Role of Levo-tetrahydropalmatine and its metabolites for management of chronic pain and opioid use disorders.

2021

Abstract Background Opioids have been prescribed to reduce suffering from pain and to enhance quality of life. Due to the addictive potential and the lack of other effective alternatives to treat severe acute and chronic pains, opioids remain a serious public health issue. While, opioids directly influence the drug-seeking behavior, tolerance and withdrawal processes, through neuroadaptation, the brain's endogenous opioid system also adapts in the presence of chronic pain and could contribute to the difficulty of treatment. Despite the seemingly obvious interaction between the presence of pain and opioid-abuse, little is known about the underlying mechanisms in the brain. Purpose To review …

Function of AtPGAP1 in GPI anchor lipid remodeling and transport to the cell surface of GPI-anchored proteins

2021

ABSTRACTGPI-anchored proteins (GPI-APs) play an important role in a variety of plant biological processes including growth, stress response, morphogenesis, signalling and cell wall biosynthesis. The GPI-anchor contains a lipid-linked glycan backbone that is synthesized in the endoplasmic reticulum (ER) where it is subsequently transferred to the C-terminus of proteins containing a GPI signal peptide by a GPI transamidase. Once the GPI anchor is attached to the protein, the glycan and lipid moieties are remodelled. In mammals and yeast, this remodelling is required for GPI-APs to be included in Coat Protein II (COPII) coated vesicles for their ER export and subsequent transport to the cell s…

A Gravitational-wave Measurement of the Hubble Constant Following the Second Observing Run of Advanced LIGO and Virgo

2021

This paper presents the gravitational-wave measurement of the Hubble constant (H 0) using the detections from the first and second observing runs of the Advanced LIGO and Virgo detector network. The presence of the transient electromagnetic counterpart of the binary neutron star GW170817 led to the first standard-siren measurement of H 0. Here we additionally use binary black hole detections in conjunction with galaxy catalogs and report a joint measurement. Our updated measurement is H 0 = km s-1 Mpc-1 (68.3% of the highest density posterior interval with a flat-in-log prior) which is an improvement by a factor of 1.04 (about 4%) over the GW170817-only value of km s-1 Mpc-1. A significant …

Phylogeny, biogeography and systematics of Dysphanieae (Amaranthaceae)

2021

After a rather turbulent taxonomic history, Dysphanieae (Chenopodioideae, Amaranthaceae) were established to contain five genera, four of which are monospecific (Cycloloma, Neomonolepis, Suckleya, Teloxys) and geographically restricted, and the fifth genus, Dysphania, having a nearly worldwide distribution and comprising ca. 50 species. This study investigates the phylogeny, biogeography and taxonomy of Dysphanieae. We studied specimens from 32 herbaria to infer morphological differences and distribution areas of the species and sampled 121 accessions representing 39 accepted species of the tribe for molecular phylogenetic analyses. The molecular phylogeny tested generic relationships of th…

A multi-phase multi-objective dynamic genome-scale model shows different redox balancing among yeast species in fermentation

2021

ABSTRACTYeasts constitute over 1500 species with great potential for biotechnology. Still, the yeastSaccharomyces cerevisiaedominates industrial applications and many alternative physiological capabilities of lesser-known yeasts are not being fully exploited. While comparative genomics receives substantial attention, little is known about yeasts’ metabolic specificity in batch cultures. Here we propose a multi-phase multi-objective dynamic genome-scale model of yeast batch cultures that describes the uptake of carbon and nitrogen sources and the production of primary and secondary metabolites. The model integrates a specific metabolic reconstruction, based on the consensus Yeast8, and a kin…

A photoswitchable helical peptide with light-controllable interface / transmembrane topology in lipidic membranes

2021

AbstractAccording to the three-step model, the spontaneous insertion and folding of helical transmembrane (TM) polypeptides into lipid bilayers is driven by three sequential equilibria: solution-to-membrane interface (MI) partition, unstructured-to-helical folding, and MI-to-TM helix insertion. However, understanding these three steps with molecular detail has been challenged by the lack of suitable experimental approaches to rapidly and reversibly perturb membrane-bound hydrophobic polypeptides out of equilibrium. Here, we report on a 24-residues-long hydrophobic α-helical polypeptide, covalently coupled to an azobenzene photoswitch (KCALP-azo), which displays a light-controllable TM/MI eq…

Analysis of Brain Functions in Men with Prostate Cancer under Androgen Deprivation Therapy: A One-Year Longitudinal Study.

2021

The relationship between cognitive decline and androgen deprivation therapy (ADT) under luteinizing hormone-releasing hormone (LHRH) analogues is unclear, and there is a scarcity of longitudinal studies considering the interaction between cognition, depressive symptoms and sleep quality in men with prostate cancer (PCa) treated with ADT. This study aimed to determine if there were differences in the scores obtained in cognitive assessment, depressive symptoms, and sleep quality after one year of ADT and determine the interrelations between sleep, mood, and cognitive status. A prospective longitudinal observational study was designed, in which a cohort of men (mean age was 70.8 years) newly …

Abstract. The formation of ice in clouds is an important processes in mixed-phase and ice-phase clouds. Yet, the representation of ice formation in numerical models is highly uncertain. In the last decade, several new parameterizations for heterogeneous freezing have been proposed. However, it is currently unclear what the effect of choosing one parameterization over another is in the context of numerical weather prediction. We conducted high-resolution simulations (Δx=250 m) of moderately deep convective clouds (cloud top ∼-18 ∘C) over the southwestern United Kingdom using several formulations of ice formation and compared the resulting changes in cloud field properties to the spread of an…

Symmetry meets AI

2021

We explore whether Neural Networks (NNs) can {\it discover} the presence of symmetries as they learn to perform a task. For this, we train hundreds of NNs on a {\it decoy task} based on well-controlled Physics templates, where no information on symmetry is provided. We use the output from the last hidden layer of all these NNs, projected to fewer dimensions, as the input for a symmetry classification task, and show that information on symmetry had indeed been identified by the original NN without guidance. As an interdisciplinary application of this procedure, we identify the presence and level of symmetry in artistic paintings from different styles such as those of Picasso, Pollock and Van…

Stationary subspace analysis based on second-order statistics

2021

In stationary subspace analysis (SSA) one assumes that the observable p-variate time series is a linear mixture of a k-variate nonstationary time series and a (p-k)-variate stationary time series. The aim is then to estimate the unmixing matrix which transforms the observed multivariate time series onto stationary and nonstationary components. In the classical approach multivariate data are projected onto stationary and nonstationary subspaces by minimizing a Kullback-Leibler divergence between Gaussian distributions, and the method only detects nonstationarities in the first two moments. In this paper we consider SSA in a more general multivariate time series setting and propose SSA method…

Spin accumulation from nonequilibrium first principles methods

2021

For the technologically relevant spin Hall effect, most theoretical approaches rely on the evaluation of the spin-conductivity tensor. In contrast, for most experimental configurations the generation of spin accumulation at interfaces and surfaces is the relevant quantity. Here, we directly calculate the accumulation of spins due to the spin Hall effect at the surface of a thin metallic layer, making quantitative predictions for different materials. Two distinct limits are considered, both relying on a fully relativistic Korringa-Kohn-Rostoker density functional theory method. In the semiclassical approach, we use the Boltzmann transport formalism and compare it directly with a fully quantu…

Bayesian Analysis of Population Health Data

2021

The analysis of population-wide datasets can provide insight on the health status of large populations so that public health officials can make data-driven decisions. The analysis of such datasets often requires highly parameterized models with different types of fixed and random effects to account for risk factors, spatial and temporal variations, multilevel effects and other sources on uncertainty. To illustrate the potential of Bayesian hierarchical models, a dataset of about 500,000 inhabitants released by the Polish National Health Fund containing information about ischemic stroke incidence for a 2-year period is analyzed using different types of models. The June 2018 CAP proposals are only marginally consistent with the ambitions of the Green Deal. This is also the case of the regulation revisions being adopted by either the Council or the European Parliament in October 2020. Making EU agriculture consistent with the Green Deal but would require a whole food chain policy that encompasses more stringent instruments on the supply side and extensive changes in eating patterns.

Increasing attention has been paid to the importance of balanced rural–urban interaction to sustainable regional development. Yet, our knowledge on the elements of network governance for such interaction is scarce. The aim of this paper is to study what kind of network governance arrangements currently exist, how they can be improved, and whether evolutionary governance paths can be identified. We analyse five existing and evolving cases of functioning rural–urban interaction in European Union (EU) member states, using a network governance framework as an analytical lens. We supplement the governance analysis with examining what kind of spatial understanding or combination of di…

Analysis of composition, morphology, and biosynthesis of cuticular wax in wild type bilberry (Vaccinium myrtillus L.) and its glossy mutant

2021

Abstract In this study, cuticular wax load, its chemical composition, and biosynthesis, was studied during development of wild type (WT) bilberry fruit and its natural glossy type (GT) mutant. GT fruit cuticular wax load was comparable with WT fruits. In both, the proportion of triterpenoids decreased during fruit development concomitant with increasing proportions of total aliphatic compounds. In GT fruit, a higher proportion of triterpenoids in cuticular wax was accompanied by a lower proportion of fatty acids and ketones compared to WT fruit as well as lower density of crystalloid structures on berry surfaces. Our results suggest that the glossy phenotype could be caused by the absence o…
