In vitro production of GHB in blood and serum samples under various storage conditions

Abstract The in vitro production of GHB was observed in freshly collected, untreated whole blood samples using glass BD-Vacutainers and polypropylene S-monovettes. GHB concentrations were determined daily over a period of one week and after 3, 6 and 9 weeks again. Furthermore, the GHB concentration in 40 untreated random whole blood samples stored at 4 °C for a longer period of time (10 samples 12 month, 10 samples 24 month and 20 samples 36 month) was also determined. Clinical success of intrauterine insemination cycles is affected by the sperm preparation time.

Objective To determine the impact of the time interval from the end of sperm preparation (TSP) to intrauterine insemination (IUI) on the outcome. Design Prospective multicentre cohort study. Setting Seven French centers (assisted reproduction group in northeastern France, four academic centers, and three clinics). Patient(s) Eight hundred sixty-two IUI cycles (709 patients) managed by gonadotropins were studied. A preliminary study on the stability of benzodiazepines in blood and plasma stored at 4 degrees C.

An approach to determine the stability of benzodiazepines and some of their metabolites (n = 13) by means of a routinely applied gas chromatographic method using electron capture detection was made in this preliminary study. Validation data of the method are given. Spiked blood and plasma samples were stored at 4 degrees C and analysed at selected times up to 240 days. The concentrations of all analytes had decreased to at least 60% of the original levels at the end of the observation period. A clear pattern of breakdown could not be established. Functionally active complement proteins C6 and C7 detected in C6- and C7-deficient individuals

SUMMARYTwo sensitive sandwich ELISAs based on monoclonal antibodies directed to native C6 and C7 allowed the detection and quantitation of these complement proteins in 20 out of 37 serum samples from individuals who had previously been classified as deficient in these proteins as assessed by immunochemical and/or functional assays. Furthermore, serum from four C6-deficient and one combined C6-/C7-deficient individual showed an increase in the terminal complement complex (TCC) and a decrease in native C6 and C7 after complement activation as assayed by specific ELISAs. A simple and flexible system, employing a column switching technique, has been designed to allow the analysis of peptides and proteins smaller than 15 kDa by molecular weight in filtered urine samples by performing a direct on-column injection utilising simultaneous sample clean-up and trace enrichment. The positively charged peptides and small proteins in the sample are attracted to the inner, negatively charged pore structure of the RAM-SCX column while the larger proteins and uncharged or negatively charged compounds are excluded. The European Culture Collections Organisation presents two new model documents for Material Deposit Agreement (MDA) and Material Transfer Agreement (MTA) designed to enable microbial culture collection leaders to draft appropriate agreement documents for, respectively, deposit and supply of materials from a public collection. An interface to allow on-line qualitative and quantitative full-plate detection and analysis of compounds separated by thin-layer chromatography (TLC) is presented. A continuous wave diode laser is employed as a desorption source. Atmospheric pressure chemical ionization mass spectrometry ionizes and subsequently identifies the desorbed sample molecules. Besides direct laser desorption on untreated TLC plates, graphite particles were used as a matrix to couple in the laser power and improve the efficiency of desorption.
