Ester Hydrolases, ATPases and Carboanhydrases

1994

Many biomolecules are esterified with carbonic acid, phosphoric acid or sulphuric acid. The enzymes responsible for the hydrolytic cleavage of the esters are the carboxylester hydrolases, phosphoesterases and sulphatases. In addition to the phosphomonoesterases, we will also consider the phosphodiesterases and nucleases that are specific for cAMP and cGMP, as well as the ATPases that cleave phosphoric acid anhydride bonds. The ion-transporting ATPases will be discussed together with comparative biochemical data on ion channels. The chapter ends with a discussion of carboanhydrases.

Nicotinic Receptors in Human Brain

1994

A vast knowledge is currently available on the molecular biology and the pharmacology of nicotinic acetylcholine receptors (nAChR) in the mammalian central nervous system (CNS) (Sargent, 1993). Only few attempts have been made to approach the expression of nAChRs at the level of functional systems, considering the different cell types involved and their connectivity. This aspect is of particular importance in order to evaluate nAChR expression under pathological conditions. Histochemical techniques have proven to be useful since immunohistochemistry and in situ hybridization can be performed on human autopsy tissue and allow for a cell type-specific localization of nAChR proteins and nAChR …

Behavioral changes over several successful agonistic encounters between male mice: Effects of type of “standard opponent”

1994

This study assessed whether two types of non-aggressive “standard opponents” (“intact” and “anosmic” group-housed males) produced similar behavioral changes in isolated OF1 male mice given several experiences of victory. Experimental groups confronted either intact or anosmic opponents every two days until they had completed four encounters. The behavioral changes were recorded using a detailed ethologically inspired analysis. These changes were clearly different depending on the opponent type. When intact opponents were used, experimental subjects increased the time spent in digging, non-social exploration, explore from a distance, and attack over encounters, but showed decreased time spen…

Cellular Immunity and Retrobulbar Fibroblasts in Graves' Ophthalmopathy

1994

In Graves' ophthalmopathy (GO), retrobulbar connective tissue is infiltrated by T cells whose role in the pathogenesis of the disease was investigated in the present work. The aims included firstly to characterize subsets of blood lymphocytes and of sessile lymphocytes cloned from a retroorbital tissue specimen. Second, in counterstimulation assays, the ability of patients' T cells to influence cultivated retrobulbar fibroblasts and in turn the enhancement of lymphocyte proliferation by retrobulbar fibroblasts was investigated. Blood lymphocytes of 16 GO patients and 12 controls isolated by density gradient centrifugation and retrobulbar fibroblasts obtained from orbital decompression were …

Significant biogenesis of chlorinated aromatics by fungi in natural environments

1994

Common wood- and forest litter-degrading fungi produce chlorinated anisyl metabolites. These compounds, which are structurally related to xenobiotic chloroaromatics, occur at high concentrations of approximately 75 mg of chlorinated anisyl metabolites kg of wood-1 or litter-1 in the environment. The widespread ability among common fungi to produce large amounts of chlorinated aromatic compounds in the environment makes us conclude that these kinds of compounds can no longer be considered to originate mainly from anthropogenic sources.

Alternating, all-trans polyenynes: Model compounds for poly(diacetylene)s with defined conjugation length

1994

The syntheses of polyenynes as model compounds for poly(diacetylene)s (PDAs) are described. Variation of properties (UV–VIS, Raman, NMR and bond geometries) as a function of the chain length was investigated. After extrapolation to infinite chain length these data were compared to those for PDAs. From UV–VIS spectra a value of λ = 551 nm (2.25 eV) was calculated corresponding to the electronic transition of a single polyenyne chain. This energy is located at the low energy end of a yellow PDA solution spectrum. From Raman scattering v(CC) = 2108–2128 cm−1 and v(CC) = 1505–1532 cm−1 were calculated after extrapolation. Similarly sp-C13C NMR data yielded a shift of δ = 100 ppm. These data are…

Determination of methamphetamine in urine samples with sodium 1,2-naphthoquinone-4-sulphonate

1994

Optimal conditions have been studied for the determination of methamphetamine in urine samples by an extractive-spectrophotometric method with sodium 1,2-naphthoquinone-4-sulphonate (NQS) as reagent. These conditions are: NaHCO3 pH 10, NQS 6.3 × 10−3 mol/l and heating for 5 min at 45°C. The accuracy and precision of the method were tested. The detection limits were 0.2 mg/l in the standard and 0.9 mg/l when 5 ml of urine sample were taken. The standard deviation of blank urine was evaluated from 12 different samples. The relative errors found in the determination of methamphetamine in urine were lower than 10% if the methamphetamine-amphetamine ratio was higher than 4.

Diffusion Measurements on Crystalline Rock Matrix

1994

AbstractA new gas flow technique is introduced such that experiments on very long samples are possible. This new technique together with increased accuracy of the measurements, allows the observation of power law tails in the break-through curves. Dispersion in these experiments can be controlled in great detail, and therefore the power law tails can be used to determine very accurately the parameters relevant in matrix diffusion. Results for rock and metal samples are shown, and they are fitted with model calculations which include both dispersion and matrix diffusion. The introduced technique, which is designed for ordinary drill cores, is suitable for scanning a large number of samples i…

Non-coordinate synthesis of MHC class II proteins and invariant chains by epidermal Langerhans cells derived from short-term in vitro culture

1994

Epidermal cells (EC) prepared from Lewis rat skin contained 2-3% class II+, LCA+ Langerhans cells (LC). LC enriched from freshly isolated EC suspensions proved highly effective accessory cells when presenting the nominal antigen OVA to an RT1.Bl-restricted ovalbumin (OVA)-specific rat T cell clone. Short-term preculture of the EC resulted in diminished OVA presenting capacity of the LC. Flow cytometry (FCM) analysis of class II and gamma chain expression revealed an up-regulation of class II on the LC's cell surface, consistent with earlier findings in mouse and human. However, while the presence of gamma chains in mouse LC was reported to decline to negligible levels during culture we obse…

Search for a new light gauge boson in decays of π0 and η

1994

Abstract We have searched for new light gauge bosons produced in π 0 and η decays by studying the kinematically well-constrained reactions p p→π 0 π 0 π 0 and p p→π 0 π 0 η , where one π 0 or the η decays through the emission of a single photon recoiling against a missing state X (where X is a long-lived weakly interacting particle or X →ν ν ). No signal has been observed and branching ratio upper limits of 6 × 10 −5 at 90% C.L. have been obtained for masses of the gauge boson lying between ∼65 MeV and 125 MeV ( π 0 decay), and 6 × 10 −5 at 90% C.L. for X masses between ∼200 MeV and 525 MeV (η decay). The π 0 -decay limit represents a factor of 4 to 8 improvement when compared to the existi…

Estrellas del apocalipsis : en torno a algunos mitos del cine de terror = Stars of the Apocalypse : Observations on a few Movie Myths

1994

Income Equality and Income Taxation

1994

In every society the distribution of income plays an essential role — not only for economists but also for social scientists and politicians. Most people agree that the laissez-faire allo-cation is not equitable but that some redistribution using taxes has to take place. Most of the literature has supposed that a Bergson- Samuelson social welfare function is the appropriate tool for capturing social values in such analyses. However, there are some objections against this approach. First, interpersonal comparability of the individual utilities is needed — an extremely strong and questionable claim. Second, most writers have worked with a concave transformation of individual cardinal utilitie…

Scoresysteme in der Notfallmedizin?

1994

The primary goals of scoring in emergency medicine are grading of the severity of the patient's condition, measurement of diagnostic and therapeutic efforts, forecasting the outcome, and support in decision making on triage and therapy. Scores can also be used as tools for measuring efficacy and controlling quality. There has been less experience with use of scoring systems to estimate quality of life. The ability to make a prognosis in an individual case is the most critical point; a score may support decisions on therapy in very specific situations only. Scores for use in emergency medicine should be based on physiological parameters, universally applicable and suitable for use throughout…

Functional consequences of prejunctional receptor activation or blockade in the iris.

1994

The iris is innervated by nerves of the sympathetic, parasympathetic, and sensory nervous systems. The terminal nerve fibres are endowed with prejunctional receptors which modulate neurotransmitter release. Activation or blockade of prejunctional receptors by drugs may have an influence on iris smooth muscle tone. Several findings are in favour of the hypothesis that prejunctional receptors may be involved in regulation of iris smooth muscle tone and/or pathophysiological events. (i). Release of acetylcholine from parasympathetic nerves of guinea-pig iris sphincter evoked by electrical stimulation is subject to autoinhibition via prejunctional M2 muscarinic receptors, and the release can be…
