Presentation of insulin and insulin A chain peptides to mouse T cells: involvement of cysteine residues.

The requirements for insulin presentation and recognition by A alpha b A beta b- and A alpha b A beta k-restricted mouse T cells were studied using a variety of derivatives of the insulin A chain. It was found that A chain peptides with irreversibly blocked Cys residues are non-stimulatory for the T cells. This suggests that at least one of the Cys residues is essential for recognition. On the other hand, all A chain peptides containing Cys residues modified in a way reversible by reaction with thiols are stimulatory yet differ in antigenic potency. Potassium antimonate was used to precipitate calcium in the cochlea of the guinea-pig. The distribution of the calcium antimonate precipitates was analysed by electron microscopy. The precipitate density was determined in different cell types in the organ of Corti by counting the number of calcium binding sites in a 10-micron 2 area. The size of the precipitates varied considerably, and thus the relative amount of the precipitable calcium was estimated only semi-quantitatively. The electrochemical oxidation of 2,5-diphenyl-3-acetylpyrrole (I) is described. The cyclic derivative 1,6a-dihydro-2,5,6a-triphenyl-3,4-diacetylbenzo[g]pyrrolo[3,2-e]indole (II) was obtained in very good yield. However, when water was present in the reaction medium, a different derivative, 4-acetyl-2-hydroxy-2,5-diphenyl-3-(4′-acetyl-2′,5′-diphenyl-3′-yl)-2H-pyrrole (III), was obtained as the main product. 2,2′,5,5′-Tetraphenyl-4,4′-diacetyl-3,3′-dipyrryl (IV), a potentially useful intermediate for the synthesis of condensed pyrroles, was synthesized by zinc reduction of III.

Lasing characteristics of a new DCM derivative under flashlamp pumping

Abstract A new DCM derivative (4-dicyanomethylene-2-methyl-6-p-diethanolamino-styryl 4H-pyran or DCM-OH), was tested under flashlamp pumping. This dye lased in the 600–700 nm range with efficiency up to 0.4% at maximum. Lasing performances of DCM-OH in methanol and water-methanol mixture are reported and compared to normal DCM.

Adamantane as a Building Block of New Araliphanes – Synthesis, Spectroscopy, and Crystal Structures Exchange of benzene units for aliphatic building blocks in cyclophanes leads to new molecules of the “araliphane” type. With adamantane the araliphanes 2 – 4 are synthesized. Their stereochemical behavior as shown by NMR studies and X-ray crystallographic analyses differs significantly from that of their aromatic counterparts. The signals of the intraannular adamantane hydrogens are strongly shifted upfield up to δ=– 2.18 (in 2). 4 is obtained as a racemic mixture of enantiomers (in the crystalline state) and shows a boat-like deformation of the benzene moiety.

The concept of major depression. III. Concurrent validity of six competing operational definitions for the clinical ICD-9 diagnosis.

The comparative validity of six operational diagnoses of major depression was evaluated in 600 psychiatric inpatients using the independently assessed clinical ICD-9 diagnoses as a yardstick. Agreement with, and positive predictive value for the ICD-9 categories of pure (endogenous and psychogenic) depression served as validation criteria; sensitivity of major depression diagnoses for detecting ICD-9 bipolar depressions was additionally used for examining the adequacy of width, time and exclusion criteria of the competing operational definitions. Midbrain vs. pontine medial longitudinal fasciculus lesions: The utilization of masseter and blink reflexes

Masseter (MR) and blink reflexes (BL) were investigated in 51 patients with internuclear ophthalmoplegia (INO) due to multiple sclerosis (28) and lacunar infarction (23). The MR was abnormal in 20 of 23 cases with bilateral INO and in 21 of 28 with unilateral INO. The R1 component of the BL (BL-R1) was abnormal in 7 of 23 patients with bilateral INO and 10 of 28 with unilateral INO. Combined MR and BL-R1 changes occurred in 8 of 28 cases with unilateral INO and 7 of 23 with bilateral INO. Electronic Shell Structure and the Crystal Field Splitting in Simple Metals Clusters

An upper limit for the number of atoms in metal clusters capable of exhibiting electronic shell structure has been estimated by comparing the energy difference between the highest occupied and the lowest unoccupied state with the crystal field splitting. The former is obtained by solving the Schrodinger equation for a spherical potential well with hard walls while the latter is obtained from the band structure of the solid. The results indicate that shell structures may persist in clusters containing as many as a million atoms.

This program is written in FORTRAN-77 for IBM 30/90 and VAX 8300 computers, and permits the simultaneous evaluation of both rate constants and initial reagent concentrations or, alternatively, rate constants and sensitivities. Coamplexation of mercury(II) compounds by crown ethers in organic media

Complexes between crown ethers having ring sizes of 15–24 atoms and 5 to 8 oxygens with HgX2 species (X = Cl, I, CN, SCN) have been prepared and studied. Interactions of the substituents X bound to mercury with groups or fragments in the crown compound are essential in determining the characteristics and stability of the complexes formed. Complexes with crowns of 18C6 ring sizes are the most favourable except for HgX2 compounds for which the size of X is larger than the macrocycle ring, as is the case for X = CF3.

The suitability of water as an oral or rectal contrast medium for abdominal CT was studied in 56 patients and compared with an iodine-containing water-soluble contrast medium (ioxital amino acid). In some cases it was impossible to differentiate gastrointestinal structures from extraluminal fluid collections (cystic tumours, ascites, abscesses) and there was poor filling of distal small bowel and colon. The routine use of water can, therefore, not be recommended. In some cases, however, water can result in improved image quality by reducing artifacts and improving the demonstration of the mucosa.

Abstract Isomerization of all-trans to 11-cis retinol has been studied in a membrane preparation from the nuclear fraction of bovine retinal pigment epithelium. When the nuclear membrane preparation deprived of endogenous retinoids is incubated with 4·5 μ m all-trans-retinol, the mean value calculated for the isomerase activity is 1·32 nmol 11-cis retinol formed hr−1 mg protein−1. Simultaneous formation of all-trans and 11-cis retinyl esters is also observed in the nuclear preparation. The open reading frame of the first intron of the mitochondrial cox1 gene (cox1I1) was expressed in Escherichia coli. The putative intron-encoded protein stimulated the formation of intra-chromosomal lac +-recombinants about threefold. No stimulation was found when the reading frame was inserted in the opposite direction, or when it was interrupted by a deletion. The intronic open reading frame did not complement recA − or recB − mutants of E. coli. In S. pombe, elimination of this intron did not abolish homologous recombination in mitochondria. A possible role of the recombinase activity in yeast mitochondria will be discussed.

Powder x-ray diffraction, d.c. and a.c. susceptibilities, and SEM have been used to study (Bi1−xPbx)2Sr2Ca2Cu3O10−δ fibers grown by the Laser Floating Zone method. The well-oriented, long-grained superconductor fiber properties are shown to be highly dependent on the partial pressure of oxygen in the growth atmosphere, as well as on fiber pulling rate. Slowly grown fibers contain initially the 2212 (80 K) phase; the 2223 (110 K) phase also appears upon annealing in air. Faster growth rates result in fibers that contain a mixture of the 2212 and 2201 phases and, in this case, long annealing procedures are necessary to observe the 2223 phase.

Summary Levels of free polyamines were monitored in needles of spruce trees [ Picea abies (L.) Karst.] during the course of two vegetation periods at five natural sites. The putrescine concentration in needles of damaged spruce trees was significantly higher than in needles of undamaged trees. This is e.g. impressively proved by the putrescine/spermidine-ratio. While undamaged trees show nearly similar putrescine and spermidine concentrations, damaged trees have five- to tenfold higher putrescine levels depending on the increasing state of damage. 38 cases of acute multilocular osteomyelitis out of a total of 165 osteomyelitis patients were treated between 1974 and 1986 at the Department of Paediatric Surgery and Paediatric Diseases at the University of Mainz. Osteomyelitis in infants was separated from juvenile osteomyelitis because of the different problems inherent to this disease depending upon the age; likewise, multilocular remote involvement was separated from regional multiple involvement. We investigate the limits of applicability of the Spitzer-Harm thermal conductivity in solar coronal loops and show that the ratio λ0/LTof electron mean-free path to temperature scale height in large-scale structures can approach the limits of the Spitzer-Harm theory. In a search for Schizosaccharomyces pombe mutants resistant to the antifungal agent papulacandin B, a morphological mutant was isolated. The mutant is round shaped in contrast to the rod shaped parental strain. Results of intracoronary stents for management of coronary dissection after balloon angioplasty.

Dissections after coronary balloon angioplasty are risk factors for acute or subacute vessel closures. Intracoronary stenting was developed to avoid these complications by pressing the intimal and medial flaps against the vessel wall, thus reducing the risk of acute thrombosis. A total of 22 stents were implanted into the coronary arteries of 15 patients with dissections after balloon angioplasty causing angina pectoris or ischemic electrocardiographic changes. Effect of obesity on left ventricular function studied by radionuclide angiocardiography

Several studies have shown a significant association of obesity with cardiovascular morbidity and mortality. The present study was carried out to investigate central and systemic haemodynamics in overweight and moderate obese, but otherwise healthy subjects, and in a lean control group to determine whether obesity can influence left ventricular performance per se. In this study an attempt has been made to eliminate misleading factors, such as diabetes, lipid abnormalities and hypertension. The Dissociation Constant of Pyruvic Acid: Determination by Spectrophotometric Measurements

Aqueous solutions of sodium pyruvate/hydrochloric acid mixtures were studied with regard to changes in the intensity of optical absorption at 317 nm wavelength, caused by varying pH and temperature. The data were evaluated to determine the pyruvic acid apparent dissociation constant at infinite dilution. Its temperature dependence followed the relation ln Kod = −(0.55 ± 0.47) − (1538 ± 135)/T or, if previous results of Pedersen at two temperatures are included, In Kod = −(0.97 ± 0.35) − (1418 ± 102)T. High-spin states in 173Os have been studied by γγ-coincidence measurements following the 146Nd(32S, 5n)173Os reaction. Additional information has also been extracted from the reaction 144Sm(32S, 2pn)173Os. Four main sequencies have been identified as the two signature branches of the bands built on the [642]52+ and [523]52− Nilsson states. The band features have been analysed within the framework of the cranked shell model. Contribution of X-Ray diffraction to the study of spin transitions in some iron (II) complexes

Abstract A review of recent single crystal investigations under constraint of iron (II) spin crossover complexes is given. The systems covered are Fe L2 (NCS)2 complexes (L = Phen, Btz) and some analogous derivatives. Emphasis is laid on the correlations between structural changes associated with spin transitions and the experimentally determined characteristics of these transitions.

Effect of 3,4-dichloroaniline on the early life stages of the zebrafish (Brachydanio rerio): results of a comparative laboratory study.

An early life-stage (ELS) test was conducted with the zebrafish and 3,4-dichloroaniline (3,4-DCA) in eight laboratories. Based on the results from all eight laboratories, an LOEC of 200 micrograms 3,4-dichloroaniline/liter applies for the early life stages of the zebrafish. Effects observed were reduction of the survival rate and malformations. If the 100 micrograms/liter concentration additionally tested by one laboratory is included in the assessment, an LOEC of 100 micrograms 3,4-DCA/liter is obtained for the survival rate and increase in length. Actin in Allomyces arbuscula

International audience; A 42 kDa protein was isolated by affinity chromatography on DNAse I-Sepharose from 24-h-old mycelia of Allomyces arbuscula. It was identified as actin by immunoblots with monoclonal antibody probe against a chicken gizzard actin. This fungal actin has a pI of 5.9 and separates into two spots on two-dimensional polyacrylamide gel electrophoresis, suggesting its dual nature. It can polymerize into 8-10 nm filaments visualized by electron microscopy.

New solutions of the hamiltonian and diffeomorphism constraints of quantum gravity from a highest weight loop representation

Abstract We introduce a highest weight type representation of the Rovelli-Smolin algebra of loop observables for quantum gravity. In terms of this representation, new solutions of the hamiltonian and diffeomorphism constraints are given. Assuming the locality of the quantum hamiltonian constraint we show that any functional depending on the generalized link class of the disjoint union of arbitrary simple loops is a solution. Finally we argue that this is the general solution in the irreducible representation space.

These reactions provide a new access to heterocyclic functionalized indoles and pyrido[c]annellated carbazoles, compounds of interest as potential antidepressive and antitumor active agents.

Severe infantile colic and food intolerance: A long-term prospective study

To determine the relationship between infantile colic and cow's milk protein intolerance (CMPI) in formula-fed infants, 70 infants (38 male, 32 female) were selected, with mean age 30.2 +/- 21.4 days, with severe colic (duration of crying greater than 4 h per day for 5 days per week). In 50 of the infants in the study group (71.4%) there was a remission of symptoms when cow's milk protein (CMP) was eliminated from the diet. Two successive challenges caused the return of symptoms in all these 50 infants.
