The plasma membrane ATPase of Kloeckera apiculata: purification, characterization and effect of ethanol on activity

Partially (6-fold) purified plasma membrane ATPase from an ethanol-sensitive yeast, Kloeckera apiculata, had an optimum pH of 6.0, an optimum temperature of 35°C, a K m of 3.6 mM ATP and a V max of 11 μmol Pi/min.mg protein. SDS-PAGE of the semi-purified plasma membrane showed a major band of 106 kDa. No in vivo activation of the ATPase by glucose was observed. Although 4% (v/v) ethanol decreased the growth rate by 50% it did not affect the ATPase. Concentrations of ethanol ≥2% (v/v) did, however, inhibit the enzyme in vitro. The characteristics of the enzyme did not change during growth in the presence of ethanol.

Low prevalence of anti-HCV antibody among Italian children

The seroprevalence of anti-HCV antibody was studied among 2,749 children and teenagers (1,438 males and 1,311 females) living in Italy. Anti-HCV antibody testing was positive by both EIA and RIBA in ten (0.36%) subjects. The positivity rate increased with age, ranging from 0 among children less than 6 years of age to 0.8% among those aged 17-19 years chi 2 linear regression = 0.038). Anti-HCV prevalence ranged from 0.2% in northeastern regions and in Apulia to 0.6% in Sicily and Sardinia (p0.005), and no difference was seen between males (0.35%, C.I. 95%: 0.04-0.66) and females (0.38%, C.I. 95%: 0.04-0.66) (Fisher's exact test = 0.565). Nutrient intake of endurance runners with ovo-lacto-vegetarian diet and regular western diet

During an endurance run (1,000 km in 20 days) it was investigated whether an ovo-lacto-vegetarian diet (OLVD) could cover the nutritional requirements of endurance athletes. A regular western diet (RWD) was used as reference. Both diets were offered with an energy content of 4,500 kcal per day and an energy percentage of carbohydrate:fat:protein of 60:30:10. The runners were divided into two dietary groups according to their usual dietary habits. The results of the 55 participants who completed the race show that runners from both groups had the same intake of energy, carbohydrate, fat and protein. When applied at low concentrations (10 micrograms/ml), staphylococcal alpha-toxin generates a small channel in keratinocyte and lymphocyte membranes that permits selective transmembrane flux of monovalent ions. Here we show that a moderate concentration (1-50 micrograms/ml) of alpha-toxin similarly produces a small pore in membranes of human fibroblasts. This process leads to rapid leakage of K+ and to a drop in cellular ATP to 10-20% of normal levels in 2 h. In the presence of medium supplemented with serum and at pH 7.4, the cells are able to recover from toxin attack, so that normal levels of K+ and ATP are reached after 6-8 h at 37 degrees C. Identification of iduronate-2-sulphatase (IDS) gene mutations in patients with mucopolysaccharidosis type II (MPS II, Hunter syndrome) allows fast and reliable carrier detection and prenatal diagnosis. We describe here three cases of prenatal diagnosis by direct detection of the gene mutation. In addition to two affected male fetuses from two different families, a 47,XXY fetus carrying both the normal and the mutant allele was diagnosed in a third family. The latter pregnancy was carried to term and the child is obviously not affected by MPS II.

Abstract The electromagnetic polarisability of charged pions, α E , has recently attracted both theoretical and experimental attention. Unfortunately the experimental results disagree with each other. We have investigated this polarisation via a QCD sum rule approach and find α E = 5.6 ± 0.5 × 10 −4 fm 3 , which is in agreement with one experiment and disagrees with the result of chiral perturbation theory.

This study investigated the development of planning and its relationship with age and other cognitive processes taken from the PASS model. Ninety-nine students in grades two, four, and eleven were administered planning, simultaneous processing, successive processing, and attention tasks. MANOVA with grade level and gender as independent variables revealed that grade level had a significant effect on planning, whereas gender did not. All but one planning variable showed a significant main effect of grade in subsequent ANOVAs. : The purpose of this paper is to shed light on the debate on market- versus bargaining-determined total earnings by examining whether models of wage drift based on wage-bargaining considerations empirically outperform models based on simple ad hoc formulations relating wage drift to excess demand for labour. The task is carried out by investigating the empirical performance of two bargaining models and two Phillips curve models in the context of data on the Finnish metal industry. It has been shown that translational diffusion coefficients, Dt, in the supercooled van der Waals liquids, orthoterphenyl, phenolphthaleindimethylether, and salol, have a weaker temperature dependence than the shear viscosity, η, at T ≲ 1.2Tg and can be described by Dt ∼ η−χ with χ < 1 whereas Dr ∼ η−1 applies for the mean rotational diffusion coefficients, Dr, down to the glass transition temperature, Tg. This apparent decoupling of translational and rotational motion has been discussed in relation with possible anomalous short time diffusion, spatial heterogeneity, and cooperative molecular motions close to Tg.

Abstract To ascertain the long-term effects of a triple procedure in the case of coexisting cataract and glaucoma, we did a retrospective study of 93 eyes that had cataract extraction, IOL implantation, and trabeculectomy within a ten-year period. Mean follow-up was 32.4 ± 21.9 months. Mean postoperative intraocular pressure (IOP) was significantly lower than preoperatively, even though fewer medications were being used. One year after surgery, 60% of eyes had a best corrected visual acuity of 20/40 or better with an average improvement of 3.8 Snellen lines. The order in thin films of a combined liquid crystalline polymer is studied by X-ray reflection. Films of thicknesses of less than 200 nm on float glass are investigated as a function of temperature. The polymer with mesogenic groups in the main and side-chains exhibits smectic and cholesteric mesophases. Measurements in the smectic phases show a Bragg peak and smectic layers are oriented parallel to the substrate. Charcoal analysis reveals various palaeo-ecological phases from the Neolithic to the Bronze Age. Agriculture starts about 7000 B.P. in favourable ecological conditions. Most of the charcoal spectra from sites on the coast represent thermomediterranean holm-oak forest; those from the inland mountains represent mesome-diterranean holm-oak forest. The Neolithic I Impressed Ware people were the first to clear the forest to plant their crops. This clearance of primary woodland resulted in the development of secondary vegetation of pine woods or scrub. Abstract Heat capacity and density measurements of pentanol (PeOH)-sodium dodecyl sulfate (NaDS)-dodecyldimethylamine oxide-water mixtures were carried out at 0.03 m PeOH as a function of the total surfactants concentration (mc) at different ratios (XNaDS). From experimental data, the apparent molar volumes (VΦ,R) and heat capacities (CΦ,R) of PeOH in the surfactants mixture solutions were calculated. As a general feature, at a given mixture composition, VΦ,R increases monotonically with mt as observed in pure surfactants. Domain shapes and monolayer structures of triple-chain phospholipids on water

Two triple-chain phospholipid isomers were investigated at the air-water interface by means of fluorescence microscopy and grazing incidence X-ray diffraction (GID). The two lipids differ only in the position of the branched chain at the glycerol backbone. Fluorescence microscopy shows different domain sharp-edged domains. In the case of dendritic domains the chains are more tilted, the deviation from hexagonal symmetry is more pronounced and hence the lattice anisotropy is larger.
