The role of network connectivity on epileptiform activity.

AbstractA number of potentially important mechanisms have been identified as key players to generate epileptiform activity, such as genetic mutations, activity-dependent alteration of synaptic functions, and functional network reorganization at the macroscopic level. Here we study how network connectivity at cellular level can affect the onset of epileptiform activity, using computational model networks with different wiring properties. The model suggests that networks connected as in real brain circuits are more resistant to generate seizure-like activity. The horse is a herbivorous animal that is used for various purposes, one of them is the production of meat intended for human consumption. Worldwide, horse meat production is led by Asia, leaving Europe in third place. Spain is not a country with a great tradition of consumption of this meat, even so, 18,275 farms are dedicated to it, sacrificing 52,908 horses per year. Physiological dependence and associated withdrawal episodes can constitute a powerful motivational force that perpetuates drug use and abuse. Using robust behavioral models of drug physiological dependence in mice, positional cloning, and sequence and expression analyses, we identified an addiction-relevant quantitative trait gene, Mpdz. Our findings provide a framework to define the protein interactions and neural circuit by which this gene's product (multiple PDZ domain protein) affects drug dependence, withdrawal and relapse.

A novel series of C-3 substituted 4,6-dichloroindole-2-carboxylic acids was synthesized to investigate the influence of different hydrogen-bond donor and acceptor groups at this specific position on the affinity to the glycine site of the NMDA receptor. These novel 3-indolylmethyl derivatives with ring-open (amines, sulfonamides, amides, ureas) and cyclic substituents (imidazolidin-2-ones, (thio)hydantoins) led to the discovery that compounds bearing a hydantoin substituent at the C-3 position of the indole nucleus are the most promising ones. The role of the substantia nigra in the control of amygdaloid paroxysmal activity.

AbstractBoth in acute and chronic cats, focal paroxysmal activity evoked in the ventro-basal complex of the amygdala has been inhibited by substantia nigra conditioning stimulation, to a greater extent, than by caudate nucleus activation. Injection of kainic acid into substantia nigra resulted in the disappearance of the caudate inhibition. It is suggested that the final control, exerted by the striatum on the amygdaloid seizures, occurs by means of the substantia nigra.

Both in acute and chronic cats entopeduncular stimulation inhibits, to a greater extent than caudate activation, focal paroxysmal activity in the ventro-basal complex of the amygdala. Lesion of entopeduncular neurons, by means of kainic acid injection, induces a decrease of the caudate inhibitory effect. It is suggested that neostriatal control of the amygdaloid seizures occurs partly through the globus pallidus.
