Mutations in the mu heavy-chain gene in patients with agammaglobulinemia.
1996
Most patients with congenital hypogammaglobulinemia and absent B cells are males with X-linked agammaglobulinemia, which is caused by mutations in the gene for Bruton's tyrosine kinase (Btk); however, there are females with a similar disorder who do not have mutations in this gene. We studied two families with autosomal recessive defects in B-cell development and patients with presumed X-linked agammaglobulinemia who did not have mutations in Btk.A series of candidate genes that encode proteins involved in B-cell signal-transduction pathways were analyzed by linkage studies and mutation screening.Four different mutations were identified in the mu heavy-chain gene on chromosome 14. In one fa…

Consistent Treatment of Relativistic Effects in Electrodisintegration of the Deuteron
1996
The influence of relativistic contributions to deuteron electrodisintegration is systematically studied in various kinematic regions of energy and momentum transfer. As theoretical framework the equation-of-motion and the unitarily equivalent S-matrix approaches are used. In a (p/M)-expansion, all leading order relativistic $\pi$-exchange contributions consistent with the Bonn OBEPQ model are included. In addition, static heavy meson exchange currents including boost terms, $\gamma\pi\rho/\omega$-currents, and $\Delta$-isobar contributions are considered. Sizeable effects from the various relativistic two-body contributions, mainly from $\pi$-exchange, have been found in inclusive form fact…
Cloning of Sponge (Geodia cydonium) and Tunicate (Botryllus schlosseri) Proteasome Subunit Epsilon (PRCE): Implications about the Vertebrate MHC-Enco…
1996
Proteasomes are large protein complexes that play a major role in selective degradation of intracellular proteins. Eukaryotes feature seven different alpha and beta subunits. Two of the vertebrate housekeeping beta-subunits have MHC-encoded homologues that can substitute for the housekeeping counterparts upon interferon-gamma induction. In the present study we report the cloning of invertebrate beta-subunit proteasome epsilon (PRCE), from the marine sponge Geodia cydonium and from the colonial tunicate Botryllus schlosseri. Sequence comparisons revealed that the sponge and tunicate proteins are strikingly similar to vertebrate and yeast PRCEs and their MHC-linked counterparts the PRCCs (als…
Trichinella spiralisThymidylate Synthase: Developmental Pattern, Isolation, Molecular Properties, and Inhibition by Substrate and Cofactor Analogues
1996
Abstract Thymidylate synthase specific activity was found to remain at a constant level in crude extracts from muscle larvae, isolated (1-15 months after infection) by pepsin-HCl digestion, as well as from adult worms ofTrichinella spiralis. The enzyme was purified and its molecular (monomer mol. wt 35 kD) and kinetic (sequential mechanism with the Kmvalues 3.1 and 19 μM for dUMP and N5,10-methylenetetrahydrofolate, respectively) properties determined. 5-Fluoro-dUMP was a competitive, slow-binding inhibitor of the parasite enzyme. N5,10-methylenetetrahydrofolate analogues 10-propargyl-5,8-dideazafolate (CB3717), ZD1694, BW1843U89, and AG337 were weaker inhibitors of the parasite than regener…
Titanium Complexes of Chelating Bis(phenolato) Ligands with Long Titanium−Sulfur Bonds. A Novel Type of Ancillary Ligand for Olefin Polymerization Ca…
1996
The sulfide-linked bis(phenol) tbmpH2 (tbmp = 2,2‘-thiobis(6-tert-butyl-4-methylphenolato)) gives a series of titanium complexes of the formula [Ti(tbmp)X2]2 or [Ti(tbmp)(L)X2] (L = two-electron ligand, X = one-electron ligand). The molecular structures of [Ti(tbmp)(OiPr)2]2 and of [Ti(tbmp){C6H4(CH2NMe2)-2}Cl] were determined by single-crystal X-ray structural analyses. An unusually long titanium−sulfur bond of ∼270 pm is observed in both complexes which contain a six-coordinate titanium center. Variable-temperature NMR spectroscopy shows a fluxional coordination of the chelating aryl group in [Ti(tbmp){C6H4(CH2NMe2)-2}Cl].
Stable expression in HEK-293 cells of the rat alpha3/beta4 subtype of neuronal nicotinic acetylcholine receptor.
1996
The alpha3/beta4 subtype of neuronal nicotinic acetylcholine receptor (nAChR) was stably expressed in human embryonic kidney (HEK) 293 cells that co-expressed a voltage-gated Ca2+ channel. alpha3/beta4-nAChR-expressing clones were identified using the fura-2 Ca2+ imaging technique, and were further characterised by single-cell and whole-cell patch-clamp studies. Acetylcholine (ACh) induced fast activating currents which showed desensitisation and inward rectification. The conductance of the ACh-activated channel was 29 pS. The order of potency of the nicotinic agonists tested was cytisine approximately = nicotine > acetylcholine. The EC50 value for ACh was 145 microM; the Hill coefficient w…
A high-temperature x-ray absorption spectroscopy study of
1996
We present for the first time a high-temperature x-ray absorption spectroscopy study of at the edge. The analysis both of the XANES and of the EXAFS regions shows that, in , a progressive localization of the 5d states following the temperature rise is accompanied at T > 350 K by a distortion of the octahedra, which increases up to the decomposition temperature . The distortion leads to the lowering of the rhenium site symmetry from and is due to the electron - phonon coupling of the 5d electrons with the and phonons and to the Jahn - Teller effect caused by the rhenium electron which becomes localized at high temperature.

Neural networks for the diagnostics of gas turbine engines
1996
The paper describes the activities carried out for developing and testing Back Propagation Neural Networks (BPNN) for the gas turbine engine diagnostics. One of the aims of this study was to analyze the problems encountered during training using large number of patterns. Each pattern contains information about the engine thermodynamic behaviour when there is a fault in progress. Moreover the research studied different architectures of BPNN for testing their capability to recognize patterns even when information is noised. The results showed that it is possible to set-up and optimize suitable and robust Neural Networks useful for gas turbine diagnostics. The methods of Gas Path Analysis furn…
Neutral heavy leptons and electroweak baryogenesis
1996
We investigate the possibility that baryogenesis occurs during the weak phase transition in a minimal extension of the Standard Model which contains extra neutral leptons and conserves total lepton number. The necessary CP-violating phases appear in the leptonic Yukawa couplings. We compute the CP-asymmetries in both the neutral and the charged lepton fluxes reflected on the bubble wall. Using present experimental bounds on the mixing angles and Standard Model estimates for the parameters related to the scalar potential, we conclude that it seems unlikely to produce the observed baryon to entropy ratio within this type of models. We comment on the possibility that the constraints on the mix…

Growth characteristics of sputter-deposited thin films
1996
Thin films of the heavy-fermion superconductor were deposited on various substrate materials in various orientations by means of a quasi-multilayer sputter process. Strongly (0001)-textured growth of the hexagonal compound was found for a uranium content in the range of 23% to 28% on sapphire and with perfect in-plane order on the latter substrate material. Atomic force microscopy and scanning electron microscopy revealed a Vollmer - Weber-like growth mode resulting in the development of large compressive strain in films on . As a result the electronic transport properties - in particular the temperature dependence of the resistivity - were strongly renormalized. Strong deviations from the …

Local Entropy Characterization of Correlated Random Microstructures
1996
A rigorous connection is established between the local porosity entropy introduced by Boger et al. (Physica A 187, 55 (1992)) and the configurational entropy of Andraud et al. (Physica A 207, 208 (1994)). These entropies were introduced as morphological descriptors derived from local volume fluctuations in arbitrary correlated microstructures occuring in porous media, composites or other heterogeneous systems. It is found that the entropy lengths at which the entropies assume an extremum become identical for high enough resolution of the underlying configurations. Several examples of porous and heterogeneous media are given which demonstrate the usefulness and importance of this morphologic…

Immunomodulatory and Hematopoietic Effects of Recombinant Human Interleukin-6 in Patients with Advanced Renal Cell Cancer
1996
Interleukin-6 (IL-6) is a cytokine with pleiotropic biologic activities on B cells, T cells, and hematopoietic progenitors. The present study was undertaken to assess pharmacodynamic effects of subcutaneous administration of IL-6 on blood counts, immunologic parameters, and acute-phase reactants. Blood samples were taken from patients with advanced renal cell cancer participating in a phase II trial of recombinant human IL-6. Multiparameter FACS analyses of peripheral blood mononuclear cells were performed using antibodies against CD3, CD4, CD8, HLA-DR, CD56, CD28, CD38, CD19, sIgM, and sIgG. Serum levels of IL-10, soluble CD23 (sCD23), sCD25, IL-1 receptor antagonist protein (IL-1RA), solu…
First observation of strong OZI rule violation in πN interactions
1996
The data of the CERN WA56 experiment that triggered the fast proton produced in the $\pi^{+}p$ and $\pi^{-}p$ interactions at beam momenta 20 GeV/c and 12 GeV/c, respectively, are used to analyse the final states $ p_{f} \phi (\omega)\pi^{\pm}$. A large excess (up to two orders of magnitude) of the $\phi/\omega$ cross sections ratio over the OZI prediction is observed.
Distribution of basal-expressed c-fos-like immunoreactive cells of the periaqueductal grey matter of the rat.
1996
The distribution of c-fos proto-oncogene expression has been studied in the periaqueductal grey matter (PAG) of non-intentional-stimulated rats by immunohistochemistry. A mean number of 53 +/- 9 Fos-like immunoreactive cells per hemiPAG, distributed into three groups, dorsolateral, lateral and ventrolateral, was found. The dorsolateral and the lateral groups appeared along the entire rostrocaudal PAG, whereas the ventrolateral group appeared only in the caudal half. These results reveal new data on the number and extent of the longitudinal columns within the hypothetical organization of the PAG. They also indicate a continuous activation of a significant population of neurones in the PAG, i…
Large deviations results for subexponential tails, with applications to insurance risk
1996
AbstractConsider a random walk or Lévy process {St} and let τ(u) = inf {t⩾0 : St > u}, P(u)(·) = P(· | τ(u) < ∞). Assuming that the upwards jumps are heavy-tailed, say subexponential (e.g. Pareto, Weibull or lognormal), the asymptotic form of the P(u)-distribution of the process {St} up to time τ(u) is described as u → ∞. Essentially, the results confirm the folklore that level crossing occurs as result of one big jump. Particular sharp conclusions are obtained for downwards skip-free processes like the classical compound Poisson insurance risk process where the formulation is in terms of total variation convergence. The ideas of the proof involve excursions and path decompositions for Mark…
Was heißt wie? Ansatz und Glossar zu Befundung und Verständnis in der HRCT der Lunge
1996
In HRCT reports multiple different, often synonymous, German and English terms are used. The variety of terms impede understanding and acceptance of HRCT. Purpose of this paper is to present a scheme, which is based on the anatomic landmarks (secondary lobule), and the density of pathologic changes, as well as a glossary from the German HRCT-literature, including suitable terms, definitions, synonyms and English terms. Low attenuation changes include emphysemas, air-filled cavities (bullae, cysts, cavitations, honeycombing) and bronchial dilatation, changes with increased density consist of diffuse (ground glass opacity, consolidation) and focal processes (reticular and nodular densities). …
HCV viraemia is more important than genotype as a predictor of response to interferon in sicily (Southern Italy)
1996
Abstract Background/Aims: To investigate host- and virus-related factors predictive of early and sustained alanine aminotransferase normalization after interferon therapy for HCV-related chronic liver disease, in an area where genotype 1 is highly prevalent. Methods: We studied 100 patients with HCV-RNA positive chronic liver disease (73 chronic hepatitis and 27 cirrhosis) undergoing alpha-interferon treatment. Thirty-four patients had an early response but relapsed, 15 patients remained into sustained response for at least 12 months after therapy, and 51 patients did not respond. Serum HCV-RNA levels were assessed by bDNA (Chiron), and genotype by LiPA (Innogenetics) and by sequencing of t…
Direct Objective Quantification of Corneal Haze after Excimer Laser Photorefractive Keratectomy for High Myopia
1996
Purpose: The purpose of the study is to measure regional distribution differences in corneal haze after excimer laser photorefractive keratectomy for high myopia. Methods: The authors developed computerized gradient edge detectors with which were analyzed digitized anterior slit-lamp photographs of 40 eyes, an average of 21.0 plus or minus 14.5 weeks after photorefractive keratectomy for high myopia (−6 to −22 diopters). A treated area and an adjacent untreated area on the anterior corneal surface, each containing six regions, were quantified, and the difference was correlated with various parameters. Results: Mean differences between scarred and clear areas for haze grade 0.5, 1.0, 2.0, 3.…
Costimulatory signalling potential of murine MHC class II‐positive T‐clone cells
1996
Activated human and rat T cells as well as mouse T-cell clones have been reported to synthesize and express major histocompatibility complex (MHC) class II molecules. However, the capacity of class II+ antigen (Ag) presenting T cells to induce proliferation of Ag-specific cloned T cells has been controversial. We analysed whether the failure of some T-cell clones to proliferate in response to Ag presented by class II+ T cells is because of a lack of costimulatory cytokine production by the antigen-presenting cells (APC). As a model system the mouse class II+ cloned BI/O4.1 T cells were used as APC in order to activate the T cell clone KIII5. This T-helper 1 (Th1) type, GAT (synthetic copoly…
Modulatory Role of Endothelial and Nonendothelial Nitric Oxide in 5-Hydroxytryptamine-induced Contraction in Cerebral Arteries after Subarachnoid Hem…
1996
OBJECTIVE : Endothelial dysfunction is claimed to play a role in the pathogenesis of delayed cerebral vasospasm after subarachnoid hemorrhage (SAH). We have examined the effect of experimental SAH on the modulatory action of endothelial and nonendothelial nitric oxide (NO) in the contractile response of goat middle cerebral artery to 5-hydroxytryptamine (5-HT). METHODS : We compared the 5-HT-induced contractile responses of cerebral arteries from control goats and from goats with SAH that had been experimentally induced 3 days earlier by delivery of autologous arterial blood into the subarachnoid space. Contractile responses were examined by recording the isometric tension in isolated cereb…
Specific stabilization of the 4F7 molecule on dendritic cells by contact allergens.
1996
Our laboratory has recently developed the monoclonal antibody 4F7 which recognizes a molecule on dendritic cells in the dermis of mice that is upregulated after application of contact allergens in vivo. Furthermore, this antibody detects an antigen on dendritic cells in spleen, lymph nodes and colon. In order to study the influence of contact allergens on the surface expression of the 4F7 molecules on dendritic cells, FACScan analysis of splenic dendritic cells was carried out after in vitro application of contact allergens. Freshly isolated splenic dendritic cells were found to be positive for 4F7, 33D1, N418 (CD11c) and MHC class II. After overnight culture the expression of the dendritic…
