Ģenētisko faktoru asociācija ar Helicobacter pylori seroloģisko prevalenci Latvijas populācijā

Baktērija Helicobacter pylori (HP) izraisa hronisku gastrītu, peptisko čūlu, kuņģa adenokarcinomu, MALT limfomu, un tās radītā infekcija ir visplašāk izplatītā cilvēku infekcija. Populācijas pētījumā atklāts, ka HP prevalence Latvijā ir gandrīz 80%, kas ir augsta. Pamatojoties uz Nīderlandes GWAS pētījumu rezultātiem, tika izvēlēti 5 gēni un to polimorfismi- PCLO (rs147900026), TLR10 (rs12233670), FCGR3A (rs147174426), TLE4 (rs138776142), ADRA1D (rs6107461)- lai noskaidrotu, vai šie polimorfismi asociējas ar HP seroprevalenci un kuņģa gļotādas atrofiju Latvijas populācijā. PSMA6 (rs2277460, rs1048990), PSMC6 (rs2295826, rs2295827) and PSMA3 (rs2348071) genetic diversity was investigated in 1438 unrelated subjects from Latvia, Lithuania and Taiwan. In general, polymorphism of each individual locus showed tendencies similar to determined previously in HapMap populations. Main differences concern Taiwanese and include presence of rs2277460 rare allele A not found before in Asians and absence of rs2295827 rare alleles homozygotes TT observed in all other human populations. The aim of this work was to study β-defensin 1 (SBD1) and β-defensin 2 (SBD2) genes in Valle del Belice dairy sheep in order to identify polymorphisms that can be utilized as markers of the analyzed genes, and search for the functional effects and roles of the identified polymorphisms (variation of the amino acid sequence of the protein and stability of mRNA molecule). The study was conducted on 300 randomly selected animals belonging to four flocks. A total of seven SNPs were identified, two in SBD1 and five in SBD2. We used a high-quality 7k SNP dataset of 1,038 Eurasian grape varieties with unique profiles to assess the population genetic diversity, structure, and relatedness, and to infer the most likely migration events. Comparisons of putative scenarios of gene flow throughout Europe from Caucasus helped to fit the more reliable migration routes around the Mediterranean Basin. The Pisum genus presents a large genetic diversity, that makers difficult its taxonomic classification. The Center for Grain Legumes Genetic Resources holds 1800 pea accessions. Some of them are lacking passport data. Previous genotyping studies have identified SNP markers specific to wild Pisum pools. Very few causal genes have been identified by quantitative trait loci (QTLs) mapping because of the large size ofQTLs, and most of them were identified thanks to functional links already known with the targeted phenotype.Here we propose to combine selection signature detection, SNP annotation, and expression analyses to identifycausal genes underlying QTLs. As a model, we chose experimental chicken lines divergently selected for onlyone trait, the abdominal fat weight, in which several QTLs were previously mapped.
