HPLC-SEC: a new approach to characterise complex wastewater effluents

ABSTRACTThis work investigates the use of HPLC-SEC to characterise dissolved organic matter (DOM) of complex wastewater effluents. A silica-based column, sodium acetate eluent and multiple detections were employed: UV-254 absorbance for humictype, and tryptophan-like (Ex/Em = 270/355) and tyrosine-like (Ex/Em = 270/310) fluorescence for protein type compounds. Effects of eluent pH, eluent ionic strength and injection volume on separation efficiency were tested. These cells release soluble mediators which induce the activation of hepatic stellate cells (HSCs), the main fibrogenic cell type within the liver. Alkaline hydrolysis/methylation-acetylation: a new technique for ultrastructural DNA cytochemistry.

SUMMARY A new technique for the visualization of DNA-containing structures in electron microscopy is described. Samples of glutaraldehyde-fixed bone marrow from rats were subjected to alkaline hydrolysis to remove RNA and the phosphate of phospho-proteins, followed by a combined blockage of protein carboxyl and amino groups through methylation-acetylation. After uranyl acetate staining of epoxy-embedded ultrathin sections, chromatin from all cell types showed a highly selective and intense electron opacity. Staining methods for DNA were also positive in semithin sections. This simple procedure could be very useful in ultrastructural cytochemistry of DNA and chromatin.

Histochemical and biochemical investigations concerning the function of larval oenocytes of Tenebrio molitor L. (Coleoptera, Insecta).

Larval oenocytes of Tenebrio molitor were investigated histochemically. In contrast to the lipid droplets of the fat body, they did not stain with Sudan black. A positive reaction for lipoproteins appeared only after destructive oxidation with sodium hypochlorite. These lipoproteins are the remnants of degenerated membranes, as revealed by ultrastructural analysis. Polyphenols could be identified in the exocuticle of exuvia, and in the newly formed procuticle. Endocuticle, epidermis and oenocytes showed no staining reaction. In oenocytes a great amount of lipase is also present which could be detected with several Tweens as substrates. Highly efficient transport of carboxyfluorescein diacetate succinimidyl ester into COS7 cells using human papillomavirus-like particles

AbstractHuman papillomavirus virus-like particles (VLPs) have recently been used to deliver genes into mammalian cells in vitro and in vivo. Here, we investigated whether VLPs may serve as an efficient carrier of low molecular weight compounds (e.g. hormones, vitamins, peptides etc.) into cells. COS7 cells were incubated with recombinant HPV-16L1/L2 VLPs labelled with the fluorescence dye carboxyfluorescein diacetate succinimidyl ester. Using flow cytometry, we demonstrate that labelled VLPs can specifically bind to the cell surface followed by their complete internalisation. Ionic liquids as solvents for the Knoevenagel condensation: understanding the role of solvent–solute interactions

The Knoevenagel condensation in ionic liquids has been demonstrated as a strongly solvent-dependent process. The objective of this study was to establish a simple and descriptive trend of solvent–solute interactions that favour the Knoevenagel condensation in ionic liquid media. The reaction between 4-(dimethylamino)benzaldehyde and ethyl cyanoacetate in several imidazolium- and pyrrolidinium-based ionic liquids and one ionic liquid mixture was studied. (η6-Benzamide)tricarbonylchromium and tricarbonyl(η6-phenylacetamide)chromium (1 and 2) have been synthesized from the corresponding amides and chromium hexacarbonyl. Decalin, a mixture of decalin and butyl acetate, and neat butyl acetate have been used as the reaction media. The first system gave best yields. Both amide complexes form adducts with non-complexed molecules of the type [(CO3)CrL] · L through hydrogen bonds, suggested on the basis of 1H and 13C NMR spectroscopy.
