Vicenin-2 (apigenin-6,8-di-C-β-d-glucopyranoside) is present in hydroalcoholic extracts of the Brazilian species Lychnophora ericoides Mart., Asteraceae, leaves, and the biological effects of this compound have been demonstrated including anti-inflammatory, antioxidant and anti-tumor effects in rat models. Uncontrolled growth, one of the fundamental properties of malignant tumors, requires a great supply of energy. This energy can be derived from the use of a variety of substrates. Besides glucose oxidation and glucose breakdown to lactic acid, the turnover of endogeneous substrates such as amino acids, free fatty acids and ketone bodies is well documented in vitro. However, under in vivo conditions, only glucose utilization has been investigated in detail, using tumor isotransplants in rodents. For human tumors, only scarce data is available, derived mainly from clinical observations rather than from systematic studies.

Bovine lactoferrin pepsin hydrolysate exerts inhibitory effect on angiotensin I-converting enzyme-dependent vasoconstriction

Abstract The inhibitory effect of a pepsin hydrolysate of bovine lactoferrin (LFH) on angiotensin I-converting enzyme (ACE) has been examined using in vitro and ex vivo functional assays. In vitro assays showed a LFH inhibitory effect on ACE activity with an IC 50 value of 0.95±0.06 mg mL −1 . Ex vivo functional assays using rabbit carotid artery segments showed a LFH inhibitory effect on ACE-dependent angiotensin I-induced contraction, but not on angiotensin II-induced contraction, suggesting that the effect of LFH is not due to antagonism of receptors for angiotensin II. Abstract Inverse correlated 2D 1H-13C NMR spectroscopy was used to follow the metabolism of vanillin (1) in vivo by plant cell suspension cultures of Nicotiana plumbaginifolia at the natural abundance of 13C. Under the non-invasive conditions four products could be detected: vanillylalcohol (2), vanillinglucoside (3) and the phenolic and benzylic glucosides of vanillylalcohol (4 and 5).

Changes in collagen cross-linking of articular cartilage are revealed by spectral reflectance imaging

Excessive cross-linking of collagen during aging may contribute to degeneration of articular cartilage. Traditionally, the amount of cross-links is derived by using destructive high-performance liquid chromatography (HPLC). However, a sensitive, non-destructive method could help to evaluate tissue integrity, including cross-links. We increased collagen cross-linking in bovine articular cartilage by controlled threose incubation. A series of 2-sulfonyliminodihydropyrimidine derivatives have been synthesized and evaluated in vivo for their antinociceptive and anti-inflammatory activities. The results were compared with that of acetyl salicylic acid. Compounds 6Ab-d and 6Be displayed an interesting analgesic profile in the acetic acid-induced abdominal contractions test. Based on the results of the carrageenan-hind paw edema test, compound 6Af showed potential anti-inflammatory activity.

Influence of bacteria on the metabolite pattern of phenol in zebra fish (Brachydanio rerio)

Summary An investigation was performed on the potential influence of bacteria in the medium on the metabolism of phenol in zebra fish. The results obtained in vitro and in vivo show that bacteria may influence biotransformation studies of xenobiotics by fish in different manner. First, it was shown that phenol conjugates were deconjugated. Hydrolysis of phenyl glucuronide was observed both in vitro and in vivo. Deconjugation of phenyl sulphate was found to a minor extent merely under in vitro conditions. Second, the xenobiotic itself, in this case phenol, was degradated to CO2.
