Transglutaminase and polyamine dependence of effector functions of human immunocompetent cells

1982

AbstractThe effects of the transglutaminase inhibitor dansyl cadaverine (DC) and the polyamine antagonist methyl glyoxal-bis-(guanylhydrazone) (MeGbG) on the response of lymphocytes towards allogeneic and lectin stimulation and on the zymosan-induced chemiluminescence of neutrophilic granulocytes was studied. Application of DC resulted in dose-dependent suppression of chemiluminiscence and lymphocyte proliferation; no difference of inhibitory potential occurred with variation of incubation time in the latter system. MeGbG was inactive in granulocytes, but inhibited lymphocyte proliferation; its effect increased with time. The experiments provide further evidence for the importance of transg…

Influence of Collective Surface Motion on the Threshold Behavior of Nuclear Fusion

1982

Fusion excitation functions for the systems $^{40}\mathrm{Ar}$+$^{112,116,122}\mathrm{Sn}$ and $^{40}\mathrm{Ar}$+$^{144,148,154}\mathrm{Sm}$ have been determined, covering cross sections ranging from several hundred millibarns down to the microbarn level. The data are interpreted with a fusion model that includes fluctuations of the barrier with an amplitude that is shown to be correlated with the collective surface properties of the nuclei. There is no need to assume an additional enhanced tunneling process.

Influence of enol acetylation on the photo-fries rearrangement of anortho-acylaryl benzoate

1982

The enol acetate 1b undergoes benzoyl-oxygen bond rupture upon irradiation, giving a mixture of benzophenones 4 and 5. The primary photo-Fries products 2 and 3 could not be isolated, due to a rapid intramolecular transacetylation. The acetophenone 6 rearranges to give the benzophenones 7 and 8 under the same conditions, but the yields are significantly lower than in the former case. Compounds 7 and 8 are separately correlated with 4 and 5 by treatment with acetic anhydride in the presence of pyridine. D-penicillamine in Wilson's disease presenting as acute liver failure with hemolysis.

1982

Wilson's disease in a young woman presenting with an acute course is described. The clinical manifestations were fulminant hepatic failure associated with marked intravascular hemolysis. Immediate D-penicillamine and high-dose steroid therapy did not influence the course of the disease. Necropsy revealed an increased hepatic copper content and cirrhosis with extensive necrosis of the liver.

Die oligomerisierung endständiger hydroxyacetylene mit rhodiumkomplexkatalysatoren

1982

The substrate and ligand influence on oligomerisation of terminal hydroxyacetylenes catalysed by rhodium complexes has been studied. In this reaction α- and β-hydroxyacetylenes were compared with each other and with an unsubstituted terminal alkyne. The concentration dependant ligand control with Ph3As and Ph3P has been compared and different ligand association behaviour is found, because only with Ph3As as ligand at intermediate concentrations cyclic trimers are selectively formed. Some brief experiments with different p- and o-substituted triphenylarsines show electronic and marked steric influences on the oligomerisation of 3-methylbutyne-1-ol-3.

Fine structure of a mandibular stretch receptor in Thrips physapus L. (Thysanoptera, Insecta)

1982

Only one non-ciliary proprioceptor is developed on the mandible of Thrips. It consists of two bipolar, multiterminal sense cells, the dendrites of which form a strand extended between the tentorium and the back mandibular margin. The dendritic terminals are embedded in an electron-dense, homogeneous matrix, which obviously represents the stimulus transmitting structure. The strand is stretched, if the mandible is moved forwards and upwards. In Thrips, the system of proprioceptors monitoring the mandibular movement is reduced extensively compared with other pterygote insects. This is linked up with the far-reaching reduction of the mobility of the mandible itself.

Studies on vinblastine-induced autophagocytosis in mouse liver. III. A quantitative study.

1982

The microtubule inhibitor vinblastine (25 mg/kg, i.p.) induces autophagocytosis in mouse hepatocytes. The formation of autophagic vacuoles, their contents, and other cellular changes after vinblastine injection in hepatocytes, were studied by light and electron microscopic morphometric analysis. The volume density of autophagic vacuoles increased significantly during the experimental period (24 h). This increase was due to the significant increase in their number, which was approximately 5-fold 4 h, 12 h and 24 h after vinblastine injection. The mean volume of the autophagic vacuoles increased significantly 1 h after vinblastine injection, at which time the formation of new autophagic vacuo…

Modulation of the nuclear-envelope nucleoside triphosphatase by poly(A)-rich mRNA and by microtubule protein.

1982

Nuclear envelopes contain a nucleoside triphosphatase which is thought to be involved in the supply of energy for nucleo-cytoplasmic RNA transport. This enzyme is stimulated most efficiently by poly(A) and to a lesser extent by poly(G) and poly(dT). Half-maximal stimulation of the enzyme from rat liver nuclei, which was associated with the poly(A)-specific endoribonuclease IV and was free from poly(A) polymerase and endoribonuclease V activity, was determined to occur at a concentration of 1.1 × 106 poly(A) molecules/nuclear ghost. Double-reciprocal plot analyses revealed a 2.8-fold stimulation of the enzyme by poly(A). Poly(A) in the hybrid form had no influence on the activity of the nucl…

1982

A silica of 14 nm pore size was reacted according to two procedures with the following silanes: γ-aminopropyltriethoxysilane (1), N-aminoethyl-N′-aminopropyltrimethoxysilane (2), N-aminoethyl-N′-aminoethyl-N″-aminopropyltrimethoxysilane (3), N-(3-triethoxysilanepropyl)-N, N-diacetic dimethylester (4), N-(3-trisodiumsilanolatepropyl)-N, N-diacetic acid disodium salt (5) and N-(3-trisodiumsilanolatepropyl)-ethylenediamine-N, N′, N′-triacetic acid trisodium salt (6). The reaction of silanes 1–4 with the silica was carried out under anhydrous conditions (procedure A). Silanes 1–6 were subjected to surface modification under essentially hydrous conditions applying a two-step procedure (procedure…

A method for fitting multi-component decay curves

1982

Abstract A generalization of a non-iterative method recently proposed by Mukoyama for the fitting of two-component decay curve is presented. Two modifications of the procedure are also suggested, by which the influence of the statistical fluctuations of the data may be reduced. Results of fairly good quality are obtained also for three- and, to a lesser extent, for four-component decay curves.

Cell fusion as a mechanism for the formation of giant cells (Langhans’ type)

1982

The formation of multinuclear giant cells of the Langhans' type in tubulo-interstitial auto-immune nephritis in the rat has been investigated by means of autoradiography. While in the majority of giant cells all nuclei were radiolabeled, in a few both labeled and unlabeled nuclei were present. This latter finding represents strong evidence in favour of the hypothesis that giant cells do not form by endomitotic processes but rather through fusion of certain precursor cells. According to previous studies this precursor cell population consists mainly of epitheloid cells, i.e. modified monocytes.

Store and recoil of elastic energy in slow and fast types of human skeletal muscles.

1982

Stretch-shortening cycle refers to the mechanical condition in which store and recoil of elastic energy occur in the skeletal muscle. This leads to a greater work output when compared to a simple shortening contraction. The subjects performed vertical jumps with and without preliminary counter-movement and with small and large knee angular displacement. The results indicated that those subjects who had more fast twitch (FT) fibers benefited more from the stretching phase performed with high speed and short angular displacement. The amounts of elastic energy stored in this phase were 30 and 26 N X kgBW-1, respectively, for FT and slow twitch (ST) type subjects. The recoil of elastic energy w…

Electron spin polarization in the photoemission of NEA GaAs1?x P x

1982

Negative electron affinity GaAs1−xPx-photocathodes emit spin-polarized electrons if irradiated with circularly polarized light. The spectra of spin polarization of photoelectrons emitted from crystals with different phosphorus contentx resemble each other in shape but shift to shorter wavelengths with increasingx. Polarization values up to 40% are observed for electrons from crystals withx=0 andx=0.38. Cathodes with higher phosphorus contents increasing fromx=0.65 tox=0.87 and tox = 1.0 deliver photoelectrons with decreasing degrees of spin polarization of 17%, 15%, and 9%, respectively, at maximum.

Alterations in the spontaneous activity of cells in the guinea pig pineal gland and visual system produced by pineal indoles

1982

The indoles serotonin (SER), melatonin (MEL), 5-methoxytryptophol (5-MTL) and 5-hydroxytryptophol (5-HTL) were administered during daytime microelectrophoretically to 240 cells in the pineal gland of the guniea-pig. The action of SER and 5-HTL was predominantly depressant on the electrical activity, MEL and 5-MTL caused an excitation in most of the units. Although MEL and 5-MTL caused fairly similar reactions on average, they appear to act on different cells. The effects of microelectrophoretically applied MEL and 5-MTL on the spontaneous or evoked activity in the visual system (retinal ganglion cells, optic tract, lateral lateral geniculate body, superior colliculus) of the guinea-pig were…

Ovicidal Activity of Maternally Applied Ecdysteroids in the Large Milkweed Bug (Heteroptera: Lygaeidae)

1982

Exogenous ecdysteroids (ecdysone, 20-hydroxyecdysone and makisterone A) administered to reproducing females of Oncopelus fasciatus (Dallas) reduced egg hatch drastically. The ovicidal activity apparently stemmed from ecdysteroids—unchanged hormone or metabolites—transferred from mother into oocyte. Different stages of embryogenesis were affected. Makisterone A, the predominant ecdysteroid in normal embryos, is by far the most effective ecdysteroid.

Coherence in Spoken and Written Discourse

1982

The article discusses two types of discourse coherence, propositional coherence and interactional coherence. These two types are related to the mode of discourse as well as to the level of style in the discourse. It takes up the concept of frame as an organised subsystem of knowledge shared by human beings, and suggests ways in which this concept could be used in studies of coherence in everyday conversations, prose texts and lyrical poetry. The article also suggests a way of describing discourse coherence in terms of the discourse topic and considers the relationship of the two types in different kinds of texts.

Oxygenation of malignant tumors after localized microwave hyperthermia

1982

The oxyhemoglobin saturation (HbO2) of single red blood cells within tumor microvessels (diameter: 3-12 micrometers) of DS-Carcinosarcoma was studied using a cryophotometric micromethod. In untreated control tumors (mean tissue temperature approx. 35 degrees C) the measured values scattered over the whole saturation range from zero to 100 sat. %, the mean being 51 sat. %. Upon heating at 40 degrees C for 30 min, the oxygenation of the tumor tissue significantly improved as compared with control conditions. After 40 degrees C-hyperthermia a mean oxyhemoglobin saturation of 66 sat. % was obtained. In contradistinction to this, after 43 degrees C-hyperthermia the tumor oxygenation was signific…

Carbohydrate binding specificity and purification by biospecific affinity chromatography of Ascidiamalaca traust. Hemagglutinins

1982

The carbohydrate specificities of Ascidia malaca serum hemagglutinins were determined by hemagglutination inhibition tests. Analysis of agglutinins against rabbit and human A, B, O erythrocytes suggests that the size of the combining site corresponds to a disaccharide with a specificity for saccharides containing a D-galacto configuration (D-melibiose, D-raffinose, D-galactose, alpha-lactose, lactulose, L-arabinose). No anomeric specificity was observed with oligosaccharides. Hydroxyl groups probably involved in hydrogen-bond formation with agglutinin binding site, were identified as carbons C2, C4, C5 and C6 of D-galactose. Absorption experiments showed that two distinct agglutinins with s…
