Stromal and Immune Cell Dynamics in Tumor Associated Tertiary Lymphoid Structures and Anti-Tumor Immune Responses

2022

Tertiary lymphoid structures (TLS) are ectopic lymphoid organs that have been observed in chronic inflammatory conditions including cancer, where they are thought to exert a positive effect on prognosis. Both immune and non-immune cells participate in the genesis of TLS by establishing complex cross-talks requiring both soluble factors and cell-to-cell contact. Several immune cell types, including T follicular helper cells (Tfh), regulatory T cells (Tregs), and myeloid cells, may accumulate in TLS, possibly promoting or inhibiting their development. In this manuscript, we propose to review the available evidence regarding specific aspects of the TLS formation in solid cancers, including 1) …

Developing human/AI interactions for chat-based customer services: lessons learned from the Norwegian government

2022

Advancements in human/AI interactions led to smartification of public services via the use of chatbots. Here, we present findings from a clinical inquiry research project in a key public service organisation in Norway. In this project, researchers and practitioners worked together to generate insights on the action possibilities offered to human service agents by chatbots and the potential for creating hybrid human/AI service teams. The project sensitised service agents to discover affordances based on their actual practices, rather than on the predefined use of chatbots. The different affordances identified can be useful for practitioners who design and deploy chatbot-based services. The a…

Modeling a Novel Variant of Glycogenosis IXa Using a Clonal Inducible Reprogramming System to Generate "Diseased" Hepatocytes for Accurate Diagnosis.

2022

The diagnosis of inherited metabolic disorders is a long and tedious process. The matching of clinical data with a genomic variant in a specific metabolic pathway is an essential step, but the link between a genome and the clinical data is normally difficult, primarily for new missense variants or alterations in intron sequences. Notwithstanding, elucidation of the pathogenicity of a specific variant might be critical for an accurate diagnosis. In this study, we described a novel intronic variant c.2597 + 5G > T in the donor splice sequence of the PHKA2 gene. To investigate PHKA2 mRNA splicing, as well as the functional consequences on glycogen metabolism, we generated hepatocyte-like ce…

Physical activity and health: Findings from Finnish monozygotic twin pairs discordant for physical activity.

2022

Genetic and early environmental differences including early health habits associate with future health. To provide insight on the causal nature of these associations, monozygotic (MZ) twin pairs discordant for health habits provide an interesting natural experiment. Twin pairs discordant for leisure-time physical activity (LTPA) in early adult life are thus a powerful study design to investigate the associations between long-term LTPA and indicators of health and wellbeing. We have identified 17 LTPA discordant twin pairs from two Finnish twin cohorts and summarize key findings of these studies in this paper. The carefully characterized rare long-term LTPA discordant MZ twin pairs have part…

Keratin/alginate hybrid hydrogels filled with halloysite clay nanotubes for protective treatment of human hair

2022

Keratin/alginate hydrogels filled with halloysite nanotubes (HNTs) have been tested for the protective coating of human hair. Preliminary studies have been conducted on the aqueous colloidal systems and the corresponding hydrogels obtained by using Ca2+ ions as crosslinkers. Firstly, we have investigated the colloidal properties of keratin/alginate/HNTs dispersions to explore the specific interactions occurring between the biomacromolecules and the nanotubes. Then, the rheological properties of the hydrogels have been studied highlighting that the keratin/alginate interactions and the subsequent addition of HNTs facilitate the biopolymer crosslinking. Finally, human hair samples have been t…

Correspondence between theory and practice of a Beerkan infiltration experiment

2022

The Beerkan infiltration experiment is carried out by inserting the ring a short depth into the soil and establishing a positive head of water on the infiltration surface for at least a part of the run. Nevertheless, the data are analyzed by assuming a fully unconfined infiltration process (ring insertion depth, d = 0 cm) and a null ponded depth of water (H = 0 cm). The influence of ring insertion and ponded water on an infiltration process of 2 h sampled every minute was tested in this numerical investigation. Five soils varying from sand to silt loam, three ring radii (5–15 cm), and the Beerkan-specific range of values for both d and H (between 0 and 1 cm) were considered. The differences…

Analytical models for the pulse shape of a superconductor-ferromagnet tunnel junction thermoelectric microcalorimeter

2022

AbstractThe superconductor-ferromagnet thermoelectric detector (SFTED) is a novel ultrasensitive radiation detector based on the giant thermoelectric effect in superconductor-ferromagnet tunnel junctions. We demonstrate analytical models and solutions in the time domain for a SFTED operated as a microcalorimeter (pulse excitation), in the linear small-signal limit. Based on these solutions, the signal current and temperature pulse response were studied for two different electrical circuit models, providing design conditions for stable and non-oscillatory response.Kindly check and confirm whether the corresponding author is correctly identified. The corresponding author is correct.

Permanent deformation and fatigue damage interaction in asphalt concrete using energy approach

2022

The interaction of fatigue and permanent deformation is very complex phenomenon and little attempt is made on this topic. This paper presents an investigation on the interaction of the two damages using the energy approach. Laboratory tests were conducted for both fatigue and permanent deformation on two different asphalt concrete in a sequential test procedure. A new failure criterion is proposed based on the dissipated energy ratio (DER). The proposed criterion for permanent deformation gives more damage indicator (inflection points) than the conventional flow number/strain rate criterion. The sequential test procedure is also found economical that can be standardized. From the study, str…

Estimation of durability of new surface courses using accelerated load test and expert's opinions

2022

There is an increasing interest among road authorities in studying sustainability assessment of road infrastructure. The durability of the surface course is a critical factor in these calculations and must be carefully considered in the analysis. New types of asphalt mixes with the potential to improve the results in the sustainability assessment have no proven track record and are thus more likely not to be used until solid performance data is available. To bridge the gap and diminish the uncertainty about the durability of Green asphalt mixes for surface courses, a combination of laboratory tests and tests in a circular road simulator were performed. Eight mixes were tested in total, four…

Post-Quantum Secure Identity-Based Encryption Scheme using Random Integer Lattices for IoT-enabled AI Applications

2022

Identity-based encryption is an important cryptographic system that is employed to ensure confidentiality of a message in communication. This article presents a provably secure identity based encryption based on post quantum security assumption. The security of the proposed encryption is based on the hard problem, namely Learning with Errors on integer lattices. This construction is anonymous and produces pseudo random ciphers. Both public-key size and ciphertext-size have been reduced in the proposed encryption as compared to those for other relevant schemes without compromising the security. Next, we incorporate the constructed identity based encryption (IBE) for Internet of Things (IoT) …

A Cogging Torque Minimization Procedure for Interior Permanent Magnet Synchronous Motors Based on a Progressive Modification of the Rotor Lamination …

2022

This article describes a simple cogging torque minimization procedure for interior permanent magnet synchronous machines (IPMSMs), consisting of a progressive modification of the rotor lamination geometry. This procedure can be generalized for the main topologies of PMSM, independently of the number of stator slots or the location of the permanent magnets. For this purpose, a basic IPMSM structure is analyzed by means of the FEM (finite element method) approach, and then, several other IPMSM geometries, obtained by adequately modifying the rotor lamination geometry of the basic IPMSM model without changing its stator configuration, are proposed and discussed. The trends of the cogging torqu…

An Innovative Structural Dynamic Identification Procedure Combining Time Domain OMA Technique and GA

2022

In this paper an innovative and simple Operational Modal Analysis (OMA) method for structural dynamic identification is proposed. It combines the recently introduced Time Domain–Analytical Signal Method (TD–ASM) with the Genetic Algorithm (GA). Specifically, TD–ASM is firstly employed to estimate a subspace of candidate modal parameters, and then the GA is used to identify the structural parameters minimizing the fitness value returned by an appropriately introduced objective function. Notably, this method can be used to estimate structural parameters even for high damping ratios, and it also allows one to identify the Power Spectral Density (PSD) of the structural excitat…

Conducting a task while reconstructing its meaning

2022

This article investigates the way an institutional task of a meeting is oriented to by different meeting participants and developed in and through local interaction. Our data come from a city organization, where a large organizational change is planned and prepared through a series of face-to-face encounters and accompanying written texts. Using the notion of recontextualization and by connecting it to the conversation analytical method and to the notion of intersubjectivity, the study examines how the institutional task that is verbalized in written form prior to the meeting is conceptualized by meeting participants in their turns of talk. By doing so, the study will particularly shed ligh…

Leisure Adjustments of Older Finnish Adults during the Early Stages of the COVID-19 Pandemic

2022

The current approach to “aging well” emphasizes the importance of active leisure participation in late adulthood. This relates to the view that leisure activities enable older adults to stay physically, mentally, and socially active, and they thereby contribute to wellbeing. In spring 2020, leisure activity engagement was significantly hampered by the COVID-19 pandemic. This study explores leisure-related experiences and adaptation strategies among Finnish older adults during the period of strict physical distancing. The data comprises letters (N = 77) written by Finnish people (aged 70–93) that were analyzed using content analysis. The study suggests that the reorganization of leisure was …

Mocking fakeness

2022

Abstract Phonetic resources, like dialects and accents, are used in ethnic humour to build up a recognisable character that pokes fun at the stereotypes associated with a particular identity, sometimes with critical and political undertones. In this article, we examine the manipulation of one such resource, aspiration, used in performing and mocking one such clichéd character, called the fake Sámi. This character has a contested history in Finnish tourism and marketing practices, and is embedded in a long-standing debate about who can use emblems of Sámi identity for economic purposes. Manipulation as an ideological tool in the political genre of Parliamentary discourses

2022

AbstractThe present study analyzes the discursive strategies of manipulation in the political genre of a discourse in Parliament with an aim to convince the audience that the Prime Minister and his party are innocent of receiving illegal cash donations from a slush fund run in the party. For that purpose, we have usedVan Dijk’s (2006)scheme of strategies of manipulation at several levels of discourse (content, lexis, topics, syntax, rhetoric, and order of discourse). Findings of the study show that the Prime Minister’s speech presents characteristics of ideological discourse, since it follows a general strategy of positive in-group and negative out-group presentation, which has an overall l…

Current and emerging drugs for the treatment of atherosclerosis: the evidence to date

2022

Introduction: Atherosclerosis can be considered a chronic inflammatory process that stands out as a dominant cause of cardiovascular disease (CVD). Since blood lipids are the leading risk factor for atherosclerosis development, lowering low-density lipoprotein cholesterol (LDL-C) and other apolipoprotein B-containing lipoproteins reduces the risk of future cardiovascular events. However, there has been significant progress in developing lipid-lowering drugs for aggressive management of dyslipidemia, the rates of CVD events remain unacceptably high, so there is great need to identify novel therapeutic pathways targeting the atherosclerosis process.Areas covered: We discussed the current guid…

Mitogenomics of the Olive Seed Weevil, Anchonocranus oleae Marshall and Implications for Its Phylogenetic Position in Curculionidae

2022

Anchonocranus oleae Marshall (Coleoptera: Curculionidae) is a seed-feeding weevil native to southern Africa; its larvae are known to develop in the fruits of the African Wild Olive and, more rarely, cultivated olives. The species has been mainly found in the Western Cape province of South Africa, but it has remained in relative obscurity because it does not seem to represent a current threat to commercial olive production. As part of an ongoing effort to produce baseline genetic data for olive-associated entomofauna in South Africa, we generated reference DNA barcodes for A. oleae collected from wild and cultivated olives and sequenced its mitogenome for assessment of the phylogenetic posit…

Metabolic plasticity of mixotrophic algae is key for their persistence in browning environments

2022

Light availability is the main regulator of primary production, shaping photosynthetic communities and their production of ecologically important biomolecules. In freshwater ecosystems, increasing dissolved organic carbon concentrations, commonly known as browning, leads to lower light availability and the proliferation of mixotrophic phytoplankton. Here, a mixotrophic algal species (Cryptomonas sp.) was grown under five increasing dissolved organic carbon concentrations to uncover the plastic responses behind the success of mixotrophs in browning environments and their effect in the availability of nutritionally important biomolecules. In addition to the browning treatments, phototrophic, …

Karl Marx: Umanismo e Materialismo

2022

In the context of the contemporary age, Karl Marx represents a classical author. His theory of historical materialism is the basis of his general vision of reality with the principles of “self-consciousness” and “class consciousness”. To the optimism of idealism, centered on pure theory, Marx opposes a new cultural model in relation to praxis and political economy. His production must therefore be read in all its phases, from youth to maturity, with the aim of interpreting historical materialism as a peculiar form of humanism and naturalism. Cytotoxic activity of several ent-kaurane derivatives of atractyligenin. Synthesis of unreported diterpenic skeleton by chemical rearrangement

2022

Atractyloside, carboxyatractyloside, their aglycon atractyligenin, and several synthetic derivatives were tested and found to be active against a panel of human tumor cell lines. Atractyligenin was subjected to oxidation, bromination, and elimination reactions, obtaining several compounds. A singular skeleton was synthesized by chemical rearrangement starting from 3 beta-bromo-2,15-diketoatractyligenin methyl ester. The synthesized compounds resulted active against all cell lines tested. In particular, 15-ketoatractyligenin methyl ester and 3 beta bromo-2,15-diketoatractyligenin methyl ester resulted the most active with IC50 values of 0.427 and 0.723 mu M against A375 melanoma cell line.
