In experiments on isolated, electrically stimulated papillary muscles or auricles of guinea pig hearts some characteristics of the inotropic action of strophanthidin-3-bromaocetate (SBA), a derivative of k-strophanthidin which is supposed to be an irreversible inhibitor of the (Na+-K+)-activated ATPase, were studied and compared with the behaviour of k-strophanthidin and digitoxin.

Effect of a "therapeutic" concentration of digitoxigenine on myocardial potassium and sodium content in Ca-poor media.

The effect of 3·10−7 g/ml digitoxigenine on cellular K and Na concentrations, [K]i and [Na]i, water content, extracellular space, and contractile force was investigated in isolated electrically stimulated left guinea-pig auricles at reduced extracellular Ca concentration (0.45 mM). Digitoxigenine, which increased contractile force without producing arrhythmias or contractures, decreased [K]i and enhanced [Na]i. Water content and extracellular space remained unchanged. It is concluded that digitoxigenine can decrease myocardial [K]i and increase [Na]i even in “therapeutic” concentrations, provided that the experiments are performed in Ca-poor media.

The variation of acetylcholine release from myenteric neurones with stimulation frequency and train length. Role of presynaptic muscarine receptors.

The effects of scopolamine on the release of 3H-acetylcholine (ACh) from the guinea-pig myenteric plexus were studied at different stimulation frequencies (0.03–10 Hz) and train lengths (1–180 pulses). Release of 3H-ACh was measured in the absence of cholinesterase inhibitors as the outflow of tritium from myenteric plexus-longitudinal muscle preparations preloaded with 3H-choline. 2. In control experiments the volley output of 3H-ACh declined with increasing train length and increasing stimulation frequency. Stimulation by one pulse produced the highest volley output. 3. H2-antihistaminics. 20. Structure-activity relationships in H2-receptor antagonists containing a 4-pyrimidone moiety.

In a series of 5,6-substituted 4- pyrimidones 1 a-v the H2-antihistaminic activity was determined (guinea-pig atrium) and lipophilicity data are given in form of Rmo -values. The influence of different substituents at position 5 or 6 of a pyrimidone moiety has been studied. Quantitative structure-activity analyses showed the importance of lipophilicity for drug activity. Additionally other physicochemical substituent-properties may play a major role.

Akute Wirkungen von Trijodthyronin (T3 10−6 und 10−5 g/ml) auf den K-Umsatz von Meerschweinchenvorhofen wurden mit Hilfe von 42K in vitro untersucht. Veranderungen der intracellularen Ionenkonzentrationen wurden nur nach 2 Std dauernder Einwirkung von T3 10−5 g/ml beobachtet. Zu diesem Zeitpunkt war die intracellulare K-Konzentration signifikant erhoht. T3 10−6 g/ml steigerte den K-Umsatz des Meerschweinchenvorhofs. K-Influx und -Efflux waren nach 2 Std Einwirkungsdauer in gleichem Ausmas gesteigert, die K-Permeabilitat war betrachtlich erhoht. Zu spateren Zeitpunkten nahmen K-Influx und -Efflux ab. In vitro pharmacological evaluation of the dichloromethanol extract fromSchinus molle l.

The pharmacological activity of the dichloromethanol extract of Schinus molle L. (SM-DCM) was analysed in in vitro models. Preincubation of the isolated guinea-pig ileum or rat uterus preparations with the extract (100 μg/mL) abolished the contractile effects of histamine and serotonin respectively. At the same dose, the extract partially reduced the contractile effects of acetylcholine on the isolated rat duodenum. A 10 μg/mL dose showed an inhibitory effect on histamine and serotonin, but not on acetylcholine-induced contractions (NS). No significant effect was found with a 1 μg/mL dose. © 1998 John Wiley & Sons, Ltd.

Adrenaline and noradrenaline were found to depress the contractions of the electrically stimulated isolated ileum of the guinea-pig; this action was related to the dose. In these experiments smaller amounts of adrenaline were required than those necessary for the depression of the spontaneous movements of the isolated rabbit duodenum.

A quantitative analysis of myocardial Ca-metabolism was carried out on isolated, isovolumetric (10 ml/min) perfused guinea pig hearts by combined determinations of the total Ca-content and kinetics of 45Ca-efflux (collecting period 60 min) (Fig. 1). The kinetics of 45Ca-uptake was estimated by extrapolating 45Ca-efflux curves of heart muscles isotopically loaded for different times (2, 5, 10, 30 60 min) to the end of the loading-period (Fig. 2).
