Hypusinated eIF5A is required for the translation of collagen.

2021

ABSTRACT Translation of mRNAs that encode peptide sequences with consecutive prolines (polyproline) requires the conserved and essential elongation factor eIF5A to facilitate the formation of peptide bonds. It has been shown that, upon eIF5A depletion, yeast ribosomes stall in polyproline motifs, but also in tripeptide sequences that combine proline with glycine and charged amino acids. Mammalian collagens are enriched in putative eIF5A-dependent Pro-Gly-containing tripeptides. Here, we show that depletion of active eIF5A in mouse fibroblasts reduced collagen type I α1 chain (Col1a1) content, which concentrated around the nuclei. Moreover, it provoked the upregulation of endoplasmic reticul…

Facilitators of and Barriers to Sustainable Development in Small and Medium-Sized Enterprises: A Descriptive Exploratory Study in Romania

2021

In the context of growing concerns regarding the deterioration of the environment and the increase in social inequalities, the concept of sustainability emerged as a response of companies, in order to contribute to community goodwill. The drivers and obstacles for the businesses engaging in sustainable policies have been explored at large by scientific literature. However, research gaps were observed, namely regarding SMEs, that tend to have a less formal and more fragmented approach to sustainability. The goal of our study is to determine the main barriers and facilitators for sustainability that Romanian SMEs face, and the connections between them and with the firms’ characteristics. Our …

Direct measurement of the mass difference of $^{72}$As-$^{72}$Ge rules out $^{72}$As as a promising $\beta$-decay candidate to determine the neutrino…

2021

We report the first direct determination of the ground-state to ground-state electron-capture $Q$-value for the $^{72}$As to $^{72}$Ge decay by measuring their atomic mass difference utilizing the double Penning trap mass spectrometer, JYFLTRAP. The $Q$-value was measured to be 4343.596(75)~keV, which is more than a 50-fold improvement in precision compared to the value in the most recent Atomic Mass Evaluation 2020. Furthermore, the new $Q$-value was found to be 12.4(40)~keV (3.1 $\sigma$) lower. With the significant reduction of the uncertainty of the ground-state to ground-state $Q$-value value combined with the level scheme of $^{72}$Ge from $\gamma$-ray spectroscopy, we confirm that th…

Luonnon monimuotoisuus ja vihreä elvytys

2021

Suomi on toistaiseksi selvinnyt koronaviruksen (COVID-19) aiheuttamasta kriisistä taloudellisesti verrokkimaita paremmin, mutta työllisyystilanne on silti heikentynyt ympäri maata ja talouden ennustetaan supistuvan noin 4,7 prosenttia vuonna 20201. Negatiivisten talousvaikutusten minimoimiseksi hallitus on suuntaamassa EU:n elpymisvälineestä varoja käytettäväksi toimiin, jotka samanaikaisesti auttavat ratkaisemaan aikamme kahta merkittävää kriisiä: ilmastonmuutosta ja luontokatoa. Kyse on aidosti vakavista kriiseistä. Esimerkiksi Maailman talousfoorumi on listannut luonnon ekosysteemien romahduksen ja ilmastonmuutoksen torjunnan epäonnistumisen sekä vaikutuksiltaan että todennäköisyydeltään…

Electrochemically triggered iodide-vacancy BiOI film for selective extraction of iodide ion from aqueous solutions

2021

Abstract The effective extraction and regeneration of radioactive iodide remains an urgent concern for safe nuclear energy utilization. Herein, we developed a novel electrochemically triggered iodide-vacancy BiOI film, which exhibited excellent I− ion extraction capacity of 328.3 mg·g−1. Especially, due to the ion vacancy trap effect, the film showed high selectivity towards I− ions in the existence of a large number of competitive anions. Additionally, the electrochemically switched ion extraction (ESIE) process with this iodide-vacancy BiOI film possessed fast extraction kinetics and high stability. More importantly, the trapped I− ions were easily desorbed from the film without the secon…

Distribution patterns of epiphytic reed-associated macroinvertebrate communities across European shallow lakes

2021

So far, research on plant-associated macroinvertebrates, even if conducted on a large number of water bodies, has mostly focused on a relatively small area, permitting limited conclusions to be drawn regarding potentially broader geographic effects, including climate. Some recent studies have shown that the composition of epiphytic communities may differ considerably among climatic zones. To assess this phenomenon, we studied macroinvertebrates associated with the common reed Phragmites australis (Cav.) Trin. ex Steud in 46 shallow lakes using a common protocol. The lakes, located in nine countries, covered almost the entire European latitudinal range (from <48°N to 61°N) and captured much …

The reliability of spanish and german electricity forward prices. Databases and price discovery process

2021

Given the existence of different databases from different sources that offer information on forward electricity prices, the need to compare them arises to guarantee that research results and trading decisions based on them are not sensitive to the database used. We worked with forward electricity prices traded over the counter, closest month to maturity, covering the period from 2010 to 2016 for the Spanish over the counter (OTC) market, and from 2008 to 2016 for the German OTC market. The goal of this paper was to test whether there were significant discrepancies between the price series provided by two of the main agencies of financial information (Thomson Reuters and Bloomberg), as well …

Deep learning for core-collapse supernova detection

2021

The detection of gravitational waves from core-collapse supernova (CCSN) explosions is a challenging task, yet to be achieved, in which it is key the connection between multiple messengers, including neutrinos and electromagnetic signals. In this work, we present a method for detecting these kind of signals based on machine learning techniques. We tested its robustness by injecting signals in the real noise data taken by the Advanced LIGO-Virgo network during the second observing run, O2. We trained a newly developed Mini-Inception Resnet neural network using time-frequency images corresponding to injections of simulated phenomenological signals, which mimic the waveforms obtained in 3D num…

All-sky search in early O3 LIGO data for continuous gravitational-wave signals from unknown neutron stars in binary systems

2021

Rapidly spinning neutron stars are promising sources of continuous gravitational waves. Detecting such a signal would allow probing of the physical properties of matter under extreme conditions. A significant fraction of the known pulsar population belongs to binary systems. Searching for unknown neutron stars in binary systems requires specialized algorithms to address unknown orbital frequency modulations. We present a search for continuous gravitational waves emitted by neutron stars in binary systems in early data from the third observing run of the Advanced LIGO and Advanced Virgo detectors using the semicoherent, GPU-accelerated, binaryskyhough pipeline. The search analyzes the most s…

Translational adaptation to heat stress is mediated by RNA 5‐methylcytosine in Caenorhabditis elegans

2021

Abstract Methylation of carbon‐5 of cytosines (m5C) is a post‐transcriptional nucleotide modification of RNA found in all kingdoms of life. While individual m5C‐methyltransferases have been studied, the impact of the global cytosine‐5 methylome on development, homeostasis and stress remains unknown. Here, using Caenorhabditis elegans, we generated the first organism devoid of m5C in RNA, demonstrating that this modification is non‐essential. Using this genetic tool, we determine the localisation and enzymatic specificity of m5C sites in the RNome in vivo. We find that NSUN‐4 acts as a dual rRNA and tRNA methyltransferase in C. elegans mitochondria. In agreement with leucine and proline bein…

Widening of Hydrous Shear Zones During Incipient Eclogitization of Metastable Dry and Rigid Lower Crust—Holsnøy, Western Norway

2021

The partially eclogitized crustal rocks on Holsnøy in the Bergen Arcs, Norway, indicate that eclogitization is caused by the interplay of brittle and ductile deformation promoted by fluid infiltration and fluid‐rock interaction. Eclogitization generated an interconnected network of millimeter‐to‐kilometer‐wide hydrous eclogite‐facies shear zones, which presumably caused transient weakening of the mechanically strong lower crust. To decipher the development of those networks, we combine detailed lithological and structural mapping of two key outcrops with numerical modeling. Both outcrops are largely composed of preserved granulite with minor eclogite‐facies shear zones, thus representing th…

Search for inelastic scattering of WIMP dark matter in XENON1T

2021

We report the results of a search for the inelastic scattering of weakly interacting massive particles (WIMPs) in the XENON1T dark matter experiment. Scattering off $^{129}$Xe is the most sensitive probe of inelastic WIMP interactions, with a signature of a 39.6 keV de-excitation photon detected simultaneously with the nuclear recoil. Using an exposure of 0.89 tonne-years, we find no evidence of inelastic WIMP scattering with a significance of more than 2$\sigma$. A profile-likelihood ratio analysis is used to set upper limits on the cross-section of WIMP-nucleus interactions. We exclude new parameter space for WIMPs heavier than 100 GeV/c${}^2$, with the strongest upper limit of $3.3 \time…

Proteomic analysis of tyrosine phosphorylation induced by exogenous expression of oncogenic kinase fusions identified in lung adenocarcinoma.

2021

Kinase fusions are considered oncogenic drivers in numerous types of cancer. In lung adenocarcinoma 5-10% of patients harbor kinase fusions. The most frequently detected kinase fusion involves the Anaplastic Lymphoma Kinase (ALK) and Echinoderm Microtubule-associated protein-Like 4 (EML4). In addition, oncogenic kinase fusions involving the tyrosine kinases RET and ROS1 are found in smaller subsets of patients. In this study, we employed quantitative mass spectrometry-based phosphoproteomics to define the cellular tyrosine phosphorylation patterns induced by different oncogenic kinase fusions identified in patients with lung adenocarcinoma. We show that exogenous expression of the kinase fu…

Global effects of ade8 deletion on budding yeast metabolism

2021

AbstractPurine auxotrophy is a typical marker for many laboratory yeast strains. Supplementation of additional purine source (like adenine) is necessary to cultivate these strains. If not supplied in adequate amounts, purine starvation sets in. We tested purine starvation effects in budding yeast Saccharomyces cerevisiae ade8 knockout. We explored effects brought by purine starvation in cellular, central carbon metabolism and in the global transcriptome level. We observed that cells cultivated in purine depleted media became significantly more tolerant to severe thermal, oxidative and desiccation stresses when compared to the cells cultivated in media with all necessary supplements. When sta…

From pirates to subscribers: 20 years of music consumption research

2021

The last two decades have witnessed an increasing scholarly interest on music consumption. This interest can be explained, at least partly, to the relevance of music as a form of cultural consumption and the profound changes the sector has undergone. This paper performs a bibliometric analysis of the literature on music consumption research. In doing so, a database comprising 455 academic documents on the fields of business, economics, and management, was reviewed following a systematic procedure. Through it we identify the intellectual roots and the methodological evolution of the field. Furthermore, text mining was applied to analyse the themes included in the research agenda and their ev…

A prehistoric jade axe from Galicia (Northwestern Iberia): Researching its origin

2021

International audience; The Vilapedre axe (Lugo, Northwest Iberia) has been traditionally considered by archaeologists as evidence of prehistoric long-distance contacts along the Atlantic Coast of France and Spain. This artefact - as other “Tumiac type” axes (long polished blades, generally butt-perforated) - would have been produced in Brittany during the Neolithic (5th millennium BCE) using jadeitite as raw material, a green-coloured rock for which there are sources in the western Italian Alps. In this paper, we have traced the possible archaeological origin of this artefact back by examining the personal files of one of its first owners, Santiago de la Iglesia. Furthermore, we have condu…

Burkitt lymphoma with a granulomatous reaction: an M1/Th1‐polarised microenvironment is associated with controlled growth and spontaneous regression

2021

Aims Burkitt lymphoma (BL) is an aggressive B-cell lymphoma that, in some instances, may show a granulomatous reaction associated with a favourable prognosis and occasional spontaneous regression. In the present study, we aimed to define the tumour microenvironment (TME) in four such cases, two of which regressed spontaneously. Methods and results All cases showed aggregates of tumour cells with the typical morphology, molecular cytogenetics and immunophenotype of BL surrounded by a florid epithelioid granulomatous reaction. All four cases were Epstein-Barr virus (EBV)-positive with type I latency. Investigation of the TME showed similar features in all four cases. The analysis revealed a p…

Minimum Leading Edge Protection Application Length to Combat Rain-Induced Erosion of Wind Turbine Blades

2021

Leading edge erosion (LEE) repairs of wind turbine blades (WTBs) involve infield application of leading edge protection (LEP) solutions. The industry is currently aiming to use factory based LEP coatings that can applied to the WTBs before they are shipped out for installation. However, one of the main challenges related to these solutions is the choice of a minimum LEP application length to be applied in the spanwise direction of the WTBs. Generally, coating suppliers apply 10–20 m of LEP onto the blades starting from the tip of the blade using the “rule of thumb”, and no studies in the literature exist that stipulate how these LEP lengths can be calculated. In this study, we extend the sc…

Europe from Below : Notions of Europe and the European among Participants in EU Cultural Initiatives

2021

In this book, Tuuli Lähdesmäki, Katja Mäkinen, Viktorija L. A. Čeginskas, and Sigrid Kaasik-Krogerus scrutinize how people who participate in cultural initiatives funded and governed by the European Union understand the idea of Europe. The book focuses on three cultural initiatives: the European Capital of Culture, the European Heritage Label, and a European Citizen Campus project funded through the Creative Europe programme. These initiatives are examined through field studies conducted in 12 countries between 2010 and 2018. The authors describe their approach as ‘ethnography of Europeanization’ and conceptualize the attempts at Europeanization in the European Union’s cultural policy as po…

Insights of Tris(2-pyridylmethyl)amine as anti-tumor agent for osteosarcoma: experimental and in silico studies

2021

Abstract Osteosarcoma (OS) is a malignant bone tumor and its occurrence is associated with high levels of microRNAs (miRNAs) and critical protein sinvolved on intracellular signaling pathways. Cisplatin and Doxorubicin are employed on chemotherapy, but their use cause side effects and contributes to multidrug resistance. Since several tripodal amines have been studied as anticancer agents, tris(2-pyridylmethyl)amine (TPA) was investigated against osteosarcoma cells. Here we show that TPA exhibits activity against MG-63 and Saos-2 cells. Cyclic voltammetry results indicate an interaction between TPA and dsDNA, denoted by blocking of the proton-assisted reduction of the 2-pyridylmethyl moiety…

In this study, we evaluated four ecotypes from Italy (Sardinia and Abruzzo), Spain (Castilla-La Mancha), and Greece (Kozani) in order to detect the existence of variability and promote the biodiversity of this crop. Thirty-one traits related to saffron flowering, flower morphology, production of spice and daughter corms, vegetative development (leaf and corm traits), and spice quality, were evaluated. In addition, a genetic analysis through three PCR-based approaches, SSRs, RAPD, and SRAP was assessed. Results highlighted a…

Loss‐of‐function variants in ARHGEF9 are associated with an X‐linked intellectual disability dominant disorder

2021

ARHGEF9 defects lead to an X-linked intellectual disability disorder related to inhibitory synaptic dysfunction. This condition is more frequent in males, with a few affected females reported. Up to now, sequence variants and gross deletions have been identified in males, while only chromosomal aberrations have been reported in affected females who showed a skewed pattern of X-chromosome inactivation (XCI), suggesting an X-linked recessive (XLR) disorder. We report three novel loss-of-function (LoF) variants in ARHGEF9: A de novo synonymous variant affecting splicing (NM_015185.2: c.1056G>A, p.(Lys352=)) in one female; a nonsense variant in another female (c.865C>T, p.(Arg289*)), that is, a…

On decoupling in Banach spaces

2021

AbstractWe consider decoupling inequalities for random variables taking values in a Banach space X. We restrict the class of distributions that appear as conditional distributions while decoupling and show that each adapted process can be approximated by a Haar-type expansion in which only the pre-specified conditional distributions appear. Moreover, we show that in our framework a progressive enlargement of the underlying filtration does not affect the decoupling properties (in particular, it does not affect the constants involved). As a special case, we deal with one-sided moment inequalities for decoupled dyadic (i.e., Paley–Walsh) martingales and show that Burkholder–Davis–Gundy-type in…

Direct time delay computation applied to the O + O2 exchange reaction at low energy: Lifetime spectrum of O3* species

2021

We report full quantum dynamical calculations for lifetimes of scattering resonances, among which are true metastable states, of the intermediate heavy ozone complex 50O3* of the 18O + 16O16O reaction, for any value of the total angular momentum quantum number J. We show that computations for nonzero values of J are mandatory in order to properly analyze resonances and time delays, with a view to establish a somewhat comprehensive eigenlife spectrum of the complex O3*. Calculations have been performed in a given low to moderate energy range, including the interval between zero-point energies (ZPEs) of reagents and product species. Quasi-bound states tend to be more numerous, and eigenlifeti…
