Spectrophotometric study of DNA interactions with ftorafur and its elementoorganic derivatives

ABSTRACTFtorafur is an antimetabolite antitumour drug successfully used for treatment of various tumours. It is generally accepted that ftorafur is converted to 5-fluoruracil. However, some data indicate direct interactions of the compound with DNA. To test this hypothesis we have performed spectrophotometric study of DNA interactions of ftorafur and some of its elementoorganic derivatives with DNA. UV-VIS spectra of the tested compounds were recorded in absence and presence of increasing amounts of DNA. Due to the scarce biological profile, the pyrazolo[1,2-a]benzo[1,2,3,4]tetrazine-3-one scaffold (PBT) has been recently explored as promising core for potential anticancer candidates. Several suitably decorated derivatives (PBTs) exhibited antiproliferative activity in the low-micromolar range associated with apoptosis induction and cell cycle arrest on S phase. Inhibition of Formation of Rev-RRE Complex by Pyronin Y

The interaction of pyronin Y, an RNA intercalating drug, with the binding of Rev protein from human immunodeficiency virus type 1 (HIV-1) to Rev-responsive element (RRE)-containing env RNA was studied. In gel retardation assays, recombinant Rev protein tightly bound to in vitro transcribed RRE RNA. Nitrocellulose-filter-binding studies revealed a dissociation constant of ≈(1–2) = 10−10M (Pfeifer et al., 1991). Pyronin Y efficiently suppressed formation of the Rev-RRE complex. At a concentration of 1 μg ml−1, complex formation was almost completely inhibited. Pyrazolo[3,4-d][1,2,3]triazolo[1,5-a]pyrimidine: a new ring system through Dimroth rearrangement

Abstract Derivatives of the new ring system pyrazolo[3,4- d ][1,2,3]triazolo[1,5- a ]pyrimidine were synthesized from the corresponding angular isomers, through a Dimroth rearrangement, in quantitative yields. Preliminary computational studies demonstrated that this class of compounds could be a good candidate as DNA intercalating agents.

Two metal complexes of 2,2′:6′,2′′-terpyridine (terpy), i.e. Cd(terpy)Cl2 and Ni(terpy)Cl2·3H2O, have been prepared and extensively characterized. The interaction of Cd(terpy)Cl2 with synthetic DNA models, poly(dA-dT)·poly(dA-dT) (polyAT) and poly(dG-dC)·poly(dG-dC) (polyGC), has been studied by CD, fluorescence and UV-vis electronic absorption spectroscopy at several metal/polynucleotide–phosphate ratios and for different NaCl concentrations. The present study was aimed at the preparation and characterization of tailor made hybrid materials, whose peculiar hosting capability could be exploited in biotechnological applications. With this purpose, the modification of K10 montmorillonite by intercalation of Tween 20 surfactant, was accomplished. The influence of two internal parameters, namely pH and surfactant/clay ratio, on the surfactant uptake ability by clay was investigated. The adsorption mechanism was elucidated on the basis of complementary kinetic and equilibrium studies and, then, corroborated by the useful information provided by the FT-IR, TGA and XRD characterization. This work studies the influence of using hydrodynamic conditions (Reynolds number, Re = 0 to Re = 600) during Ti anodization and Li+ intercalation on anatase TiO2 nanotubes. The synthesized photocatalysts were characterized by using Field Emission Scanning Electron Microscope (FE-SEM), Raman Confocal Laser Microscopy, Electrochemical Impedance Spectroscopy (EIS), Mott-Schottky analysis (M-S), photoelectrochemical hydrogen production and resistance to photocorrosion tests. Abstract Three new crystalline architectures based on acridinium trifluoromethanesulfonate salts [C13H10N]+[CF3SO3]– have been isolated as single crystals from the reaction of the dimeric hydroxo di-n-butylstannane trifluoromethanesulfonato complex [n-Bu2Sn(OH)(H2O)(CF3SO3)]2 (1) with acridine (C13H9N, Acr), in dichloromethane at room temperature. Monolayer and multilayer thin films consisting of anionic α-zirconium phosphate (α-ZrP) sheets and polycations (poly(allylamine hydrochloride) (PAH), cytochrome c) were characterized by transmission electron microscopy (TEM), ellipsometry, UV−visible absorbance spectroscopy, reflectance FT-IR, XPS, and X-ray diffraction. Titration and powder X-ray diffraction experiments confirm that exfoliation of α-ZrP begins to occur when enough tetra(n-butylammonium) hydroxide (TBA+OH-) has been added to exceed single-layer packing of TBA+ ions (x ≈ 0.50) in the intercalation compound Zr(HPO4)2-x(TBA+PO4-)x·nH2O.
