Characterization of chromatographic columns using the traditional van Deemter method is limited by the necessity of calculating extra-column variance, issue particularly relevant when modeling asymmetrical peaks eluted from monolithic columns. Filter paper disk techniques for assay of nucleotidase

A DE filter disk technique for assaying the activity of nucleotidase is described. This method is based on the observation that nucleotides bind to the filters at 5 mM Tris-HCl (pH 7.8) while nucleosides do not. As parameter for the nucleotidase activity the decrease of bound nucleotides is determined. In parallel experiments the amount of the product (nucleoside) formed can be measured by DEAE Sephadex column chromatography. The filter disk technique can be applied for the determination of vmax and Km of a nucleotidase by using different ribonucleosidase monophosphate substrates.

Abstract A multiresidue method for the analysis of 30 pesticides with different physicochemical properties in fruits and vegetables was developed. The method involves a rapid and small-scale extraction procedure based on matrix solid-phase dispersion using octadecylsilyl (C18) as the dispersant and dichloromethane as the eluent. The target pesticides were determined using column high-performance liquid chromatography/triple quadrupole-tandem mass spectrometry (LC/QqQ-MS/MS). The method was validated for sensitivity, linearity, reproducibility, and accuracy. Left, Right, or Both? On the Configuration of the Phenanthroindolizidine Alkaloid Tylophorine from Tylophora indica

The alkaloid (-)-tylophorine was isolated from a sample of Tylophora indica, and the crude extract was analyzed by HPLC/MS(n) and chiral HPLC/MS. While the literature states that the naturally occurring form of this alkaloid is the R-enantiomer and that its S-antipode is usually not found in nature, we confirmed the hypothesis of Govindachari and Nagarajan that natural levorotatory tylophorine is indeed a nearly racemic mixture with a slight excess of the R-enantiomer.

Preparation and Structure of Bicycloalkane-Condensed Aryldiaziridines Accompanied by Pyrimidines

Di-exo- and di-endo-2-aminonorbomane/enemethanamines 1-3, di-exo-oxanorbomene derivative 4 and cis-cyclohexane and trans-4-cyclohexene analogues 5, 6 were reacted with p-chlorobenzaldehyde in the presence of N-bromosuccinimide in dichloromethane. Via the reactions of 1-6, condensed diaziridines 7-12 accompanied by pyrimidine derivatives 13-16 were prepared after isolation with column chromatography. The mechanisms proposed for alternative transformations were supported by DFT calculations. The structures of the new compounds were proved by IR and NMR spectroscopy and, for 7, 9 and 12, also by means of X-ray crystallography.

High-performance micellar liquid chromatography determination of sulphonamides in pharmaceuticals after azodye precolumn derivatization

Abstract A chromatographic procedure with precolumn derivatization to form the N-(1-naphthyl)ethylenediamine dihydrochloride azodyes is proposed for the analysis of several sulphonamides (sodium sulphacetamide, sulphadiazine, sulphaguanidine, sulphamerazine, sulphamethizole, sulphamethoxazole, sulphanilamide and sulphathiazole) in pharmaceutical preparations (tablets, pills, capsules, suspensions and drops). The separation is performed with a 0.05 M sodium dodecyl sulphate/2.4% pentanol eluent at pH 7. Isolation and Characterization of an Fe(III)-Chelating Compound Produced by Pseudomonas syringae

The phytopathogenic bacterium Pseudomonas syringae produces a fluorescent pigment when it is grown in iron-deficient media. This pigment forms a very stable Fe(III) complex that was purified in this form by using a novel procedure based on ultrafiltration and column chromatography. The Fe(III) complex has a molecular weight of 1,100 and contains 1 mol of Fe(III). The pigment is composed of an amino acid moiety with three threonines, three serines, one lysine, δ- N -hydroxyornithine, and a quinoline-type fluorescent chromophore. Phosphatidylcholine-Gramicidin a Interaction Study in Non-Aqueous Solvent with a new HPLC-SEC Column

Abstract A new HPLC-SEC column, Ultrastyragel 500 A, has been assayed in order to check the separation between gramicidin A and phosphatidylcholine in tetrahydrofuran. The good resolution of both compounds has allowed the quantitation of lipid-polypeptide interaction in terms of the binding ratio parameter, BR. A BR value of 3.6 moles of phosphatidylcholine per mole of gramicidin A has been obtained for a 0.1 % (w/v) phosphatidylcholine solution as eluent. The suitability of this support for studies on lipid-polypeptide interactions in non-aqueous solvents is demonstrated.

Summary The synthesis of a novel technetium-99m labelled derivative of l-tyrosine as a potential tumour imaging agent for nuclear medicine diagnosis is reported. The synthesis involved the labelling precursor fac-[ 99m Tc(OH2)(CO)3] + which was synthesized using the commercially available Isolink 1 -labelling kit and the tyrosine derivative O-(N,Nbis(carboxymethyl)aminoethyl)-l-tyrosine trifluoroacetate. The labelled compound O( 99m Tc(I)-tricarbonyl-N,N-bis(carboxymethyl)aminoethyl)-l-tyrosine was obtained in a radiochemical yield of 70–80% within 60 min with a radiochemical purity greater than 98% without any HPLC purification step.
