Different strategies to achieve Pb-tolerance by the two Trebouxia algae coexisting in the lichen Ramalina farinacea.

2012

Lichen thalli are permeable to airborne substances, including heavy metals, which are harmful to cell metabolism. Ramalina farinacea shows a moderate tolerance to Pb. This lichen comprises two Trebouxia phycobionts, provisionally referred to as TR1 and TR9, with distinct physiological responses to acute oxidative stress. Thus, there is a more severe decay in photosynthesis and photosynthetic pigments in TR1 than in TR9. Similarly, under oxidative stress, antioxidant enzymes and HSP70 protein decrease in TR1 but increase in TR9. Since Pb toxicity is associated with increased ROS formation, we hypothesized greater Pb tolerance in this phycobiont. Accordingly, the aim of the present study was …

Analysis of Diuretics in Urine by Column-Switching Chromatography and Fluorescence Detection

1997

The potential of column-switching chromatography and fluorescence detection for the analysis of diuretics in urine is evaluated. Sample cleanup and chromatographic parameters have been optimized to achieve maximum sensitivity for the detection and quantification of some relevant diuretics. On the basis of these studies, an on-line procedure for the simultaneous determination of amiloride, furosemide, bumetanide and triamterene is presented. The linearity, precision, accuracy and sensitivity of the method are discussed. The utility of the described approach has been tested by analysing urine samples obtained after administration of bumetanide.

Determination of triamterene in urine by HPLC using fluorescence detection and column-switching

1994

A liquid chromatographic method incorporating column-switching and fluorimetric detection for the determination of triamterene in untreated urine, is described. The urine samples (5 μL) were directly introduced onto an Hypersil ODS-C18, 30 μm (20 mm×2.1 mm I.D.) pre-column. Polar urinary compounds were removed by flushing the pre-column with water for 1 min, and the analyte was then switched onto an HP-LiChrospher RP C18,5 μm (125 mm×4mm ID) analytical column using an acetonitrile/phosphate buffer gradient elution. Fluorescence detection was performed at 230 nm excitation and 430 nm emission wavelengths. The recovery of drug was 102±2% in the 0.10–20.0 μg/mL concentration range, the limit o…

Efficient two-step synthesis of water soluble BODIPY–TREN chemosensors for copper(II) ions

2017

Two simple fluorescent sensors for Cu(II) ions were synthesized in two reaction steps, using recently developed C–H functionalization reactions for boron dipyrrins, starting from a standard 8-aryl-BODIPY dye and a tris(2-aminoethyl)amine (TREN) ligand. At pH 5, the resulting BODIPY–TREN conjugates are demonstrated to be promising, highly selective, water soluble, Cu(II) sensors that can permeate the cell membrane. crosscheck: This document is CrossCheck deposited related_data: Supplementary Information identifier: Bram Verbelen (ORCID) identifier: Wim Dehaen (ORCID) copyright_licence: The Royal Society of Chemistry has an exclusive publication licence for this journal copyright_licence: Thi…

Contribution to the study of the alteration of lipase activity ofCandida rugosa by ions and buffers

1994

A semipurified C. rugosa lipase (LS) has been prepared from commercial lipase (LC) using an economical procedure. The presence of sugars and glycopeptides has been detected in LS and LC. Pure lipase only has covalently bonded sugars. The hydrolysis of olive oil catalyzed by LS and commercial lipase (LC) is sensitive to the presence of cations Na(I), Mg(II), Ca(II), and Ba(II) and to the nature of buffer. Highest enzyme activity is obtained with 0.1M Tris/HCl buffers and the combination of NaCl 0.11M and CaCl2 0.11M. Fluorescence spectroscopy analysis of LC, LS, and both pure isoenzymes lipases A and B, was used to analyze the interaction of the lipase with these effectors. Inorganic cations…

Frequency-modulation spectroscopy with blue diode lasers

2000

Frequency-modulation spectroscopy provides ultrasensitive absorption measurements. The technique is especially adaptable to diode lasers, which can be modulated easily, and has been used extensively in the near-infrared and infrared spectral regions. The availability of blue diode lasers now means that the accessible wavelength region can be increased. We successfully demonstrate wavelength-modulation spectroscopy and two-tone frequency-modulation spectroscopy for the weak second resonance line of potassium at 404.8 nm and for the transition at 405.8 nm in lead, starting from the thermally populated 6p(2) P-3(2) metastable level, information on the modulation parameters is obtained with a f…

Pyrrolo[2,3-h]quinolinones: A new ring system with potent photoantiproliferative activity

2006

A new class of compounds, the pyrrolo[2,3-h]quinolin-2-ones, nitrogen isosters of the angular furocoumarin Angelicin, was synthesized with the aim of obtaining new photochemotherapeutic agents with increased antiproliferative activity and lower undesired toxic effects than the lead compound. Two synthetic pathways were approached to allow the isolation both of the dihydroderivatives 10-17 and of the aromatic ring system 23. Compounds 10-17 showed a remarkable phototoxicity and a great UVA dose dependence reaching IC(50) values at submicromolar level. Intracellular localization of these compounds has been evaluated by means of fluorescence microscopy using tetramethylrhodamine methyl ester a…

3'-Untranslated regions of oxidative phosphorylation mRNAs function in vivo, as enhancers of translation

2000

Recent findings have indicated that the 3´-untranslated region (3´-UTR) of the mRNA encoding the β-catalytic subunit of the mitochondrial H+-ATP synthase has an in vitro translation-enhancing activity (TEA) [Izquierdo and Cuezva, Mol. Cell. Biol. (1997) 17, 5255–5268; Izquierdo and Cuezva, Biochem. J. (2000) 346, 849–855]. In the present work, we have expressed chimaeric plasmids that encode mRNA variants of green fluorescent protein in normal rat kidney and liver clone 9 cells to determine whether the 3´-UTRs of nuclear-encoded mRNAs involved in the biogenesis of mitochondria have an intrinsic TEA. TEA is found in the 3´-UTR of the mRNAs encoding the α- and β-subunits of the rat H+-ATP syn…
