Stability of ascorbic acid in adapted milk-based infant formulae during storage

A study was made of the effects of storage (for up to 17 months at 22 or 37 °C) on the ascorbic acid contents of four adapted milk-based infant formulae with the same composition except for the iron salt added (lactate or sulphate) and/or the vitamin E source involved (α-tocopherol (α-T) or α-tocopherol acetate (α-TAc)). Ascorbic acid was measured by a voltammetric method. The ascorbic acid contents of the formulae ranged from 0.77–0.84 g kg−1 immediately after manufacture to 0.41–0.48 g kg−1 after 17 months of storage. Isocratic high-performance liquid chromatographic determination of tryptophan in infant formulas.

The application to infant formulas of a method for tryptophan determination by isocratic HPLC with UV detection at 254 nm, after derivatization with phenyl isothiocyanate, was studied. Protein was hydrolysed by barium hydroxide at 120 degrees C for 8 h, followed by derivatization with phenyl isothiocyanate, HPLC and UV detection at 254 nm. The optimum chromatographic conditions (pH, ionic strength of elution solvent and eluent ratio) were established. Sterol Composition in Infant Formulas and Estimated Intake.

Sterol contents in infant formulas (IFs) from the European market were determined, and their intakes by infants between 0 and 6 months were evaluated. Total animal sterols (mg/100 mL) ranged from 1.71 to 5.46, cholesterol being the main animal sterol (1.46-5.1). In general, cholesterol and desmosterol were lower than the human milk (HM) values indicated by other authors. Total plant sterol (mg/100 mL) ranged from 3.1 to 5.0. β-Sitosterol, the most abundant phytosterol, ranged from 1.82 to 3.01, followed by campesterol (0.72-1.15), stigmasterol (0.27-0.53), and brassicasterol (0.14-0.28). Copper, iron and zinc determinations in human milk using FAAS with microwave digestion

Abstract A method for determining copper, iron and zinc in human milk was optimized and validated. It includes microwave mineralization of the sample and measuring the elements by flame atomic absorption spectroscopy (FAAS). Only 2 ml of milk is needed, and the method is free of matrix interferences. The effect of enriching milk-based beverages with plant sterols or stanols on the fatty acid composition of the products

Five plant sterol (PS)-enriched dairy products from the Spanish market were characterised for fatty acids (FA), volatile compounds and lipid stability. The ingredients used for PS enrichment by the food industry may come from different sources, thus influencing the composition and stability of the lipid fraction. In this study, the FA profile proved highly variable among samples, not agreeing with the nutritional labelling of the products. The volatile profile was characterised. Eight fruit beverages containing grape, orange and apricot, with/without iron and/or zinc and with/without milk added were analyzed for total antioxidant capacity (ORAC and TEAC methods), ascorbic acid content, and total polyphenols. The influence of cold storage (2-4 degrees C) during the product shelf-life (135 days) and antioxidant capacity after an in vitro gastrointestinal digestion were evaluated. Stability of fatty acids and tocopherols during cold storage of human milk

The stability of fatty acids and tocopherols from human milk from 8 healthy lactating mothers was determined in fresh milk and after cold storage. Refrigeration at 4 °C for 48 h or freezing at −20 °C for 30 days did not significantly decrease fatty acid contents (mg per 100 g human milk), or concentrations of α-, β-, γ- and δ-tocopherol isomers, compared with fresh milk. Results obtained in the present study showed that cold storage under the usual storage conditions in neonatal units, at home or in milk banks did not have a detrimental effect upon fatty acids and tocopherols contents in human milk. Addition of milk fat globule membrane as an ingredient of infant formulas for resembling the polar lipids of human milk

Polar lipid (PL) contents in human milk (HM) from two different geographic zones in Spain (central and coastal) were determined. These PLs were also analysed in several infant formulas (IFs), three of which contained milk fat globule membrane (MFGM), an ingredient used to resemble the PL profile of HM. Whole blood selenium content in pregnant women

The selenium content in the blood of pregnant women in the Valencian Community, Spain was determined until the end of gestation in order to assess its evolution and detect possible differences in relation to the values corresponding to non-pregnant women of fertile age. Fortification of milk with calcium: effect on calcium bioavailability and interactions with iron and zinc.

Calcium solubility, dialysability, and transport and uptake (retention + transport) by Caco-2 cells as indicators of calcium bioavailability have been estimated in the in vitro gastrointestinal digests of milk and calcium fortified milk. A significant linear correlation (p < 0.05) was obtained between calcium uptake and the amount of soluble calcium added to the cells, and also between percentage calcium uptake and the calcium measured in the analyzed samples. An enzymatic method proposed for the determination of oxalate in urine is adapted for the estimation of soluble oxalate content in beans, chickpeas and lentils. Oxalates were extracted in water by refluxing for 2 h. Stability of Plant Sterols in Ingredients Used in Functional Foods

The content of plant sterol (PS) and their oxidation products (POPs) in eight ingredients used to enrich functional foods was studied. A gas chromatographic (GC) technique with mass-spectrometric detection was used for identification, while GC with a flame ionization detector (GC-FID) was used for quantification. β-Sitosterol was the most abundant phytosterol, and the main POPs found were derived from this compound (7α/β-hydroxysitosterol, 7-ketositosterol, and sitostanetriol). Plant sterol oxides in functional beverages: Influence of matrix and storage

HPLC Method for Cyst(e)ine and Methionine in Infant Formulas

Cyst(e)ine and methionine were converted into cysteic acid and methionine sulfone by oxidizing with performic acid. The oxidized samples were then subjected to acid hydrolysis (6N HCl, 105-110°C/24 hr). After derivatization with phenylisothiocyanate, reverse phase HPLC separation was carried out at 48°C and with UV detection. Different gradients and pH values in the eluent were assayed to determine the best resolution. Analytical parameters, detection and quantification limits, linearity, precision and accuracy, were determined. The method was reliable and accurate for measuring cyst(e)ine and methionine in infant formulae.

Summary Background Cholesterol microbial transformation has been widely studied using in vitro fermentation assays, but less information is available on the biotransformation of plant sterols (PS). The excretion percentage of animal sterols (AS) (67–73%) is considerably greater than that of PS (27–33%) in feces from healthy humans following a Western diet. Plant sterols (PS) stability, antioxidant parameters, and color were studied during 6 months of storage at 4, 24, and 37 °C in three PS-enriched functional beverages. The selenium content of Spanish human milk samples and different milk-based and soy-based infant formulas has been estimated by using a flow injection hydride atomic absorption spectrometric method after microwave digestion of the organic matter. Mean values of 11.4 +/- 3.7 and 10.7 +/- 4.6 ng/ml for colostrum and transitional milk, 8.4 +/- 3.4 and 5.3 +/- 1.9 ng/ml for mature milk at 1 month and up to 2 months respectively, was obtained. The effect of storage (time and temperature) on the evolution of pentanal, hexanal, heptanal and pentane as volatile lipid oxidation products in two liquid ready-to-eat milk-cereal-based infant foods was studied. An SPME-GC method was used to this effect. Samples were stored for 9 months at 25, 30 and 37 °C and tested eight times during this period. Cholesterol Content in Human Milk during Lactation: A Comparative Study of Enzymatic and Chromatographic Methods.

This study validates a gas chromatography (GC) method for determining the sterol profile of human milk (HM) and compares it with an enzymatic-spectrophotometric (E-S) method. Good linearity ( r > 0.97) and low limits of detection and quantification were obtained with the GC method (<1.8 and <6 μg/100 g of HM, respectively).
