In experiments on isolated, electrically stimulated papillary muscles or auricles of guinea pig hearts some characteristics of the inotropic action of strophanthidin-3-bromaocetate (SBA), a derivative of k-strophanthidin which is supposed to be an irreversible inhibitor of the (Na+-K+)-activated ATPase, were studied and compared with the behaviour of k-strophanthidin and digitoxin.

The reversibility of the inhibitory action of strophanthidin, strophanthidin-3-bromoacetate, and digitoxin on the Na+-K+-ATPase activity (microsomal fraction, guinea-pig brain) was compared with the rate of decline of their positive inotropic effects (papillary muscle, guinea-pig heart) after exposure to drug-free solution. The actions of strophanthidin and digitoxin were easily reduced on both systems. Strophanthidin-3-bromoacetate, however, was found to have an irreversible blocking effect on the transport ATPase, whereas its inotropic action could be rapidly abolished.

ATPase in the normal and dystrophic developing retina of the rat

Viene descritto un aumento significativo dell'attivita specifica dell'ATPasi (attivita/mg proteine) durante lo sviluppo post-natale della retina in ratti normali ed in ratti con retinite pigmentosa ereditaria. Il rapporto tra attivita ATPasica in presenza di Mg++-Na+-K+ ed attivita ATPasica in presenza di Mg++ non varia nella retina normale, mentre esso va incontro ad un precoce declino durante lo sviluppo della retina distrofica. Il significato di questi dati e discusso molto brevemente.

The cytoplasma fraction of the bovine choroid plexus epithelial cells was found to contain a considerable ATPase activity. The influence of Na+, K+, Li+, Rb+, Cs+, Co++, Mn++, Zn++ and Fe+++ on the activity of the Mg++-dependent enzyme has been studied. The monovalent cations do not influence the enzymic activity, whereas the effect of the bi- and trivalent cations is characterized by an inhibition of the ATPase.

Prevention of the acute neurotoxic effects of phenytoin on rat peripheral nerve by H7, an inhibitor of protein kinase C.

Abstract The neurotoxic effects of a single dose of phenytoin (150 mg/kg body weight) alone or 30 min after H7 (a protein kinase C inhibitor) injection (20 mg/kg body weight) were investigated in terms of peripheral neuromuscular function and Na + ,K + -ATPase activity of the sciatic nerve. This intraperitoneal injection of phenytoin induced complete blockade of muscle action potentials in the dorsal segmental muscles of the rat tail evoked by electric stimulation of the caudal nerve and a 40% decrease in the Na + ,K + -ATPase activity of the rat sciatic nerve when compared with control values, measured as the difference between total and ouabain-insensitive ATPase activity. Control of cellular phosphatidylinositol 4,5-bisphosphate levels by adhesion signals and Rho GTPases in NIH 3T3 fibroblasts

The involvement of small GTPases of the Rho family in the control of phosphoinositide metabolism by adhesion signals was examined in NIH 3T3 fibroblasts. Abrogation of adhesion signals by detachment of cells from their substratum resulted in a time-dependent decrease in the cellular level of PtdIns(4,5)P2 by approximately 50%. This effect could be mimicked by treatment of adherent cells with Clostridium difficile toxin B and toxin B-1470, which inhibit specific subsets of Rho and Ras GTPases. Flux of protons released by wild type and ferritin over-expressor tobacco plants : effect of phosphorus and iron nutrition

Abstract Tobacco ( Nicotiana tabacum ) plants over-expressing the iron storage protein ferritin, either in the cytoplasm or in the plastids, were grown under various P and Fe conditions. The crossed effects of both the genotypes and the environmental conditions on iron and chlorophyll concentrations in leaves, ferric reductase (EC 1.6.99.13) and plasmalemma H + -ATPase (EC 3.6.3.6) activities in roots, and fluxes of H + released by roots were determined. The increase in leaf Fe concentration observed in plants over-expressing ferritin was accompanied by an increase in root ferric reductase and H + -ATPase activities.
