Site-Specific Dual-Labeling of a VHH with a Chelator and a Photosensitizer for Nuclear Imaging and Targeted Photodynamic Therapy of EGFR-Positive Tum…

2021

Simple Summary Variable domains of heavy chain only antibodies are small proteins that can be used for tumor imaging and therapy upon conjugation of functional groups. As frequently used random conjugation techniques can decrease binding to the target of interest, site-specific conjugation of these functional groups is preferred. Here, we optimized site-specific conjugation of both a chelator for binding of a radiometal and a photosensitizer to epidermal growth factor receptor (EGFR) binding VHH 7D12. We characterized this dual-labeled VHH for nuclear imaging and targeted photodynamic therapy of EGFR-expressing tumors. Abstract Variable domains of heavy chain only antibodies (VHHs) are valu…

Investigating the role of self‐control beliefs in predicting exercise behaviour: A longitudinal study

2021

Background: Engaging in exercise behaviour regularly requires a repeated investment of resources to reap the health benefits. An individual’s self-control resources, when performing a behaviour can be perceived as being recharged or depleted. The investigation on how self-control beliefs resources predict exercise behaviour is very limited in the literature. The purpose of this study was to understand how self-control beliefs predict exercise behaviour across time in a physical activity model. Methods: Participants (N = 161) were a sample of adults recruited across twelve gyms and recreation centres in a large city. Participants completed surveys across five months. Data were analysed using…

De novogenome assembly of the land snailCandidula unifasciata(Mollusca: Gastropoda)

2021

AbstractAmong all molluscs, land snails are an economically and scientifically interesting group comprising edible species, alien species and agricultural pests. Yet, despite its high diversity, the number of whole genomes publicly available is still scarce. Here, we present the draft genome assembly of the land snailCandidula unifasciata, a widely distributed species along central Europe, which belongs to Geomitridae family, a group highly diversified in the Western-Palearctic region. We performed a whole genome sequencing, assembly and annotation of an adult specimen based on PacBio and Oxford Nanopore long read sequences as well as Illumina data. A genome of about 1.29 Gb was generated w…

Psychotherapeutically Oriented Vibroacoustic Therapy for Functional Neurological Disorder: A Pilot Study

2021

Functional Neurological Disorder (FND) affects a significant number of people worldwide. Previously referred to as conversion disorder, FND is a disorder of the communication between mind and body resulting in the experience of neurological symptoms incompatible with neurological or medical diagnoses. FND patients account for a notable portion of neurologists' patients, and yet these patients are still considered some of the most difficult to diagnose and treat. This pilot case study utilized a psychotherapeutically oriented approach to vibroacoustic therapy and active music therapy methodology in the therapy process of a patient diagnosed with FND. The treatment protocol used in this study…

Amelioration of the abnormal phenotype of a new L1 syndrome mouse mutation with L1 mimetics

2021

L1 syndrome is a rare developmental disorder characterized by hydrocephalus of varying severity, intellectual deficits, spasticity of the legs, and adducted thumbs. Therapy is limited to symptomatic relief. Numerous gene mutations in the L1 cell adhesion molecule (L1CAM, hereafter abbreviated L1) were identified in L1 syndrome patients, and those affecting the extracellular domain of this transmembrane type 1 glycoprotein show the most severe phenotypes. Previously analyzed rodent models of the L1 syndrome focused on L1-deficient animals or mouse mutants with abrogated cell surface expression of L1, making it difficult to test L1 function-triggering mimetic compounds with potential therapeu…

Why are viral genomes so fragile? The bottleneck hypothesis

2021

If they undergo new mutations at each replication cycle, why are RNA viral genomes so fragile, with most mutations being either strongly deleterious or lethal? Here we provide theoretical and numerical evidence for the hypothesis that genetic fragility is partly an evolutionary response to the multiple population bottlenecks experienced by viral populations at various stages of their life cycles. Modelling within-host viral populations as multi-type branching processes, we show that mutational fragility lowers the rate at which Muller’s ratchet clicks and increases the survival probability through multiple bottlenecks. In the context of a susceptible-exposed-infectious-recovered epidemiolog…

The importance of social value in agroecological farms: adjusting the common good balance sheet to improve their sustainable management

2021

Organic farming offers the producers the possibility of focusing on their business while obtaining a competitive advantage. Nevertheless, this is centered in specific activities of the production process, thus only creating environmental value. Therefore, it is not an approach based on sustainability. As a consequence of this, it becomes necessary to enlarge this approach toward people (social value) and profitability (economic value). The objective of this work is to describe a tool based on the Economy for the Common Good model applied to the agroecological sector. We propose to adapt the Common Good Matrix to the characteristics of agroecological farms to offer a useful tool to manage an…

Analysis of Lipid Peroxidation by UPLC-MS/MS and Retinoprotective Effects of the Natural Polyphenol Pterostilbene

2021

The loss of redox homeostasis induced by hyperglycemia is an early sign and key factor in the development of diabetic retinopathy. Due to the high level of long-chain polyunsaturated fatty acids, diabetic retina is highly susceptible to lipid peroxidation, source of pathophysiological alterations in diabetic retinopathy. Previous studies have shown that pterostilbene, a natural antioxidant polyphenol, is an effective therapy against diabetic retinopathy development, although its protective effects on lipid peroxidation are not well known. Plasma, urine and retinas from diabetic rabbits, control and diabetic rabbits treated daily with pterostilbene were analyzed. Lipid peroxidation was evalu…

Homodyne Solid-State Biased Coherent Detection of Ultra-Broadband Terahertz Pulses with Static Electric Fields.

2021

We present an innovative implementation of the solid-state-biased coherent detection (SSBCD) technique, which we have recently introduced for the reconstruction of both amplitude and phase of ultra-broadband terahertz pulses. In our previous works, the SSBCD method has been operated via a heterodyne scheme, which involves demanding square-wave voltage amplifiers, phase-locked to the THz pulse train, as well as an electronic circuit for the demodulation of the readout signal. Here, we demonstrate that the SSBCD technique can be operated via a very simple homodyne scheme, exploiting plain static bias voltages. We show that the homodyne SSBCD signal turns into a bipolar transient when the stat…

Quantum correlations in PT -symmetric systems

2021

Abstract We study the dynamics of correlations in a paradigmatic setup to observe PT -symmetric physics: a pair of coupled oscillators, one subject to a gain one to a loss. Starting from a coherent state, quantum correlations (QCs) are created, despite the system being driven only incoherently, and can survive indefinitely. Both total and QCs exhibit different scalings of their long-time behavior in the PT -broken/unbroken phase and at the exceptional point (EP). In particular, PT symmetry breaking is accompanied by non-zero stationary QCs. This is analytically shown and quantitatively explained in terms of entropy balance. The EP in particular stands out as the most classical configuration…

Dual Antimicrobial and Antiproliferative Activity of TcPaSK Peptide Derived from a Tribolium castaneum Insect Defensin

2021

Antimicrobial peptides (AMPs) found in the innate immune system of a wide range of organisms might prove useful to fight infections, due to the reported slower development of resistance to AMPs. Increasing the cationicity and keeping moderate hydrophobicity of the AMPs have been described to improve antimicrobial activity. We previously found a peptide derived from the Tribolium castaneum insect defensin 3, exhibiting antrimicrobial activity against several human pathogens. Here, we analyzed the effect against Staphyloccocus aureus of an extended peptide (TcPaSK) containing two additional amino acids, lysine and asparagine, flanking the former peptide fragment in the original insect defensi…

Polymers with antiviral properties: A brief review.

2021

Viruses that are pathogenic to humans and livestock pose a serious epidemiological threat and challenge the world's population. The SARS-CoV-2/COVID-19 pandemic has made the world aware of the scale of the threat. The surfaces of various materials can be a source of viruses that remain temporarily contagious in the environment. Few polymers have antiviral effects that reduce infectivity or the presence of a virus in the human environment. Some of the effects are due to certain physical properties, e.g., high hydrophobicity. Other materials owe their antiviral activity to a modified physicochemical structure favoring the action on specific virus receptors or on their biochemistry. Current re…

Third-harmonic light polarization control in magnetically resonant silicon metasurfaces

2021

Nonlinear metasurfaces have become prominent tools for controlling and engineering light at the nanoscale. Usually, the polarization of the total generated third harmonic is studied. However, diffraction orders may present different polarizations. Here, we design an high quality factor silicon metasurface for third harmonic generation and perform back focal plane imaging of the diffraction orders, which present a rich variety of polarization states. Our results demonstrate the possibility of tailoring the polarization of the generated nonlinear diffraction orders paving the way to a higher degree of wavefront control.

Fabrication of an extremely cheap poly(3,4-ethylenedioxythiophene) modified pencil lead electrode for effective hydroquinone sensing

2021

Hydroquinone (HQ) is one of the major deleterious metabolites of benzene in the human body, which has been implicated to cause various human diseases. In order to fabricate a feasible sensor for the accurate detection of HQ, we attempted to electrochemically modify a piece of common 2B pencil lead (PL) with the conductive poly(3,4-ethylenedioxythiophene) or PEDOT film to construct a PEDOT/PL electrode. We then examined the performance of PEDOT/PL in the detection of hydroquinone with different voltammetry methods. Our results have demonstrated that PEDOT film was able to dramatically enhance the electrochemical response of pencil lead electrode to hydroquinone and exhibited a good linear co…

On a Retarded Nonlocal Ordinary Differential System with Discrete Diffusion Modeling Life Tables

2021

In this paper, we consider a system of ordinary differential equations with non-local discrete diffusion and finite delay and with either a finite or an infinite number of equations. We prove several properties of solutions such as comparison, stability and symmetry. We create a numerical simulation showing that this model can be appropriate to model dynamical life tables in actuarial or demographic sciences. In this way, some indicators of goodness and smoothness are improved when comparing with classical techniques.

The relationship between workload and burnout among nurses: The buffering role of personal, social and organisational resources

2021

Workload in the nursing profession is high, which is associated with poor health. Thus, it is important to get a proper understanding of the working situation and to analyse factors which might be able to mitigate the negative effects of such a high workload. In Germany, many people with serious or life-threatening illnesses are treated in non-specialized palliative care settings such as nursing homes, hospitals and outpatient care. The purpose of the present study was to investigate the buffering role of resources on the relationship between workload and burnout among nurses. A nationwide cross-sectional survey was applied. The questionnaire included parts of the Copenhagen Psychosocial Qu…

Etude du rôle de STING dans la biologie des lymphocytes T CD4 : application en immunothérapie anticancéreuse

2021

Recognition of danger signals by intracellular or extracellular receptors expressed by innate immune cells is crucial for the establishment of an effective adaptive immune response. STING (Stimulator of Interferon Genes) is a protein located in the endoplasmic reticulum discovered in 2008 as being essential for the detection of cytosolic DNAs and the establishment of antiviral immune response mediated by type I interferons (IFNs). The STING protein also plays a key role in spontaneous anti-tumor immune responses and the administration of STING ligands in tumor-bearing mice favors tumor growth control. These observations have been verified in clinical trials in patients with melanoma, illust…

Survey of five major grapevine viruses infecting Blatina and Žilavka cultivars in Bosnia and Herzegovina

2021

The sanitary status of grapevines has not yet been considered sufficiently in vineyards throughout Bosnia and Herzegovina (BiH). An extensive survey of five major grapevine viruses in the country was carried out in 2019. A total of 630 samples from the two dominant autochthonous cultivars, named Žilavka and Blatina, were tested by DAS-ELISA for the presence of grapevine leafroll-associated viruses (GLRaV-1 and 3), grapevine fleck virus (GFkV), grapevine fanleaf virus (GFLV) and Arabis mosaic virus (ArMV). Eighty-eight % of the samples were positive for at least one virus, and all five viruses were detected, thought with different incidence, i.e. GLRaV-3 (84%), GFLV (43%), GLRaV-1 (14%), GFk…

Public Policies to Promote Sustainable Transports : Lessons from Valencia

2021

Bicycling appears in the literature on urban mobility as a more sustainable transportation mode for future transportation, based on empirical evidence of the potential benefits of bicycling on the environment, society, and health. In this context, public interventions to promote and maintain bicycling as a sustainable practice and its positive effects are salient. This article reviews different cycling policies with respect to cycling facilities present in the literature and compares them with a case study in Valencia (Spain). We collected information on the public policies implemented in Valencia and the traffic intensity in bike lanes over detecting loops distributed around the city. The …

Teacher education programmes in physical education and sport at master’s level: Opportunities and challenges

2021

An updated checklist and biogeography of the Sardinian large branchiopods, with a focus on Spinicaudata (Crustacea, Branchiopoda)

2021

Author(s): Marrone, Federico; Alfonso, Giuseppe; Cottarelli, Vezio; Botta, Marco Massimo; Koepp, Christian; Stoch, Fabio | Abstract: The large branchiopod fauna of Sardinia is reviewed based both on literature and newly collected data. Based on the available evidence, 13 taxa are present on the island (8 Anostraca, 2 Notostraca, and 3 Spinicaudata). Among them, the finding of the spinicaudatan Leptestheria dahalacensis is new for Sardinia, while the spinicaudatans Cyzicus bucheti and Eulimnadia sp. were overlooked in the most recent synopses on the fauna of the island due to misidentifications. Conversely, Cyzicus tetracerus and Limnadia lenticularis, previously erroneously reported based o…

"You helped me out of that darkness" Children as dialogical partners in the collaborative post-family therapy research interview.

2021

Applying Dialogical Methods for Investigations of Happening of Change (DIHC), this study investigated how children who had been diagnosed with an oppositional defiant or conduct disorder participated in a collaborative post‐therapy research interview and talked about their experiences of family therapy. The results showed that the children participated as dialogical partners talking in genuine, emotional, and reflective ways. Encountered as full‐membership partners, the children also co‐constructed meanings for their sensitive experiences. However, their verbal initiatives and responses appeared in very brief moments and could easily have been missed. The collaborative post‐therapy intervie…

Semantic processing in children with autism spectrum disorder

2021
