The effects of phenylephrine on the shape of the contraction curve and on the cyclic adenosine 3',5'-monophosphate (c-AMP) content were studied in electrically driven (frequency 0.2 Hz) cat papillary muscles. All experiments were done in the presence of 1 micron propranolol in order to minimize interference from beta-adrenoceptors. 1. Phenylephrine increased the force of contraction in a concentration-dependent manner. Maximal effects (about 200% of control) occurred at 30 micron phenylephrine. 2. A negative inotropic effect of acetylcholine in the presence of several phosphodiesterase inhibitors.

The phosphodiesterase inhibitors papaverine, theophylline and 3-isobutyl-1-methylxanthine (IBMX) reveal a negative inotropic effect of acetylcholine in cat ventricular heart muscle. This effect in unrelated to beta-adrenoceptor stimulation and possibly mediated by the accumulation of cyclic GMP.

Cyclic AMP and cyclic GMP may play opposing roles in influencing force of contraction in mammalian myocardium.

CYCLIC AMP and cyclic GMP have been suggested to play opposing regulatory roles in several biological systems1. Supporting evidence for the yin yang hypothesis of opposing biological regulation has been obtained in sympathetic ganglia2,3 and pyramidal neurones in the rat cerebral cortex4. Electrical and mechanical activity of mammalian heart muscle fibres treated with papaverine

The action of papaverine on electrical and mechanical activity was investigated in ventricular and atrial heart muscle fibres from guinea-pigs and cats. 1. Papaverine (10−5 M–5×10−5M) had positive, negative or no inotropic effects in ventricular preparations; positive inotropic effects were not observed after pretreatment of the animals with reserpine. In atrial preparations, papaverine (2×10−5M) had a positive inotropic effect that was independent of endogenously stored catecholamines. 2. Effect of DB-c-AMP on mechanical characteristics of ventricular and atrial preparations of several mammalian species

Conflicting results exist about the influence of cyclic N6-2′-O-dibutyryl-AMP (DB-c-AMP) on myocardial contractile force. The present study was designed to examine whether the positive inotropic action of DB-c-AMP is restricted to certain model preparations or whether it can be assumed to represent a more general effect of the drug. Therefore, the effects of DB-c-AMP on myocardial force and on various parameters of the isometric contraction curve were examined in isolated electrically driven (0.5–2Hz) ventricular and atrial preparations of several mammalian species (cat, rabbit, calf, sheep, rat and guinea-pig). Experimental Hypertensive Intracerebral Mass Hemorrhage in Cats

About two–thirds of all patients with spontaneous intracerebral hemorrhage suffer from systemic hypertension. Additionally systemic hypertension is the main factor indicating a poor prognosis of intracerebral hemorrhage. This has been shown in our statistical evaluation of 117 cases of conservatively treated spontaneous intracerebral hemorrhage. More than 48% of the hypertensive patients died, but only 30% of the normotensive patients (Fig. 1). In an experimental study we investigated the influence of systemic hypertension and normotension on epidural pressure and on the formation of brain edema during the first 12 h after artificial intracerebral hemorrhage.

The role of the substantia nigra in the control of amygdaloid paroxysmal activity.

AbstractBoth in acute and chronic cats, focal paroxysmal activity evoked in the ventro-basal complex of the amygdala has been inhibited by substantia nigra conditioning stimulation, to a greater extent, than by caudate nucleus activation. Injection of kainic acid into substantia nigra resulted in the disappearance of the caudate inhibition. It is suggested that the final control, exerted by the striatum on the amygdaloid seizures, occurs by means of the substantia nigra.

The control of amygdaloid seizures by the globus pallidus.

Both in acute and chronic cats entopeduncular stimulation inhibits, to a greater extent than caudate activation, focal paroxysmal activity in the ventro-basal complex of the amygdala. Lesion of entopeduncular neurons, by means of kainic acid injection, induces a decrease of the caudate inhibitory effect. It is suggested that neostriatal control of the amygdaloid seizures occurs partly through the globus pallidus.

Phylogeny and prevalence of kobuviruses in dogs and cats in the UK

AbstractThe kobuviruses represent an emerging genus in the Picornaviridae. Here we have used next generation sequencing and conventional approaches to identify the first canine kobuvirus (CaKoV) from outside the USA. Phylogenetic analysis suggests that a single lineage genotype of CaKoV now exists in Europe and the USA with 94% nucleotide similarity in the coding region. CaKoV was only identified in a single case from a case–control study of canine diarrhoea, suggesting this virus was not a frequent cause of disease in this population.
