l-glutamine: a major substrate for tumor cells in vivo?

From 65 human breast cancer xenografts investigated, a net glutamine uptake was found in 13 tumors (mean +/- SE: 15.7 +/- 4.5 nmol/g per min) whereas a net release (22.5 +/- 3.3 nmol/g per min) was observed in 40 tumors. In 12 tumors neither a significant net uptake nor a net release was obvious. There is experimental evidence that glutamine is taken up by cancer cells only at arterial concentrations greater than 0.5 mM. Another parameter determining glutamine utilization by tumor cells may be the tissue oxygenation. Abstract The performance of the ion guide method in the on-line mass separation of fission products has been investigated. Activity was produced by bombarding nat. U targets with a beam of 20 MeV protons. All the mass chains from A = 95 through A = 120 have been scanned using conventional methods of nuclear spectroscopy. No chemical selectivity in the separation was observed; isotopes of highly nonvolatile elements from yttrium to palladium were produced with efficiencies equal to those for the more volatile ones. Abstract The status of the ion-guide isotope separator on-line at the Tohoku University Cyclotron is reported. The optimum ion-guide parameters were determined from test experiments using atomic and molecular ions ionized with an electric discharge inside the target chamber as well as using 64 Ga(T 1 2 = 2.6 m ) ions produced by the 64Zn(p, n) reaction. Using the present system together with a ΔE−E plastic counter telescope and a germanium detector we identified 16 short-lived nuclei including the first mass-separated nuclei 57Cu and 45V. AbstractWe describe 3 women in 1 family who had systemic urothelial leukoplakia with no underlying pathological condition of the urinary tract. Irritative clinical symptoms resolved, and the macroscopic and microscopic appearance improved after 6 months of therapy with sodium pentosan-polysulfate. Electron microscopy showed special cells in the basal layer of the cornified squamous epithelium that also have been found in the trigone of women with so-called squamous metaplasia. “Three-level shock” in ablation-refractory paroxysmal atrial fibrillation

His-bundle ablation is a very effective method to control drug-refractory supraventricular arrhythmias. We present a 69-year-old woman with paroxysmal atrial fibrillation, in whom several attempts of unipolar and bipolar His-bundle ablation were ineffective. Ultimately a stable 2/1 atrioventricular block was induced when three successive shocks of 400 W were applied. The first shock was given in that position with the largest His-potential deflection. The other two shocks were applied at catheter positions more distal and more proximal from the first one, respectively. We suppose that the initial lack of success was due to an atypical atrioventricular junction anatomy.

The conditions for coreconstitution of a bacterial ATP synthase and bacteriorhodopsin into lecithin liposomes and for light driven ATP synthesis have been optimized. A rate of maximally 280 nmol ATP min-1 mg ATP synthase-1 was achieved with monomerized bacteriorhodopsin compared with a rate of up to 45 nmol ATP min-1 mg-1 found for proteoliposomes containing bacteriorhodopsin in the form of purple membrane patches. The different rates are explained by the finding that monomeric bacteriorhodopsin is more homogeneously distributed among the liposomes than the purple membrane patches. Both soluble and nuclear oestrogen receptors have been measured in at least two separate sections from 72 endometrial cancers and 12 normal endometria. Concentration of oestrogen receptor is shown to be, in our hands, more meaningful when expressed per unit DNA than per unit protein, whether for soluble or nuclear receptor. Endometrial cancer cells from the central part of the tumour are shown to be receptor negative more frequently than those from peripheral tumour. Thus, in large cancers, biopsies from different areas are required before a tumour can be correctly designated as receptor positive, heterogeneous or receptor negative. We have studied the chromatin structure ofPenicillium chrysogenum. This fungus presents the typical nucleosomal repeat and the core DNA size characteristic of all the eukaryotes. The repeat length (about 180 base pairs) is in the range of those obtained for most fungi (160–180 base pairs) and shorter than in higher eukaryotes. Knowledge aboutP. chrysogenum chromatin structure opens the way to the study of the mechanisms of genetic regulation in this filamentous fungus.

The effects of brofaremine (CGP 11 305A), a short-acting, reversible and selective inhibitor of MAO-A, on sleep, nocturnal penile tumescence (NPT) and hormonal secretion during the night were studied during a long-term trial. Three healthy males underwent sleep-EEG and NPT recordings during consecutive nights (1) under placebo, (2) under stepwise increasing dosages of brofaremine and (3) under placebo after withdrawal. Hormone profiles were sampled during selected nights to analyze the plasma concentration of cortisol, HGH, prolactin, testosterone, LH and FSH. In anesthetized rats, the choline levels of cerebrospinal fluid and plasma obtained from blood collected from peripheral vessels (carotid artery, cardiac vessels) and from the transverse sinus were determined with a radioenzymatic assay. Cortical release of choline was studied using the "cup technique." The plasma choline level of the peripheral blood (11.5 mumol/L) was lower than that of the sinus blood. The resulting cerebral arterio-venous difference of choline was negative (3.2 mumol/L) and reflected the net release of choline from the whole brain. Some clinical studies that were performed for the purpose of assessing the potential cardiotoxicity of mitoxantrone (DHAD) have shown that repeated administrations of the drugs in some patients cause a mild impairment of cardiac functions and morphological changes in the myocardial cells qualitatively similar to those elicited by anthracyclines. Since doxorubicin has been reported to cause acute cardiac effects, probably related to its chronic cardiotoxicity, experiments were carried out on the rabbit heart to investigate whether DHAD is also able to induce acute cardiac effects. A high molecular polyanion, Liquoid, was found to inhibit at nontoxic concentrations (12-50 micrograms/ml) the natural killing (NK) and the antibody-dependent cellular cytotoxic (ADCC) activity of human peripheral blood mononuclear cells selectively. Whereas NK of the K 562 target cell was slightly or not at all affected, the spontaneous lysis of PDe-B-1, an EBV-transformed B-cell line, was strongly inhibited or even completely abolished. ADCC activity could only be inhibited by Liquoid if the target cells were mycoplasma-free, while the polyanion had no effect when mycoplasma-contaminated target cells were used. Abstract We study a simple superconvergent scheme which recovers the gradient when solving a second-order elliptic problem in the plane by the usual linear elements. The recovered gradient globally approximates the true gradient even by one order of accuracy higher in the L 2 -norm than the piecewise constant gradient of the Ritz—Galerkin solution. A superconvergent approximation to the boundary flux is presented as well.

A laser-microwave double-resonance experiment on electrodynamically trapped $^{173}\mathrm{Yb}^{+}$ ions has been performed and a value of \ensuremath{\Delta}${\ensuremath{\nu}}_{\mathrm{h}\mathrm{f}\mathrm{s}=10\phantom{\rule{0ex}{0ex}}491\phantom{\rule{0ex}{0ex}}720239.55}$\ifmmode\pm\else\textpm\fi{}0.09 Hz for the ground-state hyperfine separation has been determined. This value is corrected for small Zeeman and second-order Doppler shifts. Combined with a previous similar measurement on $^{171}\mathrm{Yb}^{+}$ and with the known ${g}_{I}$ values of both isotopes, we obtain a value of -0.004 25(6) for the differential hyperfine anomaly $^{171}\mathrm{\ensuremath{\Delta}}^{173}$.
Automated DNA-image cytometry: a prognostic tool in infiltrating bladder carcinoma?

In a retrospective study, paraffin-embedded cystectomy specimens obtained from 46 patients with bladder cancer (stage pT1 — pT4a, pN0, pN2) were analysed for tumor DNA ploidy and proliferation using automated image cytometry (LEYTAS). In 41 cases, DNA ploidy could be measured. Estimation of proliferation was possible in 26 tumors. The number of cells with a DNA content higher than 5C could be calculated in 38 of the tumors. All these three parameters are shown to correlate with patient outcome.

P-450 isoenzymes (PB1, PB2, MC1, and MC2) NADPH-cytochrome P-450 reductase, the glutathione transferases (GSTs) B and C and microsomal epoxide hydrolase (mEHb) have been studied during nitrosamine-induced hepatocarcinogenesis using immunohistochemical techniques. The investigations revealed basic differences in the expression of the individual drug metabolizing enzymes in the course of neoplastic development. Thick coal seams in the intramontane basins of the Massif Central allow a detailed study of peat/coal compaction. Three stages of compaction can be detected. Stage 1—occurred during the deposition of the peat. Coal beds which thicken and dip away from penecontemporaneous sandstones suggest rapid compaction. Stage 2—compaction of peat related to overburden pressure in some cases resulted in the formation of lakes over peat beds. The thickness of the lacustrine deposits may give an indication of the amount of compaction that took place. Abstract Collinear fast-beam laser-spectroscopy has been used to measure the hyperfine structure and isotope shift of several indium and tin isotopes. The related experimental techniques are described, including the preparation of mass-separated beams of neutron-deficient indium and tin isotopes at the GSI on-line mass separator following fusion-evaporation reactions. The deviation of the observed dependence of the charge radii upon the neutron number from the expected behaviour is briefly discussed.
Anti-glomerular basement membrane antibody-mediated glomerulonephritis due to glue sniffing

A 16-year-old girl developed rapidly progressive glomerulonephritis and renal failure. The disease was associated with high titres of antiglomerular basement membrane antibodies in serum, linear deposits of immunoglobulin G and diffuse epithelial crescents on renal biopsy. Past history revealed heavy smoking and deliberate sniffing of Pattex glue, a mixture of hydrocarbons which possibly may affect the structure of glomerular basement membrane. After treatment by repeated plasmapheresis and drug immunosuppression autoantibodies disappeared from serum but renal function was not influenced. Renal damage is a potential hazard for glue sniffing adolescents.
Characteristics of lysosomal phosphomannosyl-enzyme receptors in the rat heart

The receptor system recognizing mannose 6-phosphate groups of lysosomal enzymes has been characterized, e.g. in fibroblasts and liver cells. The purpose of this study was to demonstrate the presence of a phosphomannosyl receptor system in rat heart muscle. The characterization of receptors was accomplished with beta-N-acetylglucosaminidase (beta-GA) secreted by rat embryo fibroblasts after ammonium chloride stimulation. The receptor binding of ligand enzymes was saturated by adding increasing concentrations of beta-GA and the binding increased linearly when the content of membrane protein was increased. The enthalpies of solution and of dilution of 1-butanol and 1-pentanol were measured in micellar solutions of dodecyltrimethylammonium bromide by systematically changing the concentration of alcohols and surfactant. The enthalpies of solution at infinite dilution of alcohols at each surfactant concentration were evaluated from a linear plot. This quantity increases with surfactant concentration (up to 0.8m) with a curvature which depends on the alcohol alkyl chain length. The difficulties arising for a quantitative treatment of both the enthalpies of dilution and of solution at finite alcohol concentrations are discussed. This paper reports investigations of ZnS quasi-amorphous films by electroreflectance (ER). The films were produced by thermal evaporation and their structure determined by electron diffraction. A voltage Vo cos cut was applied through the film with two evaporated Al electrodes. A lock-in amplifier gave 2 signals, Sf at f=ω/2π frequency and S2f at 2f frequency. The S2f spectrum, characteristic of the centrosymmetric bulk component of the film, reveals tails of localized states typical of amorphous semi-conductors. Defect recovery in aluminum irradiated with protons at 20 K.

Aluminum single crystals have been irradiated with 7.0-MeV protons at 20 K. The irradiation damage and its recovery are studied with positron-lifetime spectroscopy between 20 and 500 K. Stage-I recovery is observed at 40 K. At 240 K, loss of freely migrating vacancies is observed. Hydrogen in vacancies is found to stabilize the vacancies and prolong stage III to above 280 K, where the hydrogen bound to vacancies is released. Single and multiple occupancy of hydrogen atoms at monovacancies is put forward as the reason for the two recovery stages between 280 and 400 K. A binding energy of 0.53 +- 0.03 eV is found for a hydrogen atom trapped at a monovacancy. The results are in excellent agree…

Intermediate filament composition was studied in the following endometrial stromal tumors: low-grade stromal sarcoma (endolymphatic stromal myosis), high-grade stromal sarcoma with an associated adenocarcinoma (collision tumor), diffuse clear cell stromal sarcoma and a mesodermal mixed tumor (carcinosarcoma). The tumor cells of the stromal tumors as well as the mesenchymal elements of the mixed mesodermal tumor were decorated exclusively with antibodies to vimentin.
