Deciphering the Nonsense Readthrough Mechanism of Action of Ataluren: An in Silico Compared Study

Ataluren was reported to suppress nonsense mutations by promoting the readthrough of premature stop codons, although its mechanism of action (MOA) is still debated. The likely interaction of Ataluren with CFTR-mRNA has been previously studied by molecular dynamics. In this work we extended the modeling of Ataluren's MOA by complementary computational approaches such as induced fit docking (IFD), quantum polarized ligand docking (QPLD), MM-GBSA free-energy calculations, and computational mutagenesis. Backing out of private pension provision - Lessons from Germany

Financing pensions in the EU is a challenge. Many EU countries introduced private pension schemes to compensate declining public pension levels due to reforms made necessary by demographic change. In 2001, Germany introduced the Riester pension. Ten years after introduction the prevalence rate of this voluntary private pension scheme approximates 37 %. However, numerous criticisms raise doubts that the market for Riester products is transparent. Nonsense codons suppression. An acute toxicity study of three optimized TRIDs in murine model, safety and tolerability evaluation.

Stop mutations cause 11% of the genetic diseases, due to the introduction of a premature termination codon (PTC) in the mRNA, followed by the production of a truncated protein. A promising therapeutic approach is the suppression therapy by Translational Readthrough Inducing Drugs (TRIDs), restoring the expression of the protein. Recently, three new TRIDs (NV848, NV914, NV930) have been proposed, and validated by several in vitro assays, for the rescue of the CFTR protein, involved in Cystic Fibrosis disease. Der Einfluß des Lösungsmittels auf die Teilreaktionen bei der radikalischen Polymerisation des Methylmethacrylates

Mit Hilfe der Methode der intermittierenden Belichtung (Sektormethode) in Kombination mit Stationarmessungen wird eine Absolutbestimmung der Geschwindigkeitskonstanten aller am Polymerisationsprozes beteiligten Elementarreaktionen durchgefuhrt. Hierbei wird die Viskositat des Losungsmittels insgesamt um den Faktor 170 und der Polymerisationsgrad um den Faktor 20 variiert. Es zeigt sich, das die Radikalausbeute (efficiency) bei Erhohung der Viskositat absinkt und das die Geschwindigkeitskonstante des Kettenabbruchs umgekehrt proportional der Viskositat und vollkommen unabhangig vom Polymerisationsgrad der Polymerradikale ist. Con la sentenza n. 16253/2020 la Suprema Corte ha affermato che l’assenza di nesso causale tra la cessazione di un appalto e il licenziamento del lavoratore integra un’ipotesi di manifesta insussistenza del fatto posto alla base del licenziamento - con conseguente applicabilità della tutela reintegratoria attenuta di cui all’art. 18, comma 5, della legge n. 300/1970 - non riconducibile, pertanto, agli altri casi in cui il giudice dichiara risolto il rapporto concedendo la sola tutela indennitaria forte (da dodici a ventiquattro mensilità).

Durch Kombination stationarer und instationarer Methodik (rotierender Sektor) werden die Absolutwerte der Geschwindigkeitskonstanten von Kettenwachstum und Kettenabbruch von Benzylmethacrylat unter dem Einflus verschieden viskoser Losungsmittel bestimmt. Die Radikalausbeute f hangt von der Viskositat des Losungssystems und der Art der Monomeren, dagegen nicht (oder fast nicht) von der Konzentration des Monomeren und der Geschwindigkeit der Radikalerzeugung ab.
