Simultaneous determination of third-generation synthetic cannabinoids in oral fluids using cyclodextrin-silica porous sorbents

2022

Abstract A microporous material made of cyclodextrin units covalently immobilized into a silica network has found an innovative bioanalytical application in the sample clean-up and simultaneous isolation of three synthetic cannabinoids (ADB-CHMICA, MMB-CHMICA, and MDMB-CHMCZCA) from human oral fluid through solid-phase extraction. The subsequent quantitation is carried out using liquid chromatography coupled to fluorescence detection. The optimized experimental procedure gives recoveries ranging from 76% to 96% in oral fluid samples spiked with the studied cannabinoids at the µg L-1 level. High selectivity is obtained through the extraction procedure due to the presence of cyclodextrin in t…

Probes for studying cholesterol binding and cell biology.

2011

Cholesterol is a multifunctional lipid in eukaryotic cells. It regulates the physical state of the phospholipid bilayer, is crucially involved in the formation of membrane microdomains, affects the activity of many membrane proteins, and is the precursor for steroid hormones and bile acids. Thus, cholesterol plays a profound role in the physiology and pathophysiology of eukaryotic cells. The cholesterol molecule has achieved evolutionary perfection to fulfill its different functions in membrane organization. Here, we review basic approaches to explore the interaction of cholesterol with proteins, with a particular focus on the high diversity of fluorescent and photoreactive cholesterol prob…

Host—Guest Interactions Involving Cyclodextrins: Useful Complementary Insights Achieved by Polarimetry.

2007

Abstract By means of simple polarimetry, we studied the binding abilities of native α-, β-, and γ-cyclodextrins toward a group of suitably chosen model guests. We were able to get reliable estimations of the binding constants K, spread over a wide range (from 3.7 to 12,300 M−1), allowing us to carry on interesting comparisons. A comprehensive discussion of polarimetric data, and in particular a detailed analysis of the variations ΔΘ of molar optical activities consequent to inclusion, offered us the opportunity to get useful insights into the structure and dynamic behavior of host–guest complexes.

GH57 amylopullulanase from Desulfurococcus amylolyticus JCM 9188 can make highly branched cyclodextrin via its transglycosylation activity.

2018

Abstract Desulfurococcus amylolyticus is an anaerobic and hyperthermophilic crenarchaeon that can use various carbohydrates as energy sources. We found a gene encoding a glycoside hydrolase family 57 amylolytic enzymes (DApu) in a putative carbohydrate utilization gene cluster in the genome of D. amylolyticus . This gene has an open reading frame of 1,878 bp and consists of 626 amino acids with a molecular mass of 71 kDa. Recombinant DApu (rDApu) completely hydrolyzed pullulan to maltotriose by attacking α-1,6-glycosidic linkages, and was able to produce glucose and maltose from soluble starch and amylopectin. Although rDApu showed no activity toward α-cyclodextrin (CD) and β-CD, maltooctao…

Fast enantiomeric separation of uniconazole and diniconazole by electrokinetic chromatography using an anionic cyclodextrin: application to the deter…

2000

The enantiomeric resolution of the fungicides uniconazole and diniconazole was performed using electrokinetic chromatography with cyclodextrins as pseudostationary phase (CD-EKC). A systematic evaluation of several chiral selectors was made. The anionic derivative carboxymethylated-gamma-cyclodextrin (CM-gamma-CD) was found to be the most appropriate for the enantioseparation of fungicides among all cyclodextrins tested. The influence of some experimental conditions such as nature and buffer pH, chiral selector concentration, and temperature on the enantiomeric separation of the compounds studied was also investigated. The use of a 50 mM phosphate buffer (pH 6.5) containing 5 mM CM-gamma-CD…

Cellobiose fermentation by Saccharomyces cerevisiae: Comparative analysis of intra versus extracellular sugar hydrolysis

2018

Abstract A prevalent procedure for the production of second generation bioethanol makes use of engineered yeast strains capable to hydrolyze cellobiose either in the cytosol or extracellularly. These two approaches have been compared in this study. For intracellular cellobiose hydrolysis, we initially tested three recombinant Saccharomyces cerevisiae strains that produced different cytosolic β-glucosidases and the cellodextrin transporter Po_CdtC from Penicillium oxalicum . The strain coexpressing Po_CdtC and the β-glucosidase from Neurospora crassa (NcBgl) showed the highest cellobiase activity but its growth in cellobiose was limited by sugar intake. A search of alternative cellobiose per…

Cyclodextrin‐Calixarene Nanosponges as Potential Platforms for pH‐Dependent Delivery of Tetracycline

2019

Four mixed cyclodextrin-calixarene nanosponges were tested as possible Drug Delivery Systems, using Tetracycline antibiotic as a suitable model drug. The selected nanosponges featured a different composition ratio between the two host co-monomer components, and the possible presence of ionisable amine or carboxyl groups deriving from chemical post-modification. The pH-dependent absorption and release abilities of the materials were verified; in particular release kinetics showed the occurrence of a simple first-order profile. The antibacterial activity of nanosponge-tetracycline composites suitably prepared under sterile conditions was assayed towards both Gram-positive and Gram-negative ty…

Adsorption of a dye on clay and sand. Use of cyclodextrins as solubility-enhancement agents.

2007

Abstract Laboratory-scale studies were aimed at elucidating the physico-chemical aspects on the removal process of crystal violet (CV) from waters and solid substrates. The laponite clay (RD) and sand were chosen for the double aim at investigating them as CV adsorbents for water treatment and as substrates which mime the soil components. Sand is very effective in removing CV from waters. The cyclodextrins (CDs) were exploited as solubility-enhancement agents to remove CV from the solid substrates. They are powerful solvent media because they extract the CV from sand forming water-soluble CV/CD inclusion complexes and do not show affinity for sand. Optimum performance was shown by the modif…

Application of time-of-flight aerosol mass spectrometry for the online measurement of gaseous molecular iodine.

2012

Here we present a new application of a time-of-flight aerosol mass spectrometer (TOF-AMS) for the measurement of atmospheric trace gases in real-time. Usually, TOF-AMS instruments are not sensitive to gas-phase species due to the aerodynamic particle focusing inlet system which reduces the gas phase species by a factor of about 10(7) relative to the particle phase. This efficient removal of the gas phase and the resulting high relative enrichment of particles is one reason for the very high sensitivity of TOF-AMS instruments for particle phase compounds (detection limits in the sub-μg/m(3)-range for online measurements with 1 min integration time), which allows application of the instrument…

Development of a coupled diffusion denuder system combined with gas chromatography/mass spectrometry for the separation and quantification of molecul…

2009

This study concerns the development of a coupled diffusion denuder system capable of separating and quantifying gaseous molecular iodine (I(2)) and two other highly reactive iodine species, ICl and HOI, which are collectively named activated iodine compounds (AIC). Both I(2) and AIC are key species in the atmospheric chemistry of iodine. 1,3,5-Trimethoxybenzene (1,3,5-TMB)- and alpha-cyclodextrin/(129)I(-) (alpha-CD/(129)I(-))-coated denuders proved to be suitable for the collection of gaseous AIC and I(2), respectively. The experimental collection efficiencies for AIC (tested as ICl) and I(2) agreed well with the theoretical values for gas flow rates in the range between 300 and 1800 mL mi…
