Abstract This chapter reviews alternative methods recommended for animal testing in various toxicological areas. An alternative model to achieve complete animal replacement for acute toxicity testing is not possible. Skin corrosion/irritation alternative methods have been validated and accepted. For eye irritation testing, no single method is able to replace the Draize rabbit eye test. Skin sensitization methods imply refinement and reduction of numbers of animals. Patch testing with components of water-based metalworking fluids

Water-based metalworking fluids (MWFs) may cause both irritant and allergic contact dermatitis. Several well-known MWF allergens are available for patch testing, but considering the wide variety of possible components used in MWF, our diagnostic arsenal covers only a small part of potential allergens. We therefore selected 13 frequently used MWF components that might be sensitizers and had not yet been tested routinely. Previous literature reports as well as own investigations concerning exogenously and endogenously induced Koebner-reactions in psoriatics are presented. The time interval between irritation and the Koebner-reaction is emphasized. It is proposed, that the intensity of the psoriatic reaction is mediated by circulating DHEA-deficiency. Hypothetical models are presented which allow to explain the different developmental rates of psoriatic lesions in relation to the type of irritation, the area affected and the subsequent proliferative responses.

In a multicentre study of the German Contact Dermatitis Research Group, sodium lauryl sulfate (SLS) 0.25% and 0.5% aq. has been added to routine allergen patch tests to assess its properties as a convenient diagnostic indicator of individual susceptibility to irritation at the time of patch testing.
