An exon junction complex‐independent function of Barentsz in neuromuscular synapse growth

2021

The exon junction complex controls the translation, degradation, and localization of spliced mRNAs, and three of its core subunits also play a role in splicing. Here, we show that a fourth subunit, Barentsz, has distinct functions within and separate from the exon junction complex in Drosophila neuromuscular development. The distribution of mitochondria in larval muscles requires Barentsz as well as other exon junction complex subunits and is not rescued by a Barentsz transgene in which residues required for binding to the core subunit eIF4AIII are mutated. In contrast, interactions with the exon junction complex are not required for Barentsz to promote the growth of neuromuscular synapses.…

From obesity to Alzheimer's disease through insulin resistance

2021

Alzheimer's disease is one of the most frequent forms of dementia. It is a progressive neurodegenerative disease, characterized by presence of amyloid plaques and neurofibrillary tangles in the brain. Obesity is regarded as abnormal fat accumulation with deleterious impact on human health. There is full scientific evidence that obesity and the metabolic comorbidities (e.g., insulin resistance, hyperglycaemia, and type 2 diabetes) are related to Alzheimer's disease and likely in the causative pathway. Numerous studies have identified several overlapping neurodegenerative mechanisms, including oxidative stress, mitochondrial dysfunction, and inflammation. In this review, we present how obesit…

Speaking out against everyday sexism : Gender and epistemics in accusations of “mansplaining”

2021

In everyday interaction, subtle manifestations of sexism often pass unacknowledged and become internalised and thus perceived as “natural” conduct. The introduction of new vocabularies for referring to previously unnamed sexist conduct would presumably enable individuals to start problematising hitherto unchallengeable sexism. In this paper, we investigate whether and how these vocabularies empower people to speak out against sexism. We focus on the use of the term “mansplaining” which, although coined over 10 years ago, remains controversial and contested. Using Conversation Analysis and Membership Categorisation Analysis, this paper excavates the interactional methods individuals use to f…

Cytotoxicity of 4-hydroxy-N-(naphthalen-1-yl)-2-oxo-2H-chromene-3-carboxamide in multidrug-resistant cancer cells through activation of PERK/eIF2α/AT…

2021

After decades of research, multidrug resistance (MDR) remains a huge challenge in cancer treatment. In this study, the cytotoxic of 4-hydroxy-N-(naphthalen-1-yl)-2-oxo-2H-chromene-3-carboxamide (MCC1734) has been investigated towards multidrug-resistant cancer cell lines. MCC1734 exerted cytotoxicity on cell lines expressing different mechanisms of drug resistance (P-glycoprotein, BCRP, ABCB5, EGFR, p53 knockout) to a different extent. Interestingly, sensitive CCRF-CEM cells and multidrug-resistant P-gp-overexpressing CEM/ADR5000 cells represented similar sensitivity towards MCC1734, indicating MCC1734 can bypass P-gp-mediated resistance. Microarray-based mRNA expression revealed that MCC17…

Cardiovascular outcomes after cardiac resynchronization therapy in cardiac amyloidosis

2021

Abstract Aims Cardiac resynchronization therapy (CRT) is highly effective in dilated cardiomyopathy (DCM) patients with impaired left ventricular ejection fraction (LVEF) and left bundle block branch. In cardiac amyloidosis (CA) patients, left ventricular dysfunction and conduction defects are common, but the potential of CRT to improve cardiac remodelling and survival in this particular setting remains undefined. We investigated cardiovascular outcomes in CA patients after CRT implantation in terms of CRT echocardiographic response and major cardiovascular events (MACEs). Methods and results Our retrospective study included 47 CA patients implanted with CRT devices from January 2012 to Feb…

Un Punto di vista sul design italiano

2021

The text proposes a reading of the history of design as a narrative struct…

Severe asthma: One disease and multiple definitions

2021

Abstract Introduction There is, so far, no universal definition of severe asthma. This definition usually relies on: number of exacerbations, inhaled therapy, need for oral corticosteroids, and respiratory function. The use of such parameters varies in the different definitions used. Thus, according to the parameters chosen, each patient may result in having severe asthma or not. The aim of this study was to evaluate how the choice of a specific definition of severe asthma can change the allocation of patients. Methods Data collected from the Severe Asthma Network Italy (SANI) registry were analyzed. All the patients included were then reclassified according to the definitions of U-BIOPRED,…

Food: Ordinary Practice or Extraordinary Experience?

2021

Food and the practice of cooking hold a privileged place in contemporary aesthetics, as attested by the extensive literature that has been devoted to this topic. Food has been addressed in both Anglo-American and European studies from multiple points of view, including a cognitivist, pragmatist, phenomenological, everyday and somaesthetic perspective. In this essay I will try to identify a path that allows us to hold together these readings through the ordinary-extraordinary dichotomy. First, I shall analyze food through the lens of the extraordinary, taking into consideration some examples in which food is presented as a true work of art in museums or as an exceptional experience in increa…

Morphology, Rheological and Mechanical Properties of Isotropic and Anisotropic PP/rPET/GnP Nanocomposite Samples

2021

The effect of graphene nanoplatelets (GnPs) on the morphology, rheological, and mechanical properties of isotropic and anisotropic polypropylene (PP)/recycled polyethylene terephthalate (rPET)-based nanocomposite are reported. All the samples were prepared by melt mixing. PP/rPET and PP/rPET/GnP isotropic sheets were prepared by compression molding, whereas the anisotropic fibers were spun using a drawing module of a capillary viscometer. The results obtained showed that the viscosity of the blend is reduced by the presence of GnP due to the lubricating effect of the graphene platelets. However, the Cox–Merz rule is not respected. Compared to the PP/rPET blend, the GnP led to a slight incre…

The Role of Vitamin Deficiency in Liver Disease: To Supplement or Not Supplement?

2021

Over the past few years, growing interest has been shown for the impact of dietary requirements and nutritional factors on chronic diseases. As a result, nutritional programs have been reinforced by public health policies. The precise role of micronutrients in chronic liver disease is currently receiving particular attention since abnormalities in vitamin levels are often detected. At present, treatment programs are focused on correcting vitamin deficiencies, which are frequently correlated to higher rates of comorbidities with poor outcomes. The literature reviewed here indicates that liver diseases are often related to vitamin disorders, due to both liver impairment and abnormal intake. M…

Frailty Is Associated with Oxidative Stress in Older Patients with Type 2 Diabetes

2021

Aging has increased the prevalence of frailty, and type 2 diabetes (T2D) has also increased in prevalence. Diabetes and oxidative stress (OS) have been shown to be related to frailty. However, the exact mechanism by which it occurs is not fully known. Our aim was to analyze body composition in community-dwelling older diabetic people treated in our center and to evaluate the possible relation between OS, frailty, and body composition. We included 100 adults older than 65 years with T2D. We found that 15% were frail and 57% were prefrail. The patients included in the nonrobust group showed increased levels of OS. Our study shows that the presence of T2D in the geriatric population is associa…

Vectofocimetry: Dioptric power measurement in manual focimeters using power vectors

2021

Abstract Dioptric power measurement is classically achieved using manual focimeters. In this contribution, we present vectofocimetry as a novel methodology to be easily implemented in manual focimeters for improving their capabilities regarding dioptric power measurement. The new procedure is conceptually based on power vector formalism of dioptric power which represents the dioptric power as three values in the form of [M, J0, J45], it uses an iterative process of blur reduction, and it only requires a few modifications in the device that are easy-to-adapt and cost-effective. As a result, dioptric power measurements are easily and quickly obtained with comparable results to the reported on…

Comparison of inorganic chlorine in the Antarctic and Arctic lowermost stratosphere by separate late winter aircraft measurements

2021

Stratospheric inorganic chlorine (Cly) is predominantly released from long-lived chlorinated source gases and, to a small extent, very short-lived chlorinated substances. Cly includes the reservoir species (HCl and ClONO2) and active chlorine species (i.e., ClOx). The active chlorine species drive catalytic cycles that deplete ozone in the polar winter stratosphere. This work presents calculations of inorganic chlorine (Cly) derived from chlorinated source gas measurements on board the High Altitude and Long Range Research Aircraft (HALO) during the Southern Hemisphere Transport, Dynamic and Chemistry (SouthTRAC) campaign in austral late winter and early spring 2019. Results are compared to…

Is prolonged febrile syndrome associated with reactive thrombocytosis a possible association in the diagnosis of Takayasu’s arteritis? A case report

2021

Takayasu’s arteritis is a rare, systemic, inflammatory vasculitis of large blood vessels with an unknown aetiology that more frequently affects women of childbearing age with progression to stenosis, fibrosis or thrombogenesis. Clinical manifestations are associated either with inflammation of the vascular wall (including fever, myalgia, arthralgia, weight loss) or the development of aneurysms and extensive vascular lesions, which creates challenges for a differential diagnosis. This current report presents the case of a female Caucasian patient, aged 23 years, that presented herself repetitively at the hospital reporting symptoms including fever, productive cough, myalgia associated with …

Cortical and Cerebellar Oscillatory Responses to Postural Instability in Parkinson's Disease

2021

Introduction: Posture and balance dysfunctions critically impair activities of daily living of patients with progressing Parkinson's disease (PD). However, the neural mechanisms underlying postural instability in PD are poorly understood, and specific therapies are lacking. Previous electrophysiological studies have shown distinct cortical oscillations with a significant contribution of the cerebellum during postural control tasks in healthy individuals. Methods: We investigated cortical and mid-cerebellar oscillatory activity via electroencephalography (EEG) during a postural control task in 10 PD patients with postural instability (PDPI+), 11 PD patients without postural instability (PDPI–…

Improved $K_{e3}$ radiative corrections sharpen the $K_{\mu 2}$--$K_{l3}$ discrepancy

2021

The measurements of $V_{us}$ in leptonic $(K_{\mu 2})$ and semileptonic $(K_{l3})$ kaon decays exhibit a $3\sigma$ disagreement, which could originate either from physics beyond the Standard Model or some large unidentified Standard Model systematic effects. Clarifying this issue requires a careful examination of all existing Standard Model inputs. Making use of a newly-proposed computational framework and the most recent lattice QCD results, we perform a comprehensive re-analysis of the electroweak radiative corrections to the $K_{e3}$ decay rates that achieves an unprecedented level of precision of $10^{-4}$, which improves the current best results by almost an order of magnitude. Research on Unrealistic Optimism among HoReCa Workers as a Possible Future Hotspot of Infections

2021

As we are facing a new surge of the highly infectious delta variant of COVID-19, there is an urgent need for research to reduce the harm before this next wave hits. In the present paper, we present data that is alarming. We have found that HoReCa (hotels, restaurants, and catering services) workers, who are highly exposed to many new social interactions in close contact, present an unrealistic optimism (UO) bias: they perceive themselves as less at risk to this virus in comparison to others. From the literature, we already know that individuals holding this view are less involved in preventive actions and present more risky behaviors. In the face of the delta variant, this leads to the conc…

Abstract Despite islands contributing only 6.7% of land surface area, they harbor ~20% of the Earth’s biodiversity, but unfortunately also ~50% of the threatened species and 75% of the known extinctions since the European expansion around the globe. Due to their geological and geographic history and characteristics, islands act simultaneously as cradles of evolutionary diversity and museums of formerly widespread lineages—elements that permit islands to achieve an outstanding endemicity. Nevertheless, the majority of these endemic species are inherently vulnerable due to genetic and demographic factors linked with the way islands are colonized. Here, we stress the great variation of islands…

Experimental characterization of a novel configuration of thermoelectric refrigerator with integrated finned heat pipes

2021

Abstract Thermoelectric refrigerating systems (TER) are environment-friendly technologies. They have several advantages such as low maintenance costs, are less cumbersome and long lifetime. However, these systems have low coefficient of performance and are limited in achieving low temperature levels. This study investigates a new configuration of TE refrigerator employing a novel arrangement of heat sinks with integrated flat and finned heat pipes. A mathematical model is firstly developed to predict transient temperatures and cooling limit time. An experimental characterization is then performed and it includes optimization of energy consumption, cooling time, temperature levels and TE ref…

Patient-Specific Analysis of Ascending Thoracic Aortic Aneurysm with the Living Heart Human Model

2021

In ascending thoracic aortic aneurysms (ATAAs), aneurysm kinematics are driven by ventricular traction occurring every heartbeat, increasing the stress level of dilated aortic wall. Aortic elongation due to heart motion and aortic length are emerging as potential indicators of adverse events in ATAAs

Oleuropein multicompartment nanovesicles enriched with collagen as a natural strategy for the treatment of skin wounds connected with oxidative stres…

2021

Aim: Collagen-enriched transfersomes, glycerosomes and glytransfersomes were specifically tailored for skin delivery of oleuropein. Methods: Vesicles were prepared by direct sonication and their main physicochemical and technological properties were measured. Biocompatibility, protective effect and promotion of the healing of a wounded cell monolayer were tested in vitro using fibroblasts. Results: Vesicles were mainly multicompartment, small (∼108 nm), slightly polydispersed (approximately 0.27) and negatively charged (~-49 mV). Oleuropein was incorporated in high amounts (approximately 87%) and vesicles were stable during four months of storage. In vitro studies confirmed the low toxicit…
