Two-dimensional 1 H NMR spectra of ferricytochrome c 551 from Pseudomonas aeruginosa
1993
AbstractThe full assignment of 1H NMR signals of heme proton resonances of ferricytochrome c551 from Pseudomonas aeruginosa has been performed by means of 2D NMR experiments. This technique allows the complete and unequivocal assignment of all heme resonances, including methylene resonances of the propionic groups, directly implicated in the pH dependence of the redox properties of cytochrome c551.
Physostigmine and Neuromuscular Transmission
1993
Single channel studies carried out in cultured rat myoballs and cultured hippocampal neurons, and ion flux studies performed on Torpedo electrocyte membrane vesicles, showed that physostigmine (Phy), a well-established acetylcholinesterase inhibitor, interacts directly with nicotinic acetylcholine receptors (nAChR). Low concentrations (0.1 microM) of Phy activate the receptor integral channel, whereas higher concentrations blocked the channel in its opened state. In contrast to channel activation by acetylcholine (ACh) and classical cholinergic agonists, however, Phy was capable of activating the nAChR channel even when the ACh binding sites were blocked by competitive antagonists, such as …
Localization and mRNA steady-state level of cellular fibronectin in rat liver undergoing a CCl4-induced acute damage or fibrosis.
1993
Abstract In an attempt to investigate cellular fibronectin synthesis and deposition during acute liver damage and fibrogenesis, we used the presence of the additional type III-related ED-A domain of cellular fibronectin as a characteristic for distinguishing it from the plasma form. Using site-specific antibodies, we localized cellular fibronectin deposition in the necrotic pericentral areas of acutely damaged liver tissue after a single CCl 4 -gavage, whereas in control liver only trace amounts of cellular fibronectin were detectable along the sinusoids. Upon several CCl 4 -administrations leading to liver fibrosis, cellular fibronectin deposits were accumulated in the fibrotic septa. Nort…
Lysis of human pancreatic adenocarcinoma cells by autologous HLA-class I-restricted cytolytic T-lymphocyte (CTL) clones.
1993
From the primary site of a pancreatic adenocarcinoma (patient BE) a permanent cell line (MZ-PC-2) was established in tissue culture. In the course of mixed lymphocyte-tumor-cell cultures (MLTC) with autologous blood-derived lymphocytes, we isolated CTL clones that lysed autologous tumor cells but not autologous EBV-transformed B cells (EBV-B) and not K562. Pre-treatment of MZ-PC-2 cells with IFN-gamma was required to obtain significant lysis in 4-hr cytotoxicity assays. IFN-gamma was superior to IFN-alpha in that respect. Among MLTC responder lymphocytes, tumor-reactive CTL proliferated more strongly in response to MZ-PC-2 cells treated with IFN-gamma than to untreated tumor cells. Three CT…
Molecular structures of chlorophyll a aggregates: spectroscopic and molecular modeling study
1993
Molecular structures of chlorophyll a aggregates have been studied. Spectroscopic properties of these aggregates have been studied by absorption and fluorescence spectroscopy in hydrocarbon solution at various temperatures. Observed spectroscopic shifts were interpreted in terms of simple exciton theory. Exciton splittings were estimated from computer optimized models of previously suggested Chl a aggregate structures.
Locally Frozen Defects in Random Sequential Adsorption with Diffusional Relaxation
1993
Random sequential adsorption with diffusional relaxation, of two by two square objects on the two-dimensional square lattice is studied by Monte Carlo computer simulation. Asymptotically for large lattice sizes, diffusional relaxation allows the deposition process to reach full coverage. The coverage approaches the full occupation value, 1, as a power-law with convergence exponent near 1/2. For a periodic lattice of finite (even) size $L$, the final state is a frozen random rectangular grid of domain walls connecting single-site defects. The domain sizes saturate at L**0.8. Prior to saturation, i.e., asymptotically for infinite lattice, the domain growth is power-law with growth exponent ne…

The Phenomenon of a High Triglyceride Response to an Oral Lipid Load in Healthy Subjects and Its Link to the Metabolic Syndrome
1993
Excessive postprandial triglyceride (TG) responses despite normal fasting TG levels have been described in single cases within small groups of healthy subjects and in patients with obesity or precocious atherosclerosis, known to be associated with high insulin fasting levels. To clarify this association, fasting and postprandial TG and insulin levels were studied in 113 healthy young (25.7 +/- 2.6 years), normal weight (body mass index 20.8 +/- 2.3 kg/m2) male subjects who were selected from among 117 subjects on the basis of TG fasting levels200 mg/dl. After a 12-hour fast a standardized liquid lipid load was administered containing 58 g mainly saturated fat and 1,017 kcal energy. Both fas…

Altered pore-forming properties of proteolytically nicked staphylococcal alpha-toxin
1993
Staphylococcal alpha-toxin is a single-chain polypeptide with a molecular weight of 34,000 that hexamerizes in lipid bilayers to form pores of 1-1.5 nm effective diameter in membranes. We demonstrate that limited proteolysis of purified alpha-toxin with proteinase K generates a hemolytically active product that yields one major protein band of 17-18 kDa in SDS-polyacrylamide gel electrophoresis. The 17-18-kDa protein band harbors two major fragments of similar size representing the N- and C-terminal halves, which remain associated with each other in non-denaturing buffers but dissociate in 6 M urea. Dissociation in urea leads to loss of hemolytic activity. In contrast, unnicked alpha-toxin …
Vesamicol blocks the recovery, by recycling cholinergic electromotor synaptic vesicles, of the biophysical characteristics of the reserve population.
1993
The effect of vesamicol on the ability of recycling cholinergic synaptic vesicles to recover, during a period of post-stimulation rest, the biophysical properties of the reserve pool was studied in prestimulated perfused blocks of the electric organ of the electric ray, Torpedo marmorata, a tissue rich in cholinergic synapses. The effect of the drug was analysed by high-resolution centrifugal density-gradient fractionation in a zonal rotor of the extracted vesicles. The two vesicle fractions were identified by their ATP and acetylcholine content and the recycled vesicles by their acquisition of [3H]acetylcholine derived from [3H]acetate in the perfusate. Vesamicol (10 microM) blocked the up…
N-string vertices in string field theory.
1993
We give the general form of the vertex corresponding to the interaction of an arbitrary number of strings. The technique employed relies on the ``comma" representation of String Field Theory where string fields and interactions are represented as matrices and operations between them such as multiplication and trace. The general formulation presented here shows that the interaction vertex of N strings, for any arbitrary N, is given as a function of particular combinations of matrices corresponding to the change of representation between the full string and the half string degrees of freedom.
The approximate subdifferential of composite functions
1993
This paper deals with the approximate subdifferential chain rule in a Banach space. It establishes specific results when the real-valued function is locally Lipschitzian and the mapping is strongly compactly Lipschitzian.
Deterministic chaos and the first positive Lyapunov exponent: a nonlinear analysis of the human electroencephalogram during sleep
1993
Under selected conditions, nonlinear dynamical systems, which can be described by deterministic models, are able to generate so-called deterministic chaos. In this case the dynamics show a sensitive dependence on initial conditions, which means that different states of a system, being arbitrarily close initially, will become macroscopically separated for sufficiently long times. In this sense, the unpredictability of the EEG might be a basic phenomenon of its chaotic character. Recent investigations of the dimensionality of EEG attractors in phase space have led to the assumption that the EEG can be regarded as a deterministic process which should not be mistaken for simple noise. The calcu…

Muscle strength and serum testosterone, cortisol and SHBG concentrations in middle-aged and elderly men and women.
1993
Forty healthy males (M) and females (F) divided into two different age groups i.e. M50 years (range 44–57; n= 9), F50 years (range 43–54; n= 9), M70 years (range 64–73; n= 11) and F70 years (range 63–73; n= 11) volunteered as subjects for examination of muscle cross-sectional area (CSA) and maximal voluntary isometric force production characteristics of the leg extensor muscles and serum androgen and sex hormone binding globulin (SHBG) concentrations. The CSA in the male groups was greatly larger (P < 0.01) than in the female groups and both elderly groups demonstrated slightly (n.s.) smaller values in the CSA than the two middle-aged groups. Maximal force of 2854 ± 452 N in M50 was greater…

TheA 1-and the ϕ-meson in QCD
1993
Recently we used a method of stable analytic extrapolation to test whether strictly local duality between the asymptotic and the resonance region, which is believed to be valid in QCD, appears already at the present stage of QCD calculations. Probing the QCD ϱ-amplitude, we found a prominent bump structure in the resonance region, i.e. a rather direct evidence for the validity of local duality. In the present paper we confirm and extend this result by giving two further applications, theA1-meson and the ϕ-meson. While we do not meet theA1, as was to be expected since other sum rules can not separate it from the continuum, too, the extrapolation of the ϕ-amplitude leads to an enormous bump. …
The human gene encoding cytokeratin 20 and its expression during fetal development and in gastrointestinal carcinomas
1993
The differentiation of the predominant cell types of the mucosal epithelium of the mammalian gastrointestinal tract is characterized by increasing amounts of an intermediate-sized filament (IF) protein designated cytokeratin (CK) 20 which is a major cellular protein of mature enterocytes and goblet cells. Here we report the isolation of the human gene encoding CK 20, its complete nucleotide sequence and the amino acid sequence deduced therefrom that identifies this polypeptide (mol. wt. 48553) as a member of the type I-CK subfamily. Remarkable, however, is the comparably great sequence divergence of CK 20 from all other known type I-CKs, with only 58% identical amino acids in the conserved …
Application Of Microholography for Ground-based In Situ Measurements in Stratus Cloud Layers: A Case Study
1993
Abstract A holographic droplet and aerosol recording system (HODAR) has been designed and operated in situ in a low-level stratus cloud layer adopting the Fraunhofer in-line technique to measure sizes and velocity vectors of cloud droplets as well as to determine sizes and shapes of other hydrometeors (e.g., graupel). The particle-size radii covered by this ground-based instrument range from three to several hundred micrometers. A case study conducted on the Kleiner Feldberg Mountain Observatory near Frankfurt, Germany, during November 1990 documents the temporal evolution of the cloud-droplet size distribution over a period of 19 h during which the size distribution changed from monomodal …
Search for contact interactions in the reactionse + e −→l + l − ande + e −→γγ
1993
Contact interactions are searched for using the differential cross sections for the reactions e + e -→ e + e -, e + e -→ µ + µ -, e + e -→ τ + τ - and e + e -→γγ measured at 12 energies around the Z peak and corresponding to about 20 pb-1 of cumulated luminosity. Four-fermion contact term models assuming various chiralities of lepton currents are fitted to the lepton data and lower limits on the energy scale Λ of such terms are set at 95% c.l. The limits vary in the range 0.9 4.7 TeV, depending on the model and on the lepton flavour. The eeγγ contact terms are searched for assuming various chiralities. Limits on the energy scale Λ between 79 and 130 GeV are extracted from the data. The resu…
