Multiple voltage‐gradient gel electrophoresis system

A new device, based on the principle of voltage-gradient gel electrophoresis, was developed in order to enhance differentiation of the distance across the range of molecular masses in the electrophoretic fractionation of nucleic acids in an agarose matrix. The apparatus has a series of modular parallel plates, placed slantwise to allow reiteration of the voltage gradient effect along the gel. This subjects DNA fragments of variable length to differential runnings according to their original position in the gel. Fluorescence of soil humic acids and their fractions obtained by tandem size exclusion chromatography-polyacrylamide gel electrophoresis

Humic acids (HAs) extracted from soils of different origin (chernozem, ferralsol and ranker) and their fractions (A, B and C+D) obtained by tandem size exclusion chromatography–polyacrylamide gel electrophoresis were investigated by steady-state fluorescence spectroscopy in the emission mode. Independently of HA source, high molecular size fractions A and B are shown to be weakly fluorescent. The main fluorophores, especially those emitting at long wavelength (around 500–510 nm), are contained in the polar and low molecular size fractions C+D. Molecular size and net charge of isoforms of pathogenesis-related (PR) chitinase, beta-1,3-glucanase and peroxidase were studied in uninfected barley (Hordeum vulgare L., v. Karat) leaves and in barley leaves infected with the pathogenic fungus Drechslera teres f. teres (Sacch.) Shoem. Effect of α-factor on individual wall mannoproteins fromSaccharomyces cerevisiae acells

Treatment of Saccharomyces cerevisiae a cells with α-factor partially inhibits mannosylation of the high Mr mannoproteins, although there is an increase in the total amount of these molecules present in the wall. They show a similar mobility in SDS-acrylamide gels to those from untreated mnn2 cells. No other significant effects on wall mannoproteins have been observed, except a decrease in the amount of the 29 kDa species.

The antigenic properties of somatic (SoP) and excretory-secretory products (ESP) of Echinostoma caproni were studied in experimentally infected rats. Sodium dodecyl sulphate-polyacrylamide gel electrophoresis demonstrated a total of 15 and 9 major polypeptides, in the molecular weight range 10-165 kDa, in SoP and ESP products, respectively. Further analysis revealed some common as well as several unique polypeptides for each type of extract. A total of 2 polypeptides, weighing 58 and 115 kDa, were found in both types of antigen. Indirect enzyme-linked immunosorbent assay analysis using both types of antigens showed that rats developed a weak and slow response in infections with E. caproni. …

The plasma membrane ATPase of Kloeckera apiculata: purification, characterization and effect of ethanol on activity

Partially (6-fold) purified plasma membrane ATPase from an ethanol-sensitive yeast, Kloeckera apiculata, had an optimum pH of 6.0, an optimum temperature of 35°C, a K m of 3.6 mM ATP and a V max of 11 μmol Pi/min.mg protein. SDS-PAGE of the semi-purified plasma membrane showed a major band of 106 kDa. No in vivo activation of the ATPase by glucose was observed. Although 4% (v/v) ethanol decreased the growth rate by 50% it did not affect the ATPase. Concentrations of ethanol ≥2% (v/v) did, however, inhibit the enzyme in vitro. The characteristics of the enzyme did not change during growth in the presence of ethanol.

Studying the separation behavior of various native carbonic anhydrase isozymes from mammalian erythrocytes we found that the migration of these enzymes differs from that of the marker proteins commonly used in gradient gel electrophoresis. In alkaline buffer systems the enzymes from human, bovine, rabbit, and canine erythrocytes start to migrate with a size apparently 6 to 12 times larger than their monomeric size, then gradually lose in apparent size and finally end up in a size equivalent to their monomeric mol mass.
