ChemInform Abstract: Tandem Nucleophilic Addition-Intramolecular Aza-Michael Reaction: Facile Synthesis of Chiral Fluorinated Isoindolines.
2011
A highly stereoselective synthesis of fluorinated 1,3-disubstituted isoindolines is described. To this end, a tandem reaction consisting of a diastereoselective addition of fluorinated nucleophiles to Ellman’s N-(tert-butanesulfinyl)imines followed by an intramolecular aza-Michael reaction has been developed. This strategy allows for the construction of isoindolines bearing several degrees of fluorination (mono-, di-, or trifluoromethyl as well as heavier fluorinated groups). In the majority of all cases, the products are formed as single isomers.
Stereoselective Synthesis of 3-Substituted and 3,4-Disubstituted Piperidine und Piperidin-2-one Derivatives
2012
The stereoselective synthesis of 3-substituted and 3,4-disubstituted piperidine and piperidin-2-one derivatives was achieved starting from 2-pyridone. After N-galactosylation and subsequent O-silylation, nucleophilic addition of organometallic reagents proceeded with high regio- and stereoselectivity at 4-position. Substituents at position 3 were stereoselectively introduced by reaction of electrophiles with amide enolates of the N-galactosyl-2-piperidones.
An Unexpected Three-Component Condensation Leading to Amino(3-oxo-2,3-dihydro-1H-isoindol-1-ylidene)acetonitriles.
2005
The reaction of 2-carboxybenzaldehyde with primary amines in the presence of cyanide leads to the formation of 2-substituted amino(3-oxo-2,3-dihydro-1H-isoindol-1-ylidene)acetonitriles. These compounds originate from the condensation of 2-carboxybenzaldehyde with the amine and two molecules of hydrogen cyanide and represent a novel class of isoindolinones.
STUDIEN ZUM VORGANG DER WASSERSTOFFÜBERTRAGUNG 611
1982
Abstract In arylsulfonyl halides, the half-wave potentials of the corresponding chlorides and fluorides differ by more than 1000 mV, the fluoride being more negative; the influence of para-substituents is small for the chlorides, large for the fluorides. In agreement with the half-wave potentials, arylsulfonyl chlorides are considerably more reactive chemically than the corresponding fluorides. The O-selectivity found for P(O)F compounds is not observed in arylsulfonyl fluorides. Studies of competitive ester formation using primary and secondary alcohols and various arylsulfonyl chlorides yielded no clear analogy to the half-wave potentials. The primary alcohol is always sulfonated in prefe…
ChemInform Abstract: Preparation of N,O-Aminals as Synthetic Equivalents of H2C=NAr and (H2C=NHAr)+ Ions: Neutral- and Acid-Promoted Transformations.
1988
A general method for the synthesis of N,O-aminals derived from primary aromatic amines is described. The reactivity of these compounds under neutral and acidic conditions has been studied and the title compounds can be envisaged as general purpose H2CNAr or (H2CNHAr)+ equivalents. N,O-Aminals have been converted into perhydrotriazines by moderate heating and into bis(4-aminoaryl)methane derivatives or N-benzylarylamines, respectively when heated in acidic media with pH control. Reduction of N,O-acetals with sodium cyanoborohydride has revealed that the C–O bond is broken exclusively in acidic media.
Unprecedented intermolecular transamidation reaction of N-carbamoylmethyl-N'-tosylguanidines.
2003
[reaction: see text] N-Carbamoylmethyl-N'-tosyl guanidine 2 reacts easily with primary alkylamines to afford substituted carboxamides 3. The reaction proceeds via a five-membered-ring intermediate 5, which could be isolated, and features a rare example of an intermolecular transamidation reaction under mild conditions.
ChemInform Abstract: Addition of Nucleophiles to Fluorinated Michael Acceptors.
2016
A series of nucleophiles, including primary and secondary amines, primary alcohols, and thiols, as well as diethyl malonate and nitromethane, were added to different fluorinated Michael acceptors including 2-fluoroalk-1-en-3-ones and 2-fluoro-1-phenylprop-2-en-1-one. The resulting β-substituted α-fluoro ketones were isolated in 34–92 % yield, depending on the substrate and the nucleophile. The best yields were obtained with secondary amines and with p-methylthiophenol.
Preparation of 5‐bromo‐6‐phenylimidazo[2,1‐b][1,3,4]thiadiazol‐2‐ylamines
2008
The reaction of primary or secondary amines with 2,5-dibromo-6-phenylimidazo[2,1-b][1,3,4]-thiadiazole (5) leads to a chemoselective replacement of the 2-Br substituent. The process represents a convenient route to the corresponding 2-ylamines 7a-d. Hydrazine reacts in an analogous fashion (5 → 7e). The structure determinations are based on an X-ray crystal structure analysis and on one- and two-dimensional NMR measurements.
