Background: Systemic lupus erythematosus (SLE) is a chronic inflammatory autoimmune disease and patients are under an increased risk for cardiovascular (CV) events and mortality. The increased CV risk for patients with SLE seems to be caused by a premature and accelerated atherosclerosis, attributable to lupus-specific risk factors (i.e., increased systemic inflammation, altered immune status), apart from traditional CV risk factors. To date, there is no established experimental model to explore the pathogenesis of this increased CV risk in SLE patients. Methods: Here we investigated whether MRL-Faslpr mice, which develop an SLE-like phenotype, may serve as a model to study lupus-mediated v…

Applicability of a single‐use bioreactor compared to a glass bioreactor for the fermentation of filamentous fungi and evaluation of the reproducibili…
2021
Abstract The implementation of single‐use technologies offers several major advantages, e.g. prevention of cross‐contamination, especially when spore‐forming microorganisms are present. This study investigated the application of a single‐use bioreactor in batch fermentation of filamentous fungus Penicillium sp. (IBWF 040‐09) from the Institute of Biotechnology and Drug Research (IBWF), which is capable of intracellular production of a protease inhibitor against parasitic proteases as a secondary metabolite. Several modifications to the SU bioreactor were suggested in this study to allow the fermentation in which the fungus forms pellets. Simultaneously, fermentations in conventional glass b…
Fully coupled functional equations for the quark sector of QCD
2021
We present a comprehensive study of the quark sector of $2+1$ flavour QCD, based on a self-consistent treatment of the coupled system of Schwinger-Dyson equations for the quark propagator and the full quark-gluon vertex. The individual form factors of the quark-gluon vertex are expressed in a special tensor basis obtained from a set of gauge-invariant operators. The sole external ingredient used as input to our equations is the Landau gauge gluon propagator with $2+1$ dynamical quark flavours, obtained from studies with Schwinger-Dyson equations, the functional renormalisation group approach, and large volume lattice simulations. The appropriate renormalisation procedure required in order t…
SARS-CoV-2-Specific Memory T Lymphocytes From COVID-19 Convalescent Donors: Identification, Biobanking, and Large-Scale Production for Adoptive Cell …
2021
Syndrome coronavirus 2 (SARS-CoV-2) pandemic is causing a second outbreak significantly delaying the hope for the virus’ complete eradication. In the absence of effective vaccines, we need effective treatments with low adverse effects that can treat hospitalized patients with COVID-19 disease. In this study, we determined the existence of SARS-CoV-2-specific T cells within CD45RA– memory T cells in the blood of convalescent donors. Memory T cells can respond quickly to infection and provide long-term immune protection to reduce the severity of COVID-19 symptoms. Also, CD45RA– memory T cells confer protection from other pathogens encountered by the donors throughout their life. It is of vita…
Anomaly-free leptophilic axionlike particle and its flavor violating tests
2021
Motivated by the recent Xenon1T result, we study a leptophilic flavor-dependent anomaly-free axionlike particle (ALP) and its effects on charged-lepton flavor violation. We present two representative models. The first one considers that the ALP origins from the flavon that generates the charged-lepton masses. The second model assumes a larger flavor symmetry such that more general mixings in the charged-lepton are possible, while maintaining flavor-dependent ALP couplings. We find that a keV ALP explaining the Xenon1T result is still viable for lepton flavor violation and stellar cooling astrophysical limits. On the other hand, if the Xenon1T result is confirmed, future charged-lepton flavo…
Selective Implantation of Diamines for Cooperative Catalysis in Isoreticular Heterometallic Titanium–Organic Frameworks
2021
[EN] We introduce the first example of isoreticular titanium-organic frameworks, MUV-10 and MUV-12, to show how the different affinity of hard Ti(IV) and soft Ca(II) metal sites can be used to direct selective grafting of amines. This enables the combination of Lewis acid titanium centers and available -NH, sites in two sizeable pores for cooperative cycloaddition of CO2 to epoxides at room temperature and atmospheric pressure. The selective grafting of molecules to heterometallic clusters adds up to the pool of methodologies available for controlling the positioning and distribution of chemical functions in precise positions of the framework required for definitive control of pore chemistr…
Intra-Helical Salt Bridge Contribution to Membrane Protein Insertion.
2021
ABSTRACTSalt bridges between negatively (D, E) and positively charged (K, R, H) amino acids play an important role in protein stabilization. This has a more prevalent effect in membrane proteins where polar amino acids are exposed to a very hydrophobic environment. In transmembrane (TM) helices the presence of charged residues can hinder the insertion of the helices into the membrane. This can sometimes be avoided by TM region rearrangements after insertion, but it is also possible that the formation of salt bridges could decrease the cost of membrane integration. However, the presence of intra-helical salt bridges in TM domains and their effect on insertion has not been properly studied ye…
Don’t trust the modern environmental epidemiologists!
2021
Air pollution is responsible for more than 400.000 premature deaths/year in Europe – so German newspapers and other media complained at 9 September 2020, refering to the European Environment Agency [1]. If you ask the scientic base of this statement with severe political and economic consequences, you will find numerous studies since many years. Nearly all these studies describe “associations”, “interactions” ore model calculations regarding air pollution and regional incidences, but for the author not a convincing proof for causality. Correlations don’t proof causal interactions, they include a hight risk of stork- -statistics. A similar case exist in the risk-level of NOx, decided by the …
PLGA Nanoparticles Co-encapsulating NY-ESO-1 Peptides and IMM60 Induce Robust CD8 and CD4 T Cell and B Cell Responses
2021
Contains fulltext : 232076.pdf (Publisher’s version ) (Open Access) Tumor-specific neoantigens can be highly immunogenic, but their identification for each patient and the production of personalized cancer vaccines can be time-consuming and prohibitively expensive. In contrast, tumor-associated antigens are widely expressed and suitable as an off the shelf immunotherapy. Here, we developed a PLGA-based nanoparticle vaccine that contains both the immunogenic cancer germline antigen NY-ESO-1 and an α-GalCer analog IMM60, as a novel iNKT cell agonist and dendritic cell transactivator. Three peptide sequences (85-111, 117-143, and 157-165) derived from immunodominant regions of NY-ESO-1 were se…
Autocorrelation Metrics to Estimate Soil Moisture Persistence From Satellite Time Series: Application to Semiarid Regions
2021
Satellite-derived soil moisture (SM) products have become an important information source for the study of land surface processes in hydrology and land monitoring. Characterizing and estimating soil memory and persistence from satellite observations is of paramount relevance, and has deep implications in ecology, water management, and climate modeling. In this work, we address the problem of SM persistence estimation from microwave sensors using several autocorrelation metrics that, unlike traditional approaches, build on accurate estimates of the autocorrelation function from nonuniformly sampled time series. We show how the choice of the autocorrelation estimator can have a dramatic impac…
Phonons of hexagonal BN under pressure: Effects of isotopic composition
2021
Raman scattering experiments on isotopically enriched hexagonal boron nitride have been performed under pressure up to 11 GPa at room temperature. The sublinear increase of the Raman-active E2g mode frequencies has been characterized. The pressure behavior has been analyzed by means of a bond-stiffness–bond-length scaling parameter γ which takes into consideration the vast differences in a- and c-axis compressibilities. The interlayer shear mode exhibits a γ parameter similar to that of graphite, and the mode frequency in isotopically pure samples separates faster at low pressures as a result of van der Waals interactions. Because of the extremely low a-axis compressibility, the intralayer …
Kinetic and static perimetry after 16 years and additional OCT-A analysis in eyes with long-lasting optic disc drusen
2021
The aim of the study is to evaluate the progression of visual field (VF) defects over 16 years of observation and to assess abnormalities in vessels and retinal nerve fibre layer (RNFL) thickness in patients with optic disc drusen (ODD). Both static automated perimetry (SAP) and semi-automated kinetic perimetry (SKP) were performed in 16 eyes of 8 patients (mean age 54 years) with ODD among 26 eyes of 13 patients examined 16 years before. The area of I2e, I4e, III4e, and V4e isopters was measured in deg2. The MD and PSD parameters were estimated using SAP. Optical coherence tomography angiography (OCT-A) was additionally performed in 16 ODD eyes and 16 eyes of 8 healthy subjects to estimate…
Consistent Treatment of Axions in the Weak Chiral Lagrangian.
2021
We present a consistent implementation of weak decays involving an axion or axion-like particle in the context of an effective chiral Lagrangian. We argue that previous treatments of such processes have used an incorrect representation of the flavor-changing quark currents in the chiral theory. As an application, we derive model-independent results for the decays $K^-\to\pi^- a$ and $\pi^-\to e^-\bar\nu_e a$ at leading order in the chiral expansion and for arbitrary axion couplings and mass. In particular, we find that the $K^-\to\pi^- a$ branching ratio is almost 40 times larger than previously estimated.
Innovative method to estimate state of charge of the hydride hydrogen tank: application of fuel cell electric vehicles
2021
International audience; Significant attention has been paid to metal hydrides (MH) as an environmentally friendly and safe way to store hydrogen. This technology has considerable potential for the application of embedded hydrogen storage in fuel cell electric vehicles, but its widespread application faces a major problem in terms of estimating the remaining hydrogen amount in the tank. In this work, a new method is proposed for estimating the state of charge (SoC) of the hydrogen hydride tank (HHT) by application of piezoelectric material. The idea is to cover the entire inner wall of the metal-hydride tank with a layer of piezoelectric material. During the process of hydrogen absorption, t…
Range-wide phylogeography and taxonomy of the marine rock pools dweller Tigriopus fulvus (Fischer, 1860) (Copepoda, Harpacticoida)
2021
In the light of the wide distribution and ecological importance of the genus Tigriopus in coastal rock pool habitats, and of its frequent use in aquaculture and as a model organism, we investigated the identity of the Atlantic–Mediterranean Tigriopus populations and elucidated their taxonomy and patterns of morphological and genetic diversity. In order to reach these goals, an “integrative taxonomy” multisource approach was implemented. First, we investigated the constancy and taxonomical value of the morphological characters currently used to distinguish among the Tigriopus species occurring in the Mediterranean and in Eastern Atlantic area north of the Tropic of Cancer, and checked the ac…

Il neonato che “sa di sale”
2021
Pseudohypoaldosteronism type 1 (PHA1) is a rare genetic disease due to the peripheral resistance to aldosterone. Clinical spectrum with neonatal onset includes salt loss, hyponatremia, hypochloraemia, hyperkalaemia, metabolic acidosis and increased plasmatic levels of aldosterone. Two forms of the disease - renal and systemic – have been described, which are genetically distinct and with wide clinical expressivity. The most severe generalized PHA1 is caused by mutations in the genes encoding for the subunits of the epithelial sodium channels (ENaC). The paper reports the case of a newborn of the first pregnancy of healthy and consanguineous Sicilian parents, with a clinical and hormonal pic…
Timeout procedure in paediatric surgery: effective tool or lip service? A randomised prospective observational study.
2021
BackgroundFor over a decade, the preoperative timeout procedure has been implemented in most paediatric surgery units. However, the impact of this intervention has not been systematically studied. This study evaluates whether purposefully introduced errors during the timeout routine are detected and reported by the operating team members.MethodsAfter ethics board approval and informed consent, deliberate errors were randomly and clandestinely introduced into the timeout routine for elective surgical procedures by a paediatric surgery attending. Errors were randomly selected among wrong name, site, side, allergy, intervention, birthdate and gender items. The main outcome measure was how freq…

2021
Background: We investigated whether the number, duration and intensity of respiratory arousals (RA) on C3-electroencephalographic (EEG) recordings correlate with polysomnography (PSG)-related disease severity in obstructive sleep apnea (OSA) patients. We also investigated if every patient might have an individual RA microstructure pattern, independent from OSA-severity. Methods: PSG recordings of 20 OSA patients (9 female
Photon frequency shift in curvature based Extended Theories of Gravity
2021
We study the frequency shift of photons generated by rotating gravitational sources in the framework of curvature based Extended Theories of Gravity. The discussion is developed considering the weak-field approximation. Following a perturbative approach, we analyze the process of exchanging photons between Earth and a given satellite, and we find a general relation to constrain the free parameters of gravitational theories. Finally, we suggest the Moon as a possible laboratory to test theories of gravity by future experiments which can be, in principle, based also on other Solar System bodies.
Characterization of 150 Wheat Cultivars by LC-MS-Based Label-Free Quantitative Proteomics Unravels Possibilities to Design Wheat Better for Baking Qu…
2021
Wheat (Triticum aestivum ssp. aestivum) contributes to 20% of the human protein supply, delivers essential amino acids and is of fundamental importance for bread and pasta quality. Wheat proteins are also involved in adverse human reactions like celiac disease (CD), wheat allergy (WA) and non-celiac wheat sensitivity (NCWS). Using liquid chromatography-mass spectrometry (LC-MS)-based label-free quantitative (LFQ) proteomics of aqueous flour extracts, we determined 756 proteins across 150 wheat cultivars grown in three environments. However, only 303 proteins were stably expressed across all environments in at least one cultivar and only 89 proteins thereof across all 150 cultivars. This und…

Chemically Fueled Volume Phase Transition of Polyacid Microgels
2021
Abstract Microgels are soft colloids that show responsive behavior and are easy to functionalize for applications. They are considered key components for future smart colloidal material systems. However, so far microgel systems have almost exclusively been studied in classical responsive switching settings using external triggers, while internally organized, autonomous control mechanisms as found in supramolecular chemistry and DNA nanotechnology relying on fuel‐driven out‐of‐equilibrium concepts have not been implemented into microgel systems. Here, we introduce chemically fueled transient volume phase transitions (VPTs) for poly(methacrylic acid) (PMAA) microgels, where the collapsed hydr…
The starry sky, a territorial commons?
2021
International audience; Flagstaff, Arizona, United States, April 15, 1958: the city council adopts the ordinance n°440 which aims at “preventing a rapid deterioration of the visibility of the starry sky”. This regulation results from the mobilization of the Lowell Observatory astronomers against certain public lighting devices. Through this political act, the elected officials place their city at the epicenter of an emergent international movement: the ‘Dark-sky movement’. The first replica of this movement is felt in 1972 in Tucson, Arizona. From a discomfort perceived by astronomers from the Lowell and Kitt Peak observatories, the idea of ‘light pollution’ emerges and begins to spread thr…
Evolutionary priming and transition to the ectomycorrhizal habit in an iconic lineage of mushroom-forming fungi: is preadaptation a requirement?
2021
AbstractThe ectomycorrhizal symbiosis is an essential guild of many forested ecosystems and has a dynamic evolutionary history across kingdom Fungi, having independently evolved from diverse types of saprotrophic ancestors. In this study, we seek to identify genomic features of the transition to the ectomycorrhizal habit within the Russulaceae, one of the most diverse lineages of ectomycorrhizal fungi. We present comparative analyses of the pangenome and gene repertoires of 21 species across the order Russulales, including a closely related saprotrophic member of Russulaceae. The ectomycorrhizal Russulaceae is inferred to have originated around the Cretaceous-Paleogene extinction event (73.…
