Development and characterization of microsatellite markers in the Colorado potato beetle, Leptinotarsa decemlineata

The characterization of 11 Leptinotarsa decemlineata microsatellite loci isolated using the fast isolation by AFLP of sequences containing repeats (FIASCO) protocol is reported. Three to 15 alleles per locus were detected in 54 beetles collected from four populations of L. decemlineata. The mean number of alleles was 7.4 ± 3.2 and the level of expected heterozygosity ranged from 0.451 to 0.798. The total exclusionary probabilities using these loci for the first and the second parent were 0.982 and 0.999, respectively. Forensics of birds of prey by DNA fingerprinting with 32P-labeled oligonucleotide probes.

Paternity tests on confiscated families of eight species of birds of prey were carried out successfully by DNA fingerprinting with 32P-labeled oligonucleotide probes. Variations in the number of hybridized fragments, depending on the species of birds, are observed using the same probe, as well as differences of polymorphism by hybridizing the DNA samples with several oligonucleotide probes.

Dissemination of wine-related yeasts by migratory birds

Summary The present work was undertaken to evaluate the contribution of migratory birds in the environmental dissemination of yeasts. Four sites (Mazara del Vallo, Lampedusa, Ustica and Linosa), representing the main stop-over points in Sicily, were analysed during spring and autumnal bird migration and 349 birds (belonging to 10 families) were ringed and analysed for yeast presence. Rapid characterization of wild and collection strains of the genus Zygosaccharomyces according to mitochondrial DNA patterns

Several wild and collection strains of the genus Zygosaccharomyces were characterized using a rapid and simple method of restriction analysis of mitochondrial DNA. Patterns obtained with three endonucleases (HaeIII, HinfI and RsaI) made it possible to differentiate each species and to identify the wild strains, isolated from the same spoiled concentrated must, as belonging to the species Z. rouxii. The HinfI restriction enzyme produced a strain-specific pattern which allowed us to recognize that the seven wild isolates belonged to only three strains.

Congruence in genetic markers used to describe Mediterranean and Atlantic populations of European hake (Merluccius merluccius L. 1758)

Summary Eight samples of the hake, Merluccius merluccius L., from the Mediterranean basin (370 fishes total) and one from the Atlantic ocean (50 fishes) were analysed in order to assess genetic variability and describe genetic population structure. Five polymorphic protein coding loci were scored (ADH*, PGI-1*, PGI-2*, PGM* and SOD-1*) in eight samples, together with a haplotype variation of four samples, obtained from polymerase chain reaction/restriction fragment length polymorphism (PCR–RFLP) analysis on the mitochondrial DNA control region. Molecular characterization of Colletotrichum strains derived from strawberry

Strains of Colletotrichum species derived from diseased strawberry plants from a wide geographical range were studied using mitochondrial and ribosomal DNA RFLPs, and acetyl and propionyl esterase isoenzymes. Two major species aggregates were detected, centred on C. acutatum and C. gloeosporioides respectively, with significant further subdivision. There were apparent discrepancies in the hierarchical nesting of some taxon groups based on the different molecular techniques. Strains assigned to C. acutatum fell into several rDNA RFLP groups, but there was less variation in mtDNA RFLP band patterns. Lack of seasonal changes in mitochondrial DNA variability of a Drosophila subobscura population

Restriction site analysis of mtDNA of 550 isofemale lines corresponding to different seasonal samples of a single geographic population of Drosophila subobscura was carried out. The distribution pattern of haplotypes was similar to that observed for the entire range of the species on the European continent: two haplotypes were equally and highly frequent, and a set of sporadic haplotypes were almost never present in more than one seasonal sampling. Molecular characterisation of the species of the genus Zygosaccharomyces.

The restriction fragments polymorphisms of the mitochondrial DNA and the PCR fragment that comprised the internal transcribes spacers and the 5.8S rRNA gene, together with the electrophoretic karyotypes of 40 strains from the 10 species of the genus Zygosaccharomyces, including the new species Z. lentus were examined. The RFLP's of the ITS-5.8S region showed a specific restriction pattern for each species, including the new species Z. lentus. Characterization of polymorphisms in the toxin A and B genes of Clostridium difficile.

We have used six independent polymerase chain reactions (A1–A3 and B1–B3) for amplification of the entire sequence of the two toxin genes tcdA and tcdB of several Clostridium difficile strains. With this approach we have detected (1) restriction site polymorphisms which are distributed all over the genes, and (2) deletions that could be found only in tcdA. Characteristic differences between strains were mainly focused to the 5′ third of tcdB (B1 fragment) and/or the 3′ third of tcdA (A3 fragment). The possible use of our approach for typing of C. difficile toxin genes is discussed.

Genetic typing of C4A and B alleles is routinely carried out by high-voltage agarose gel electrophoresis.
