Selectivity of Neutrophil 5-Lipoxygenase and Cyclo-oxygenase Inhibition by an Anti-inflammatory Flavonoid Glycoside and Related Aglycone Flavonoids
1988
Abstract A newly described plant-derived flavonoid, hypolaetin-8-glucoside, which has anti-inflammatory and gastroprotective actions in-vivo, and its corresponding aglycone, hypolaetin, have been compared with 14 other flavonoids for inhibition of eicosanoid generation via the 5-lipoxygenase and cyclo-oxygenase pathways in elicited rat peritoneal leukocytes stimulated with calcium ionophore. Comparable results for the inhibitory profiles of the compounds were obtained using either radioimmunoassay of released eicosanoids or radio-TLC of metabolites formed from labelled arachidonate, but there were differences in absolute potency of the inhibitors. Hypolaetin-8-glucoside was a weak but selec…
Brain density in depression: methodological and psychopathological aspects
1988
The relationship between brain density, measured by computerized tomography (CT), and severity of depression was investigated in 44 patients with a major depressive episode according to DSM-III. In order to limit methodological problems, correlations between both the Brief Psychiatric Rating Scale (BPRS) and the Bech-Rafaelsen Melancholia Scale (BRMS) with density values were controlled for age, different ventricle measurements, brain size, and density and size of the skull. The BRMS score correlated inversely with density of the right thalamus, the right head of the caudate, and with parietal grey matter and occipital regions of both hemispheres. Similar, but nonsignificant results, were o…
Differential effects of potassium channel blockers on neurohypophysial release of oxytocin and vasopressin. Evidence for frequency-dependent interact…
1988
Isolated rat neurohypophyses were fixed by their stalks to a platinum wire electrode and superfused with Krebs-HEPES solution. Vasopressin and oxytocin released into the medium were determined by specific radioimmunoassays. Hormone secretion was increased by electrical stimulation of the pituitary stalk at different frequencies. The effects of several potassium channel blockers, tetraethylammonium (TEA) ions, 4-aminopyridine (4-AP) and 3,4-diaminopyridine (3,4-DAP) were tested. The release of vasopressin and oxytocin evoked by electrical stimulation with 900 pulses at 15 Hz (about 900 and 1,000 μU, respectively) was about 10 times higher than that evoked by 900 pulses at 3 Hz. Both 10 and 3…
Blood lactate production and recovery from anaerobic exercise in trained and untrained boys.
1988
Blood lactate production and recovery from anaerobic exercise were investigated in 19 trained (AG) and 6 untrained (CG) prepubescent boys. The exercises comprised 3 maximal test performances; 2 bicycle ergometer tests of different durations (15 s and 60 s), and running on a treadmill for 23.20±2.61 min to measure maximal oxygen uptake. Blood samples were taken from the fingertip to determine lactate concentrations and from the antecubital vein to determine serum testosterone. Muscle biopsies were obtained from vastus lateralis. Recovery was passive (seated) following the 60 s test but that following the treadmill run was initially active (10 min), and then passive. Peak blood lactate was hi…
Multiple Peptide Hormone Producing Adenocarcinoma of Lung with Neurotensin and CRF-like Immunoreactivity
1988
Summary A pulmonary cancer of a 43 year-old-man was classified according to WHO-criteria as a poorly differentiated adenocarcinoma because of demonstration of mucinous substances in tumor cells and in rarely occurring acinar lumina. Immunohistochemical reactivity for several peptide hormones was detected in the predominant solid parts of the carcinoma. In many tumor cells an immunoreactivity to neurotensin, bombesin and — according to the elevated serum levels — to ACTH, CRF and calcitonin was found. Therefore the present lung cancer must be defined as a combined carcinoma composed of mucous producing and endocrine cell types. Because peptide hormone production is not restricted to the clas…
Entzündungshemmende wirkstoffe. XI. Ringschlußreaktion an 5-phenyl-2-thiobiuret
1988
The reaction of 5-phenyl-2-thiobiuret (1) with α-chlorocarbonyl compounds 2 leads to the structure type of N-phenyl-N1-(thiazolyl)urea (3). The new compounds exhibit particularly antiinflammatory, anthelminthic, and trichomonacidal activity.
Mass spectra of halogenated esters: 8. Methyl esters of 2,3-dichloro-, bromochloro- and dibromopropenoic acids
1988
The mass spectral fragmentation of methyl esters of E and Z isomers of 2,3-dichloro-, 2-bromo-3-chloro-, 3-bromo-2-chIoro- and 2,3-dibromopropenoic acids have been investigated. The M+˙ peak is shown with all isomers, the [M − OCH3]+, [M − X]+, [M − OCH3 − CO]+, [M − OCH3 − CO − X]+˙ and [M − OCH3 − CO − X − X]+ ions constituting abundant peaks in all spectra. The results, particularly from the bromochloro isomers, show that a halogen atom is eliminated from the 3- rather than the 2- position and from the Z rather than the E isomer. Bromine as a bulky atom is preferentially lost.
Immunosuppressant therapy of thyroid eye disease.
1988
Thyroid eye disease is attributed to an autoimmune process where both cellular and humoral immunity play a role. In this report, after a short introduction dealing with immunopathogenesis of the disease, immunosuppressant therapy is discussed. Treatment with glucocorticoids (as the standard substance), nonsteroid immunosuppressants (azathioprine, cyclophosphamide, lately cyclosporin) and with the immunomodulatory substance ciamexone is reviewed. Retroorbital irradiation as a "local" immunosuppressive method and plasmapheresis are also discussed. While systemic glucocorticoids and to a lesser extent orbital radiotherapy are routinely administered for severe Graves' ophthalmopathy, nonsteroid…

Contact-dependent inhibition of growth of normal diploid human fibroblasts by plasma membrane glycoproteins.
1988
Homeostasis in vivo is maintained by a highly complex network of positive and negative signals. At the cellular level, this regulatory microenvironment can be divided, in a simplified fashion, into two major compartments: the humoral compartment, including compounds such as hormones, growth factors and nutrients, and the contact-environment compartment, including cell-cell and cell-matrix interactions. At least in cultures of diploid, non-transformed cells, cell-cell and cell-matrix interactions have been shown to be of major importance for the regulation of growth as well as of differentiation. Although until now the glycoprotein involved in the contact-dependent inhibition of growth has n…
Cloned T helper cells reverting to a resting state develop increasing sensitivity in their antigen-mediated interaction with accessory cells.
1988
A cloned murine T cell line, KIII5, specific for the polypeptide poly-L(Tyr,Glu)-poly-D,L-Ala--poly-L-Lys [(T,G)-A--L] was compared at different stages after antigenic stimulation with respect to the conditions required for the reinduction of growth by varying concentrations of antigen presented on different types of accessory cells (AC). We show that the dose of antigen necessary for inducing half maximal proliferation in the presence of splenic AC shifts to considerably lower concentrations when the T cell blasts revert to a resting state (100 micrograms/ml on day 7 to 10 micrograms/ml on day 21-35). During the same time period the expression of interleukin 2 (IL2) receptor and the reacti…
Functional reconstitution of a proton-translocating system responsive to fusicoccin
1988
Crude fusicoccin binding proteins and a partially purified plasma membrane H+-transporting ATPase (EC 3.6.1.34), both solubilized from maize tissues, were simultaneously inserted into liposomes by the freeze-thaw method. ATP-driven intravesicular acidification in the proteoliposomes, measured by the fluorescence quenching of the dye 9-amino-6-chloro-2-methoxyacridine, markedly increased upon addition of fusicoccin to the reconstituted system. This effect could not be observed when binding sites and ATPase preparations were separately reconstituted into the proteoliposomes, thus demonstrating that fusicoccin binding to its receptor is a prerequisite for ATPase stimulation.

Mass spectroscopic analysis of excimer laser ablated material from human corneal tissue.
1988
The clinical feasibility of photorefractive keratectomy depends on the surface structure of the ablated cornea. Two factors that influence the remodeled surface are the homogeneity of the energy distribution and the properties of the laser source (energy and wavelength). Currently, the homogeneity of the beam is difficult to control. The second factor, laser source properties, was the focus of this study. We investigated the effect of laser wavelength and energy by analyzing the reaction products of photoablation. We monitored the fragments produced by UV-laser ablation of human corneas using mass spectroscopy in the range of 0 to 100 atomic mass units. At 248 nm (KrF), average photon energ…
The intraclonal and interclonal phenotypic heterogeneity in a rhabdomyosarcoma cell line with abortive imitation of embryonic myogenesis
1988
Three distinct subpopulations (A, B, C) derived from a dimethylbenzanthracene-induced rat rhabdomyosarcoma were established as permanent cell lines. Although the clonal nature of each of these subpopulations was confirmed by repeated recloning procedures, a striking intraclonal phenotypic heterogeneity was observed. By means of immunofluorescence microscopy and transmission electron microscopy, it could be shown that these subpopulations closely recapitulate stages of embryonic rhabdomyogenesis both in vitro and in vivo, but differ in their particular range of maximum differentiation. Embryonic rhabdomyogenesis is imitated most perfectly by subpopulation C, in which multinuclear myotubes ar…
Wilms tumours diagnosed by routine physical examinations.
1988
Sir: The prognosis of stage I and -II Wilms tumours with favourable histology is good, reaching 97% (stage I) and 92% (stage II) 5 year disease-free survival rates [1]. With these figures in mind the question arises whether the overall prognosis of Wilms tumours could be improved by presymptomatic routine physical examinations in an attempt to avoid stage III to V primary diagnoses. In the Federal Republic of Germany six routine physical examinations (RPE) are offered to infants, and two additional ones to children between 12 and 48 months. The 1980-1987 German Wilms tumour study [1] listed 313 patients, 29 of whom (9%) were diagnosed by such routine examinations. As shown in Table 1, the R…

N-Terminal amino acid sequence analysis indicates that isolated atrial amyloid is derived from atrial natriuretic peptide
1988
Isolated atrial amyloid, the most frequent senile cardiac amyloid type, was chemically analysed. Amyloid fibrils obtained from a patient (NIP) were extracted and the predominant low-molecular-weight polypeptide (approximately 3.5 kDa, designated ASc2 NIP) was isolated by size exclusion high performance liquid chromatography in 60% formic acid. N-Terminal amino acid sequence analysis of this polypeptide was identical to that of the atrial natriuretic peptide alpha-hANP for the first 12 residues determined.
Radioactively labelled epoxides. Part VI. tritium-labelled mono- and dimethyl substituted phenyl oxiranes (styrene oxides)
1988
Tritium-labelled (E)- and (Z)-2,3-dimethyl-2-phenyl oxirane 4, (E)- and (Z)-2-methyl-3-phenyl oxirane 7 and 2,2-dimethyl-3-phenyl oxirane 11 have been prepared by reduction of the corresponding bromoketones with sodium borotritide to the corresponding bromohydrins followed by cyclization to the oxiranes. These oxiranes were successfully used as diagnostic substrates to distinguish between different forms of epoxide hydrolase and glutathione transferase.

Crystal growth and hysteresis phenomena of rubidium tetrachlorozincate Rb2ZnCl4
1988
Abstract Phase transitions in Rb2ZnCl4 crystals grown by various methods are examined with differential scanning calorimetry and dielectric measurements. For most of the studied samples the permittivity along the ferroelectric axis shows a large thermal hysteresis over a wide temperature range including the incommensurate phase and the lock-in point. This hysteresis depends on the presence of different defects which appear during crystal growth or after a thermal and mechanical treatment. It is shown that dielectric measurements are quite well suited to characterize the crystals exhibiting on incommensurate phase and a ferroelectric phase.
PATTERNS OF PUFFING ACTIVITY AND CHROMOSOMAL POLYMORPHISM IN DROSOPHILA SUBOBSCURA . IV. EFFECT OF INVERSIONS ON GENE EXPRESSION
1988
We have observed that, contrary to a common assumption, the puffing patterns manifest in the salivary chromosomes of Drosophila subobscura are modified by chromosomal inversions as well as by genic content. An inversion effect is apparent in the E and A chromosomes of five strains coming from four different natural populations. An effect due to the geographical location of the populations is also detected in the J and O chromosomes. The chromosomal and geographic effects are distinguishable but not contradictory. Indeed, a statistical test using the DK2 coefficient of distance shows that, for a given chromosomal arrangement, strains of different geographic origin exhibit puffing patterns si…

Scoring Criteria for Electrodermal Habituation: Further Research
1988
In the context of Levinson and Edelberg's critique of scoring criteria for electrodermal habituation, the present study examined the question of whether trials-to-habituation scores based on two no-response trials are superior to scores based on three no-response trials. Male students (N=120) performed two identical habituation experiments on two consecutive days and their skin conductance responses based on a short latency window of 1–3 s were analyzed. In each experiment subjects received 20 presentations of a 1000 Hz tone at 65dB. Results showed that three-trials scores were higher overall and that the distributions of three- and two-trials scores differed. On the other hand, the twoscor…
Gefäßrekanalisation mit einem elektrischen Thermokauter-Katheter -In-vitro-Erfahrungen mit einem neuen Kathetersystem
1988
Recanalisation of arteriosclerotic occlusions by means of laser angioplasty, Kensey catheter and the "hot tip" catheter is undergoing early, successful clinical trials. The thermo-cautery catheter described in this paper uses the well known technique of cutting with high frequency electric currents in order to achieve vascular recanalisation. Seven attempts were made to recanalise occlusions at post mortem; four were successful, but perforation occurred in three. Histologically we found a narrow rim of carbonisation of tissues surrounded by a zone of homogenisation and a transitional zone.

Muscle metabolic profile and oxygen transport capacity as determinants of aerobic and anaerobic thresholds
1988
Aerobic and anaerobic thresholds determined by different methods in repeated exercise tests were correlated with cardiorespiratory variables and variables of muscle metabolic profile in 33 men aged 20–50 years. Aerobic threshold was determined from blood lactate, ventilation, and respiratory gas exchange by two methods (AerT1 and AerT2) and anaerobic threshold from venous lactate (AnTLa), from ventilation and gas exchange (AnTr) and by using the criterion of 4 mmol·l−1 of venous lactate (AnT4mmol). In addition to ordinary correlative analyses, applications of LISREL models were used. The 8 explanatory variables chosen for the regression analyses were height, relative heart volume, relative …
Effect of Thyroid Hormones on Urea Biosynthesis and Related Processes in Rat Liver*
1988
The results of the few studies on the effect of the thyroid status on nitrogen metabolism have been inconclusive and/or contradictory. In an attempt to elucidate this important relationship, we have studied the effect of experimental hypo- and hyperthyroidism on urea biosynthesis and related processes. We have found that the capacity of the liver to synthesize urea was increased in hypothyroid rats, as were the activities of the urea cycle enzymes; there were also changes in the activities of some related enzymes and in the levels of intermediates and amino acids. Isolated hepatocytes from these rats showed an increased capacity for urea synthesis. In hyperthyroid rats the picture was more …

Pränatale Diagnostik der Sialidose, eines Defektes des lysosomalen Enzyms Neuraminidase
1988
