[3H]-DA release evoked by low pH medium and internal H+ accumulation in rat hypothalamic synaptosomes: involvement of calcium ions
2003
The pH fluctuations have been often interpreted as an insufficient regulation or as a consequence of the onset of pathological events, such as ischemia, in which a significant decrease in pH levels occurs. Neurotransmitter release appears to be affected by pH drop significantly. In this study, we investigated the effect of an extracellular and an intracellular acidification on tritiated dopamine release ([3H]-DA release), from superfused rat hypothalamic synaptosomes. When compared to basal release, extracellular acidification, due to a reduction in the external pH of the nominally carbonic-free superfusion media, provoked a significant increase in [3H]-DA release that showed a sensitivenes…
Electrical Coupling in Ensembles of Nonexcitable Cells: Modeling the Spatial Map of Single Cell Potentials
2015
We analyze the coupling of model nonexcitable (non-neural) cells assuming that the cell membrane potential is the basic individual property. We obtain this potential on the basis of the inward and outward rectifying voltage-gated channels characteristic of cell membranes. We concentrate on the electrical coupling of a cell ensemble rather than on the biochemical and mechanical characteristics of the individual cells, obtain the map of single cell potentials using simple assumptions, and suggest procedures to collectively modify this spatial map. The response of the cell ensemble to an external perturbation and the consequences of cell isolation, heterogeneity, and ensemble size are also ana…
Mapping of a binding site for ATP within the extracellular region of the Torpedo nicotinic acetylcholine receptor beta-subunit.
1997
Using 2,8,5'-[H-3]ATP as a direct photoaffinity label for membrane-bound nicotinic acetylcholine receptor (nAChR) from Torpedo marmorata, we have identified a binding site for ATP in the extracellular region of the beta-subunit of the receptor. Photolabeling was completely inhibited in the presence of saturating concentrations of nonradioactive ATP, whereas neither the purinoreceptor antagonists suramin, theophyllin, and caffeine nor the nAChR antagonists alpha-bungarotoxin and d-tubocurarine affected the labeling reaction. Competitive and noncompetitive nicotinic agonists and Ca2+ increased the yield of the photoreaction by up to 50%, suggesting that the respective binding sites are allost…
Vertebrate Hedgehog is secreted on two types of extracellular vesicles with different signaling properties
2014
Hedgehog (Hh) is a secreted morphogen that elicits differentiation and patterning in developing tissues. Multiple proposed mechanisms to regulate Hh dispersion includes lipoprotein particles and exosomes. Here we report that vertebrate Sonic Hedgehog (Shh) is secreted on two types of extracellular-vesicles/exosomes, from human cell lines and primary chick notochord cells. Although largely overlapping in size as estimated from electron micrographs, the two exosomal fractions exhibited distinct protein and RNA composition. We have probed the functional properties of these vesicles using cell-based assays of Hh-elicited gene expression. Our results suggest that while both Shh-containing exo-ve…
Different intracellular cation-content present in right and left ventricle dependent on varying extracellular Ca2+-concentrations.
1979
Abstract The different intracellular cation-contents present in the right and left ventricle depend on varying Ca2+-concentrations. The effect of extracellular Ca2+-concentrations varied within the physiological range has been studied on an isolated guinea pig heart preparation, showing excellent stable experimental conditions. By increasing the extracellular Ca2+-concentration from 0.45 m m to 3.6 m m the tissue contents of calcium and potassium were increased dependent on [Ca]e whereas that of Na was reduced. This was due to a change in the composition of the intracellular cations as the extracellular space (inulin) and water-content of the heart muscle tissue were not influenced by incre…
Extracellular space and electrolyte distribution in cortex and white matter of dog brain in cold induced oedema
1973
24 hours after a circumscribed cold injury of the cortex dog brains were perfused from the lateral ventricle and the frontal subarachnoidal space to the cisterna magna with an artificial CSF containing trace amounts of35S-labelled thiosulphate. Simultaneously the extracellular tracer was administered intravenously. Extracellular fluid volume was estimated and found to be increased from 10 to 15% in the oedematous cortex and from 10 to 27% in the oedematous white matter. The actual size of ECS in oedematous white matter, however, must be larger as indicated by the relative alterations of thiosulphate distribution, tissue water, sodium and chloride. Apparently a small part of the fluid accumu…

Blood volume and extracellular space (ECS) of the whole body and some organs of the rat.
1978
Methods are described for estimation of blood volume and extracellular space (ECS) in the whole body and in some organs with 51Cr, 14C-thiocyanate and 3H-inulin. A mean blood volume of 47 ml/kg, a thiocyanate space of 350 ml/kg and a inulin space of 288 ml/kg were determined in the rat. The corresponding values of organs are shown in figures 1--3.
Effect of a "therapeutic" concentration of digitoxigenine on myocardial potassium and sodium content in Ca-poor media.
1972
The effect of 3·10−7 g/ml digitoxigenine on cellular K and Na concentrations, [K]i and [Na]i, water content, extracellular space, and contractile force was investigated in isolated electrically stimulated left guinea-pig auricles at reduced extracellular Ca concentration (0.45 mM). Digitoxigenine, which increased contractile force without producing arrhythmias or contractures, decreased [K]i and enhanced [Na]i. Water content and extracellular space remained unchanged. It is concluded that digitoxigenine can decrease myocardial [K]i and increase [Na]i even in “therapeutic” concentrations, provided that the experiments are performed in Ca-poor media.
Biological properties of extracellular vesicles and their physiological functions
2015
The authors wish to thank Dr R Simpson and Dr D Taylor for critical reading of the manuscript and acknowledge the Horizon 2020 European Cooperation in Science and Technology programme and its support of our European Network on Microvesicles and Exosomes in Health & Disease (ME-HaD; BM1202 www.cost.eu/COST_Actions/bmbs/Actions/BM1202). In the past decade, extracellular vesicles (EVs) have been recognized as potent vehicles of intercellular communication, both in prokaryotes and eukaryotes. This is due to their capacity to transfer proteins, lipids and nucleic acids, thereby influencing various physiological and pathological functions of both recipient and parent cells. While intensive invest…
