Contribution of polyadenylate sequences to the translational efficiency of globin messenger RNAs

mRNAs from reticulocyte polysomes were fractionated by chromatography on poly(U)-Sepharose and thermal elution. The molar ratio of alpha- to beta-globin mRNA was found to be 2:1 and 1:1 respectively in short- and long-poly(A) size classes. Translational analyses indicated that the globin mRNAs containing long poly(A) tracts (with a mean length of about 70 nucleotides) directed protein synthesis with higher rates than did mRNA containing short poly(A) tracts (15-35 nucleotides). Analysis of Diuretics in Urine by Column-Switching Chromatography and Fluorescence Detection

The potential of column-switching chromatography and fluorescence detection for the analysis of diuretics in urine is evaluated. Sample cleanup and chromatographic parameters have been optimized to achieve maximum sensitivity for the detection and quantification of some relevant diuretics. On the basis of these studies, an on-line procedure for the simultaneous determination of amiloride, furosemide, bumetanide and triamterene is presented. The linearity, precision, accuracy and sensitivity of the method are discussed. The utility of the described approach has been tested by analysing urine samples obtained after administration of bumetanide.

Crystallization and preliminary X-ray analysis of strictosidine synthase and its complex with the substrate tryptamine

Strictosidine synthase (STR1) is a central enzyme that participates in the biosynthesis of almost all plant monoterpenoid indole alkaloids. After heterologous expression in Escherichia coli, crystals of STR1 and its substrate complex with tryptamine were obtained by the hanging-drop technique at 302–304 K with potassium sodium tartrate tetrahydrate as precipitant. All crystals belong to space group R3. The native STR1 crystals diffract to 2.95 Å and have unit-cell parameters a = b = 150.3, c = 122.4 Å. The tryptamine complex crystals diffract to 2.38 Å, with unit-cell parameters a = b = 147.3, c = 122.3 Å.

Tumor micromilieu often shows pronounced acidosis forcing cells to adapt their phenotype towards enhanced tumorigenesis induced by altered cellular signalling and transcriptional regulation. In the presents study mechanisms and potential consequences of the crosstalk between extra- and intracellular pH (pH(e), pH(i)) and mitogen-activated-protein-kinases (ERK1/2, p38) was analyzed. Data were obtained mainly in AT1 R-3327 prostate carcinoma cells, but the principle importance was confirmed in 5 other cell types. FIA-fluorimetric determination of thiamine.

A flow injection-fluorimetric determination of thiamine is reported. The procedure is based on the oxidation of the analyte with potassium hexacyanoferrate(III) immobilized on an anionic exchange resin; the fluorescence is monitored in aqueous basic solution. Concentrations of the vitamin of 0.1-4 ppm have been determined; the relative standard deviation was 1.8%. The injection rate was 28 samples/h. The influence of other substances and the determination of the drug in a pharmaceutical formulation are also reported.
Effects of a vitamin D3 analogue in a rat model of bladder outlet obstruction

OBJECTIVES To explore the effect of the vitamin D3 analogue, BXL-628, on some of the consequences of bladder outlet obstruction (BOO), e.g. hypertrophy and loss of contractile function, as vitamin D3 and BXL-628 inhibit prostate and bladder cell growth in vitro, and there are receptors for vitamin D in rat and human bladder. MATERIAL AND METHODS In female rats, BOO was produced by a standardized method; one group received daily BXL-628 (150 µg/kg per day) and the remaining rats received vehicle.
