Respiration and low cAMP-dependent protein kinase activity are required for high-level expression of the peroxisomal thiolase gene in Saccharomyces c…

1996

Transcription of genes for peroxisomal proteins is repressed by glucose and induced by oleate. At least for the peroxisomal thiolase gene (POT1) there is a third regulatory mechanism, mediated by the transcription factor Adr1p, which is responsible for the high-level expression of the gene in stationary phase. Here we show that a region in the POT1 promoter that extends from positions -238 to -152 mediates this mechanism, and we suggest that Adr1p acts indirectly on POT1. We have also analyzed the role of the cAMP-dependent protein kinase (PKA) in the transcriptional regulation of POT1. PKA exerts a negative control: the high, unregulated PKA activity in a bcy1 mutant maintains POT1 transcr…

Rigidity and Dynamics of Random Spring Networks

1996

The static and dynamic elastic properties of two-dimensional random networks composed of Hookean springs are analyzed. These networks are proved to be nonrigid with respect to small deformations, and the floppy mode ratio is calculated exactly. The vibrational spectrum is shown to consist only of zero-frequency and localized modes. The exponential decay of the amplitude and velocity of the transient wave front are shown to be exactly described by a quasi-one-dimensional model of noninteracting paths of propagation.

Modeling urban growth by cellular automata

1996

International audience; The structural development of human settlements can be characterized as a complex highly feedbacketed process. The assumption that this process is governed by rather few fundamental laws stimulated a considerable research in the field of urban growth during the last decade. Aiming at the comprehension of the basic underlying dynamics different approaches from the field of self-organizing systems have been proposed. In this paper we present a "cellular model" of urban growth dynamics based on the work of White, Engelen and Uljee. As a starting point this model throws some light on the mechanism of urban growth. But even more important, it raises a lot of questions con…

Different effects of interferons, interleukin‐1β and tumor necrosis factor‐α in normal (OSE) and malignant human ovarian epithelial cells

1996

Ovarian cancer arises mostly from the ovarian surface epithelium. The aim of our study was to compare the effects of cytokines in ovarian surface epithelial (OSE) cells and in ovarian carcinoma cells. Proliferation and expression of surface antigens (CA-125 and classes I and II antigens of the major histocompatibility complex [MHC]) were measured in OSE cells obtained from 7 different patients and 7 ovarian carcinoma cell lines. Proliferation of OSE cells remained unaffected by interferon (IFN)-α or IFN-γ, whereas interleukin-I (IL-I) and tumor necrosis factor (TNF) increased cell growth. Proliferation of ovarian carcinoma cells was reduced by both types of IFN as well as TNF but was not af…

Small-angle X-ray scattering reveals differences between the quaternary structures of oxygenated and deoxygenated tarantula hemocyanin

1996

Small-angle X-ray scattering (SAXS) curves have been recorded for the oxygenated and deoxygenated states of the 4 x 6-meric hemocyanin from the tarantula Eurypelma californicum. A comparison of the curves shows that the quaternary structures of the two states are different by three criteria, which all indicate that the hemocyanin is less compact in the oxygenated compared to the deoxygenated form: (a) The radius of gyration is 8.65 +/- 0.05 nm for the deoxy- and 8.80 +/- 0.05 nm for the oxy-form. (b) The maximum particle dimension amounts to 25.0 +/- 0.5 nm for the deoxy- and to 27.0 +/- 0.5 nm for the oxy-form. (c) A dip in the intramolecular distance distribution function p(r) is more pro…

Spermatocytes of the caddisfly Potamophylax rotundipennis (Trichoptera, Insecta): a fine structure study with emphasis on synaptonemal complex plates…

1996

Abstract Electron microscopy of ultrathin sections was used to study the restructuring of primary spermatocytes in a caddisfly, Potamophylax rotundipennis (Limnephilidae). Spindle structure was also examined using light microscopy of dividing spermatocytes lysed in a microtubule-stabilizing buffer. The bulk of pachytene spermatocytes was usual in that the nuclei contained tripartite synaptonemal complexes (SCs). The SCs were attached end-on to the inner face of the nuclear envelope and loosely surrounded by electron-dense chromatin. Cells of this type gave rise to late prophase I spermatocytes, where SCs were missing and chromatin condensation was advanced. By metaphase I, a conventional bi…

Forward-AngleH3e(e,e′π±)Coincident Electroproduction and the Search forΔ's in the Ground State ofH3e

1996

Forward-angle coincident electroproduction cross sections of charged pions from ${}^{3}\mathrm{He}$ have been measured at electron energies ${E}_{0}\phantom{\rule{0ex}{0ex}}=\phantom{\rule{0ex}{0ex}}855$, 675, 600, and 555 MeV. The overall features of the data for energy transfers of $\ensuremath{\omega}\phantom{\rule{0ex}{0ex}}=\phantom{\rule{0ex}{0ex}}370$ to 430 MeV with pions detected along the momentum transfer axis are reproduced in terms of a microscopic model, including pole terms, final state rescattering and produced and preformed $\ensuremath{\Delta}$ resonances. Separation of the cross section into its longitudinal and transverse parts was performed at ${Q}^{2}\phantom{\rule{0ex…

Treatment of advanced adenocarcinomas of the exocrine pancreas and the gallbladder with 5-fluorouracil, high dose levofolinic acid and oral hydroxyur…

1996

BACKGROUND To date there is no established chemotherapeutic treatment for patients with unresectable locally advanced and/or metastatic carcinomas of the exocrine pancreas or the gallbladder. A multicenter Phase II trial has been performed by the Southern Italy Oncology Group with the aim of evaluating the clinical effectiveness and tolerability of weekly 5-fluorouracil (5-FU) in modulation with intravenous (i.v.) high dose levofolinic acid and oral hydroxyurea. METHODS A total of 70 patients fulfilling the standard eligibility for a Phase II study were enrolled in the trial. Forty patients had advanced pancreatic adenocarcinoma and 30 had advanced gallbladder carcinoma. The treatment sched…

Electron-phonon heat transport in arrays of Al islands with submicrometer-sized tunnel junctions.

1996

We present experimental evidence of the effect of electrode volume and its shape on thermalization of small metallic islands for single electron tunneling. We have investigated the power law and the magnitude of the thermal transport and found that it obeys the common ${T}^{5}$ law for electron-phonon coupling only for the smallest islands studied, and in other cases considered, with cooling fins attached to the islands, the coupling per unit volume is weaker and it rather follows a law $\ensuremath{\propto}{T}^{p}$, where $pl5$. We attribute this to local hot electrons adjacent to the tunnel junctions.

Restoration of Clostridium difficile toxin-B-inhibited phospholipase D by phosphatidylinositol 4,5-bisphosphate.

1996

Receptor signalling to phospholipase D (PLD) in human embryonic kidney (HEK) cells stably expressing the m3 muscarinic acetylcholine receptor apparently involves Rho proteins. Since phosphatidylinositol 4,5-bisphosphate [PtdIns(4,5)P2] has been recognized as an essential cofactor for PLD activity and since activated Rho proteins have been reported to stimulate the synthesis of PtdIns(4,5)P2, we studied whether in HEK cells PLD activity is regulated by PtdIns(4,5)P2 and, in particular, whether PtdIns(4,5)P2 can restore PLD activity inhibited by Clostridium difficile toxin B, which inactivates Rho proteins. Addition of MgATP to permeabilized HEK cells increased basal PLD activity and potentia…

Geochemistry of gases and waters discharged by the mud volcanoes at Paternò, Mt. Etna (Italy)

1996

Approximately 20 km south of Mt. Etna craters, at the contact between volcanic and sedimentary formations, three mud volcanoes discharge CO2-rich gases and Na–Cl brines. The compositions of gas and liquid phases indicate that they are fed by a hydrothermal system for which temperatures of 100–150 °C were estimated by means of both gas and solute geothermometry. The hydrothermal system may be associated with CO2-rich groundwaters over a large area extending from the central part of Etna to the mud volcanoes. Numerous data on the He, CH4, CO2 composition of the gases of the three manifestations, sampled over the past 5 years, indicate clearly that variations are due to separation processes of…

Down-regulation of nuclear binding activities of OXBOX-REBOX transcription factors during cellular senescence.

1996

Functional capacity of mitochondria declines during aging and this impairment may have a major role in aging process. Several observations indicate that transcriptional efficiency is reduced during aging. Our purpose was to find out whether aging and cellular senescence affect the nuclear binding activities of transcription factors which bind to OXBOX-REBOX sequence present in promoter regions of numerous nuclear genes encoding mitochondrial proteins. These factors regulate and coordinate the expression of mitochondrial proteins. We observed a strong down-regulation in the nuclear binding activities of OXBOX-REBOX factors in replicatively senesced human WI-38 and IMR-90 fibroblasts. On the …

Perturbations of spacetime: gauge transformations and gauge invariance at second order and beyond

1996

We consider in detail the problem of gauge dependence that exists in relativistic perturbation theory, going beyond the linear approximation and treating second and higher order perturbations. We first derive some mathematical results concerning the Taylor expansion of tensor fields under the action of one-parameter families (not necessarily groups) of diffeomorphisms. Second, we define gauge invariance to an arbitrary order $n$. Finally, we give a generating formula for the gauge transformation to an arbitrary order and explicit rules to second and third order. This formalism can be used in any field of applied general relativity, such as cosmological and black hole perturbations, as well …
