Susceptibility to collagen-induced arthritis is modulated by TGFβ responsiveness of T cells

2004

The objective of our study was to determine the regulatory effects that endogenous transforming growth factor beta (TGFbeta) exerts on T cells in the pathogenesis of collagen-induced arthritis (CIA). CIA was induced in transgenic mice expressing a dominant negative TGFbeta type II receptor in T cells under the control of the human CD2 promoter. Clinical and histological arthritis scores were determined and experiments on disease induction and the healing phase of disease were performed. The proliferation and cytokine production of draining lymph node cells in vitro were analyzed. Transgenic mice were more susceptible to induction of CIA. The overall incidence was higher in transgenic mice t…

Mycobacterium tuberculosis Immune Response in Patients With Immune-Mediated Inflammatory Disease

2021

Subjects with immune-mediated inflammatory diseases (IMID), such as rheumatoid arthritis (RA), have an intrinsic higher probability to develop active-tuberculosis (TB) compared to the general population. The risk ranges from 2.0 to 8.9 in RA patients not receiving therapies. According to the WHO, the RA prevalence varies between 0.3% and 1% and is more common in women and in developed countries. Therefore, the identification and treatment of TB infection (TBI) in this fragile population is important to propose the TB preventive therapy. We aimed to study the M. tuberculosis (Mtb) specific T-cell response to find immune biomarkers of Mtb burden or Mtb clearance in patients with different TB …

Chapter 13 Antiphospholipid Antibodies and Vasculitis

2009

Abstract Besides the large group of primary vasculitis diseases, vasculitis in autoimmune diseases is most commonly associated with systemic lupus erythematosus (SLE), scleroderma, rheumatoid arthritis, insulin-dependent diabetes mellitus, and dermatomyositis. The antiphospholipid syndrome (APS), as a relatively recently recognized autoimmune disorder, may also present with a variety of vasculitis features. These non-typical vasculitis syndromes have to be considered very carefully in the APS, and vasculitis manifestations presenting as systemic involvement in APS patients have to be identified very early to prevent patients from rapidly developing organ damage. The aim of this review artic…

Endothelial progenitor cells: Are they displaying a function in autoimmune disorders?

2016

Endothelial Progenitor Cells (EPCs) are bone marrow derived cells able to differentiate in mature endothelial cells (EC) contributing to the generation of new vessels, connecting to fibronectin, and forming colonies and/or colony forming units. Since circulating EPCs can be actively considered part of endothelial damage in several cardiovascular diseases and autoimmune disorders the possibility to have a measure for endothelium damage should be considered of interest to predict the patient out-come. At the same time the EPCs proliferative and regenerative role could be considered for therapeutic applications. Studies have been performed to elucidate the role of EPCs in Systemic Sclerosis an…

ChemInform Abstract: Cyclooxygenase Inhibitors - Current Status and Future Prospects

2010

Prostaglandins are formed from arachidonic acid by the action of cyclooxygenase and subsequent downstream synthetases. Two closely related forms of the cyclooxygenase have been identified which are now known as COX-1 and COX-2. Both isoenzymes transform arachidonic acid to prostaglandins, but differ in their distribution and their physiological roles. Meanwhile, the responsible genes and their regulation have been clarified. COX-1, the pre-dominantly constitutive form of the enzyme, is expressed throughout the body and performs a number of homeostatic functions such as maintaining normal gastric mucosa and influencing renal blood flow and platelet aggregation. In contrast, the inducible for…

AB1358 Infectious antibodies repertoire in rheumatoid arthritis

2018

Background The incidence of infectious diseases in the RA ADAPTHERA study by ELISA antibody screening and the differences in infectious distribution in active or low active disease patients was explored. Methods Sera from 88 naive RA patients out of the ADAPTHERA study cohort, disease duration Results 82% RA patients were found to be positive for HSV1 +2 IgG (2% IgM positive), 8% for Adenovirus (IgA), 77% (IgG), and 1% (IgM). 99% for EBV-IgG (no IgM positive). 53% (IgG), and 26% (IgM) for CMV. 38% for HP-IgG and 15% for IgA and 79% for P-B19-IgG (3% IgM). 6% for Borrelia-IgM and 14% for IgG. A slightly increase was found for EBV sera positivity (99% IgG), compared to the normal population. …

AB1035 INTESTINAL MICROBIOTA COMPOSITION OF ADULT PATIENTS WITH FAMILIAL MEDITERRANEAN FEVER AND HEALTHY CONTROLS (THE RHEUMA-BIOTA STUDY)

2020

Background:Although Familial Mediterranean Fever (FMF) is a monogenic disease, microbiota composition may play role in the pathogenesis or phenotypic expression. Objectives:We aim to evaluate the intestinal microbiota composition in patients with FMF and to compare with healthy controls. Methods:In this prospective cohort study, a group of 10 adult patients with FMF and 10 age-appropriate healthy controls, for which there was strict inclusion/exclusion, were enrolled. Fecal samples were stored at -80°C until DNA extraction. A region of the 16S rRNA gene (V3-V4) was selected and sequencing was performed on the Illumina MiSeq platform at the Sequencing and Bioinformatics Service of FISABIO foun…

Effects of exercise on life quality in a rheumatoid arthritis patient

2020

Rheumatoid arthritis (RA) is a chronic-degenerative disease characterized by symptoms that significantly impact on the functional capacity and thus, the ability to carry out daily functional activities reducing the quality of life (QoL). The aim was to evaluate the effects of a strength and conditioning (S&C) program on the quality of life (QoL), the self-perception disease impact, the self-fatigue perception and cardiovascular risk factors in a patient with rheumatoid arthritis (RA) and cardiovascular disease (CVD). 34-year-old woman (height: 1.40m; weight: 36.1kg) diagnosed with RA and CVD underwent a six-month S&C program with aerobic training 2-3 times a week in 1-3 sets of 5-15 minutes…

Fluorine-containing drugs approved by the FDA in 2019

2020

Abstract Eleven new fluorine-containing FDA-approved drugs have been profiled and details of their discovery and preparation are discussed. Therapeutic areas include schizophrenia, migraine, multiple sclerosis, insomnia, rheumatoid arthritis, anti-tuberculosis, breast cancer, lymphoma kinase inhibitor, serotonin receptor antagonist. New pharmaceuticals feature four examples of aromatic fluorine, three aromatic CF3 group, three aliphatic CF3 and one compound with aromatic CF3O group. Furthermore, among the new compounds, six are chiral and seven are derived from tailor-made amino acids.

ChemInform Abstract: Investigations Concerning the COX/5-LOX Inhibiting and Hydroxyl Radical Scavenging Potencies of Novel 4,5-Diaryl Isoselenazoles.

2008

The aim of this study was to investigate 4,5-diaryl isoselenazoles as multiple target non-steroidal anti-inflammatory drugs (MTNSAIDs) which can intervene into the inflammatory processes via different mechanisms of action creating a new class of compounds. Here we describe the synthesis of COX/LOX inhibitors which additionally reduce the level of reactive oxygen species, such as hydroxyl radicals which are well known for supporting inflammation processes in Parkinson's disease, Alzheimer's disease and rheumatoid arthritis.
