Computational Study of Nanosized Drug Delivery from Cyclodextrins, Crown Ethers and Hyaluronan in Pharmaceutical Formulations

Abstract The problem in this work is the computational characterization of cyclodextrins, crown ethers and hyaluronan (HA) as hosts of inclusion complexes for nanosized drug delivery vehicles in pharmaceutical formulations. The difficulty is addressed through a computational study of some thermodynamic, geometric and topological properties of the hosts. The calculated properties of oligosaccharides of D-glucopyranoses allow these to act as co-solvents of polyanions in water. In crown ethers, the central channel is computed. Chemical gels of scleroglucan obtained by cross-linking with 1,w-dicarboxylic acids: Synthesis and characterization

Chemical gels obtained by cross-linking reaction between scleroglucan and I, omega-dicarboxylic acids having a number of carbon atoms from 4 to 8 in the chain, have been prepared and characterised. The networks show properties correlated with the number of carbon atoms of the acid and with the molar ratio carboxylic acid/glucose unit of the polymer (r) employed in the cross-linking reaction. Chemical conjugation of dexamethasone to a polyaspartamide and in vitro evaluation studies

Two macromolecular conjugates of dexamethasone containing different drug amounts were synthesized using PHEA as the polymeric carrier and a succinic group as spacer. The content of linked drug was equal to 25.3% w/w (conjugate A) and 12.7% w/w (conjugate B). Both polymeric conjugates, unlike the free drug, were water-soluble and the amount of unlinked drug was evaluated to be approximately about 0.01% w/w. Both conjugates were relatively stable in vitro at pH 7.4 whereas in the presence of esterase only the conjugate B was able to release drug under the used experimental conditions. Characterization of interaction between tricyclic structures containing pharmaceuticals, their models and humic substances.

Their persistence and wide consumption identify pharmaceuticals as “emerging pollutants”. The complexation of pharmaceuticals containing adamantine ring structures and their model substances with humic acids (HA) of different origins was compared using fluorescence spectroscopy as a function of pH, humic acid concentration, ionic strength, and molecular mass of HA. Binding constants between the studied pharmaceuticals and humic acids were calculated. A combination of dynamic and static quenching processes as indicated by nonlinear Stern-Volmer plots and high Kd values were positively correlated with the concentration of carboxyl groups in the studied humic acids. Tailor‐Made Amino Acids and Fluorinated Motifs as Prominent Traits in Modern Pharmaceuticals

Structural analysis of modern pharmaceutical practices allows for the identification of two rapidly growing trends: the introduction of tailor-made amino acids and the exploitation of fluorinated motifs. Curiously, the former represents one of the most ubiquitous classes of naturally occurring compounds, whereas the latter is the most xenobiotic and comprised virtually entirely of man-made derivatives. Herein, 39 selected compounds, featuring both of these traits in the same molecule, are profiled. The total synthesis, source of the corresponding amino acids and fluorinated residues, and medicinal chemistry aspects and biological properties of the molecules are discussed.
The effect of pH on polymorph formation of the pharmaceutically active compound tianeptine.

The anti-depressant pharmaceutical tianeptine has been investigated to determine the dynamics of polymorph formation under various pH conditions. By varying the pH two crystalline polymorphs were isolated. The molecular and crystal structures have been determined to identify the two polymorphs. One polymorph is an amino carboxylic acid and the other polymorph is a zwitterion. In the solid state the tianeptine moieties are bonded through hydrogen bonds. The zwitterion was found to be less stable and transformed to the acid form. During this investigation an amorphous form was identified.

6-(4-Chlorophenyl)-7-phenyl-2,3-dihydro-1H-pyrrolizines with functional groups at position 5 of the heterocyclic moiety were synthesized and tested. To determine their antiinflammatory activity bovine blood was used as enzyme source for the cyclooxygenase and 5-lipoxygenase, respectively. The iminoxy acetic acid derivative and the iminotetrazole selectively inhibit the 5-lipoxygenase, all the other compounds show medium or low affinity to the active sites of cyclooxygenase and 5-lipoxygenase. In general all compounds inhibit 5-lipoxygenase more effectively than cyclooxygenase. Investigations Concerning the Correlation of COX-1 Inhibitory and Hydroxyl Radical Scavenging Activity

The aim was to study the COX-1 inhibiting efficacy in context with hydroxyl radical scavenging properties of compounds bearing a carboxylic acid and ester function, respectively. In general, the acids are more potent radical scavengers than the corresponding esters but there is no clear correlation with their COX-1 inhibiting potencies. A feasible scavenging mechanism of carboxylic acids is discussed.

The indoles 2a-2c react with diethoxycarbenium tetrafluoroborate (1) to furnish the indolecarbaldehydes 3a-3d. In the thermodynamically controlled reaction of 3-methylindole (2a) with 1 the tris(indolyl)methane 4 and diskatole (5), are formed in addition. The limitations of these reactions are discussed and evidence is presented for a C-3-ipso-attack and a Wagner-Meerwein rearrangement, respectively, leading to the formation of 3b or 3d. Fluidity of liposome membranes doped with organic tin compounds: ESR study.

The kinetics of change in the fluidity of liposome membranes, obtained in the process of sonication of Egg Yolk Lecithin (EYL), with the admixture of organic tin compounds, was investigated. Five compounds were selected for the research: three differed in the length of hydrocarbon chains, (CH(3))(4)Sn, (C(2)H(5))(4)Sn, and (C(3)H(7))(3)SnCl, whereas two differed in the number of aromatic rings, (C(6)H(5))(2)SnCl(2) and (C(6)H(5))(3)SnCl.
