A new method to isolate lichen algae by using percoll® gradient centrifugation

AbstractA rapid method to isolate intact functional algae from the lichens Evemia prunastri and Ramalina farinacea has been developed. This method is based on the use of Percoll® gradients after mechanical disruption of lichen thalli. Results obtained show that the algal preparations were virtually free of contamination by fungal hyphae. The purified algal cells were photosynthetically active and without symptoms of photoinhibition, which indicates their functional integrity. This method may be used for the isolation of intact algae from a broad range of lichen species.

Sources of geomaterials in the Sicani Mountains during the Early Middle Ages: A case study of Contrada Castro, central western Sicily

From 2017, an unknown rural settlement in Contrada Castro at Corleone (Palermo Province, western Sicily) was investigated as part of the `Harvesting Memories Project¿. The stratigraphic sequence, supported by radiocarbon dating, has demonstrated a reoccupation of a pre-Roman site during the transition between the Byzantine and Islamic periods. In particular, the main occupation occurred in the late 8th¿9th century when pottery kilns and a probable warehouse were constructed. Infantile intranuclear rod myopathy.

This report concerns three unrelated floppy infants, two girls and one boy, each biopsied at the age of 1 month. They were hypotonic since birth and required artificial ventilation. The two girls died at the ages of 4 and 3½ months, respectively, the boy is still alive at the age of 2 years, but requires assisted ventilation. Each of the three infants showed, by muscle biopsy, abundant intranuclear rods, the boy and one girl also had sarcoplasmic rods, which were not present in the other girl's muscle. Fluorescence In Situ Hybridization (FISH) is a powerful technique for localizing specific DNA targets directly in the fixed tissue or cells. Bacterial artificial chromosome (BAC) as well as commercial probes, which could be supplied ready for use or concentrated and must be diluted following the manufacturers instructions, can be used. The technique requires 2 days, as an overnight incubation of the FISH probes is needed for optimal hybridization. The critical steps include deparaffinization of tissue sections, optimal pretreatment (target retrieval and protein digestion), and probe hybridization. Relationships between soil water repellency and microbial community composition under different plant species in a Mediterranean semiarid forest

Abstract Soil water repellency (SWR) can influence many hydrological soil properties, including water infiltration, uneven moisture distribution or water retention. In the current study we investigated how variable SWR persistence in the field is related to the soil microbial community under different plant species (P. halepensis, Q. rotundifolia, C. albidus and R. officinalis) in a Mediterranean forest. The soil microbial community was determined through phospholipid fatty acids (PLFA). Il saggio esamina le peculiarità del barocco dissonante di Gesualdo Bufalino. In Bufalino l’artificio dello stile sottende una tensione ideologica. L’intensità espressiva e l’eccesso verbale cercano di compensare un vuoto e costituiscono, nel sentire dello scrittore, un formidabile mezzo «per contrastare l’ossificazione del mondo». Bufalino rivendica quindi il valore strutturale della propria scelta barocca, dichiarando che la sua scrittura è sorretta da un «barocco borrominiano» in cui «l’ornato è funzione», in mancanza del quale «l’architettura cadrebbe».

For the alkaloid raumacline (2), which is a biotransformation product of ajmaline (1) in Rauwolfia serpentina cell cultures, an efficient ‘one-pot’ synthesis was developed using a NaBH4/riboflavin/light-mediated transformation of 1 into 2 with a total yield of 86%.

The effects of elevated gaseous SO2 concentrations in the lichens Evernia prunastri (L.) Ach. and Ramalina farinacea (L.) Ach. were investigated by means of gas exchange, modulated chlorophyll fluorescence analysis and antioxidant enzyme assays. The response to SO2 of the studied species differed markedly. Net photosynthetic rates were more adversely affected in E. prunastri than in R. farinacea. In addition, processes dependent on thylakoid membrane integrity such as PSII-mediated electron flow and nonphotochemical quenching were reduced to a greater extent by exposure to SO2 in E. prunastri.
