CD63 and Dna Mismatch Repair Protein Expression in Prostate Cancer
2021
Abstract Protein expression levels in immunohistochemistry and molecular biomarkers have been reported for their ability to predict recurrence, progression, development of metastases, and patient survival. The molecular features in low- and high-grade prostate cancer can differ and influence treatment decision and prognosis. The objective of the current study was to compare the expression of exosomal biomarkers CD63 and mismatch repair proteins (MSH2, MSH6, MLH1, and PMS2) by immunohistochemistry (IHC) in tissue of patients with prostate cancer and benign hyperplasia. Altogether, 62 patients with prostate acinar adenocarcinoma and 20 patients with prostate benign hyperplasia were enrolled i…
Novel functional chitosan and pectin bio-based packaging films with encapsulated Opuntia-ficus indica waste
2021
Abstract The aim of this work was to study the effect of adding red prickly pear, as wasted source of antioxidants on various characteristics of novel functional (antioxidant) chitosan and pectin film formulations. Four types of prickly pear material were used: fruit pulp or peel, as powdered extracts and as aqueous extracts. Prickly pear peel extract was shown to possess higher antioxidant properties than fruit pulp. The antioxidant power of extracts was pretty high, around 1000 mgAAE∙100 g−1 for extract and that of all films around 100 mgAAE∙100 g−1 for films. Even though some changes in thickness, moisture content, water solubility, oxygen permeability and water vapor permeability of fil…
Proton reduction by phosphinidene-capped triiron clusters
2021
Bis(phosphinidene)-capped triiron carbonyl clusters, including electron rich derivatives formed by substitution with chelating diphosphines, have been prepared and examined as proton reduction catalysts. Treatment of the known cluster [Fe3(CO)9(µ3-PPh)2] (1) with various diphosphines in refluxing THF (for 5, refluxing toluene) afforded the new clusters [Fe3(CO)7(µ3-PPh)2(κ2-dppb)] (2), [Fe3(CO)7(µ3-PPh)2(κ2-dppv)] (3), [Fe3(CO)7(µ3-PPh)2(κ2-dppe)] (4) and [Fe3(CO)7(µ3-PPh)2(µ-κ2-dppf)] (5) in moderate yields, together with small amounts of the corresponding [Fe3(CO)8(µ3-PPh)2(κ1-Ph2PxPPh2)] cluster (x = -C4H6-, -C2H2-, -C2H4-, -C3H6-, -C5H4FeC5H4-). The molecular structures of complexes 3 a…
The effect of alertness and attention on the modulation of the beta rhythm to tactile stimulation
2021
Abstract Beta rhythm modulation has been used as a biomarker to reflect the functional state of the sensorimotor cortex in both healthy subjects and patients. Here, the effect of reduced alertness and active attention to the stimulus on beta rhythm modulation was investigated. Beta rhythm modulation to tactile stimulation of the index finger was recorded simultaneously with MEG and EEG in 23 healthy subjects (mean 23, range 19–35 years). The temporal spectral evolution method was used to obtain the peak amplitudes of beta suppression and rebound in three different conditions (neutral, snooze, and attention). Neither snooze nor attention to the stimulus affected significantly the strength of…
High temperature oxidation of higher manganese silicides
2021
Abstract The oxidation kinetics and mechanisms of higher manganese silicides (HMS) MnSi1.75, MnSi (1.75-x)Gex, MnSi(1.75-x)Alx (with x = 0.005 and 0.01)were studied and the effects of densification methods and dopant concentration discussed. Oxidation experiments were conducted using thermogravimetry (TGA), while post characterization with X-ray Photoelectron Spectroscopy (XPS) and Scanning Electron Microscope (SEM) showed that spark plasma sintering (SPS) is a better densification method than hot pressing (HP). Except for undoped HMS, HMS doped with 0.5at% Ge had the lowest oxidation rate. Stable formation of a SiO2 protective layer was the main reason for improved oxidation resistance in …

Evaluation of tumor immune contexture among intrinsic molecular subtypes helps to predict outcome in early breast cancer
2021
BackgroundThe prognosis of early breast cancer is linked to clinic-pathological stage and the molecular characteristics of intrinsic tumor cells. In some patients, the amount and quality of tumor-infiltrating immune cells appear to affect long term outcome. We aimed to propose a new tool to estimate immune infiltrate, and link these factors to patient prognosis according to breast cancer molecular subtypes.MethodsWe performed in silico analyses in more than 2800 early breast cancer transcriptomes with corresponding clinical annotations. We first developed a new gene expression deconvolution algorithm that accurately estimates the quantity of immune cell populations (tumor immune contexture,…
Healthy mistrust or complacent confidence? : Civic vigilance in the reporting by leading newspapers on nuclear waste disposal in Finland and France
2021
Trust and confidence have been identified as crucial for efforts at solving the conundrum of high-level radioactive waste management (RWM). However, mistrust has its virtues, especially in the form of “civic vigilance”—healthy suspicion towards the powers that be. This article examines civic vigilance in the form of “watchdog journalism,” as practiced by the leading Finnish and French newspapers—Helsingin Sanomat (HS) and Le Monde (LM)—in their RWM reporting. Although both countries are forerunners in RWM, Finland constitutes a Nordic “high-trust society” while France has been characterized as a “society of mistrust.” Employing the methods of frame analysis, key RWM-related news frames were…
L’amore condiviso, l’Arte. Sulla formazione giovanile di Giuseppe Schirò e di Luigi Pirandello (1884-1890)
2021
The retrived epistolary of Pirandello and Schirò helps reconstruct the relationship between the two young writers, both protagonists of different and private love affairs. The backdrop against which their lives unfolded were the Classical College Vittorio Emanuele where they met; the Palermo of the Liberty style of the end of the 19th century and the cultural context dominated by reknowned and famous Sicilian intellectuals such as their professors Mario Villareale and Pier Giacinto Giozza, or the poets Mario Rapisardi and Eliodoro Lombardi, and other outstanding figures such as Giuseppe Pipitone Federico, Giuseppe Pitrè and Salvatore Salomone-Marino. This third part of the essay considers t…
Tritium retention in plasma facing materials of JET ITER-Like-Wall retrieved from the vacuum vessel in 2012 (ILW1), 2014 (ILW2) and 2016 (ILW3)
2021
Abstract ITER-Like-Wall (ILW) project has been carried out at Joint European Torus (JET) to test plasma facing materials relevant to International Thermonuclear Experimental Reactor – ITER [1]. Limiters and an upper dump plate of the vacuum vessel are made of bulk beryllium tiles, whereas for the divertor bulk tungsten and tungsten-coated carbon fibre (CFC) composite tiles are used. During the shutdowns in ILW1 (2012), ILW2 (2014) and ILW3 (2016), selected beryllium tiles were removed from the vacuum vessel. In this study, tiles from three positions were analysed, and analysis results were compared regarding both the tile position in the vacuum vessel and differences in the exploitation con…

A Methodological Framework to Discover Pharmacogenomic Interactions Based on Random Forests
2021
The identification of genomic alterations in tumor tissues, including somatic mutations, deletions, and gene amplifications, produces large amounts of data, which can be correlated with a diversity of therapeutic responses. We aimed to provide a methodological framework to discover pharmacogenomic interactions based on Random Forests. We matched two databases from the Cancer Cell Line Encyclopaedia (CCLE) project, and the Genomics of Drug Sensitivity in Cancer (GDSC) project. For a total of 648 shared cell lines, we considered 48,270 gene alterations from CCLE as input features and the area under the dose-response curve (AUC) for 265 drugs from GDSC as the outcomes. A three-step reduction t…

Endotrophin, a pro-peptide of Type VI collagen, is a biomarker of survival in cirrhotic patients with hepatocellular carcinoma
2021
Aim: Type VI collagen, is emerging as a signaling collagen originating from different types of fibroblasts. A specific fragment of Type VI collagen, the pro-peptide, is also known as the hormone endotrophin. We hypothesized that this fibroblast hormone would be of particular relevance in cancer types with a high amount of fibrosis activity, namely for outcome in hepatocellular carcinoma (HCC) cirrhotic patients. Patients & methods: Plasma C6M, PRO-C6 and alphafeto-protein (AFP) were assessed in 309 patients with mixed etiologies (hepatitis C, hepatitis B, alcohol and nonalcoholic fatty liver) diagnosed as cirrhotics, cirrhotics with HCC, noncirrhotics and healthy controls. Progression-f…
Editors Introduction: Morphology, Organism, Evolution
2021
In biology, interest in form was the prerogative of developmental biology, while it was practically neglected by evolutionary biology. This situation has changed a lot in recent decades and has led to a reinterpretation of the concept of evolution and evolutionism focusing more on the problem of form and morphology. In Italy, especially Alessandro Minelli, one of the editors of this issue, has dedicated his studies to the need to communicate form to structure, to reconnect morphology and evolution. This theme is a highly relevant one for philosophy, inasmuch as the question of form and morphology, since the days of Goethe and Bergson, has always been considered as the starting point for a p…
Heart rate responses during the modified six-minute walk test among Special Olympics athletes
2021
Objective: The construct validity of the modified six-minute walk test (m6MWT) was examined by addressing the following questions: 1) do participants attain a steady state plateau in heart rate (HR) within the range of 110-150 beats per minute? 2) at what percentage of their predicted HR maximum were participants during each minute of the test? Material and Methods: Participants were 36 (male=56%) Special Olympics athletes aged 34.4 years (SD = 12.7). Following familiarization, participants completed the m6MWT while wearing a Polar Team 2 heart rate monitor. Results: By the end of the first minute, average heart rate was 120bpm, and a factorial repeated measures ANOVA revealed that steady s…
Myositis/myasthenia after pembrolizumab in a bladder cancer patient with an autoimmunity-associated HLA: Immune–biological evaluation and case report
2021
Pembrolizumab (mAb to PD-1) has been recently approved for the therapy of pretreated urothelial cancer. Despite the efficacy, it is often accompanied by unpredictable and sometime severe immune-related (ir) adverse events (AEs). Here, we report the clinical and immune–biological characterization of a patient with a metastatic bladder cancer who developed myositis signs (M) and a myasthenia-like syndrome (MLS) during treatment with pembrolizumab. The patient presented an autoimmunity-associated HLA haplotype (HLA-A*02/HLA-B*08/HLA-C*07/HLA-DRB1*03) and experienced an increase in activated CD8 T-cells along the treatment. The symptomatology regressed after pembrolizumab discontinuation and a …

Causes and consequences of DNA damage-induced autophagy.
2021
Abstract Autophagy is a quality control pathway that maintains cellular homeostasis by recycling surplus and dysregulated cell organelles. Identification of selective autophagy receptors demonstrated the existence of pathways that selectively degrade organelles, protein aggregates or pathogens. Interestingly, different types of DNA damage can induce autophagy and autophagy-deficiency leads to genomic instability. Recent studies provided first insights into the pathways that connect autophagy with the DNA damage response. However, the physiological role of autophagy and the identity of its targets after DNA damage remain enigmatic. In this review, we summarize recent literature on the target…

This study aimed at identifying frequency and patterns of safety incidents in our NICU.Methods: A 6-month prospective clinical study was performed from May to October 2019 in a German 10-bed level III NICU. A voluntary, anonymous reporting system was introduced, and all neonatal team members were invited to complete paper-based questionnaires following each particular safety incident. Safety incidents were defined as safety-related events that were considered by the reporting team member as a “threat to the patient's well-being” which “should ideally not occur agai…
Canopy status modulates formation of wood rays in scots pine under hemiboreal conditions
2021
Abstract Non-structural carbohydrates (NSC) reserves are crucial for trees to cope with weather extremes, thus to ensure their survival and ecological plasticity. The NSC reserves can depend on social status, suggesting uneven plasticity of trees at the stand level. In stemwood of Scots pine (Pinus sylvestris L.), which is a widespread and important species, NSC reserves are stored in parenchyma in wood rays (WR). The quantity of WRs is adjusted intra-annually, allowing retrospective analysis of factors affecting their formation. Accordingly, the differences in WR quantity in stemwood of dominant and intermediate (canopy trees with reduced and narrow crowns) maturing Scots pine were assesse…
UAV-Aided Multi-Antenna Covert Communication Against Multiple Wardens
2021
In this paper, we propose a UAV-aided covert communication scheme assisted by a multi-antenna jammer to maximize the transmission rate between a ground transmitter and a UAV receiver against several randomly distributed wardens. The transmitter adopts the maximum ratio transmission, while the jammer zero-forces its transmitted signal at the UAV to disturb the monitoring at wardens without interfering the legitimate transmission. First, we analyze the detection performance and derive the optimal threshold for each warden to minimize its detection outage probability (DOP). Then, with the worst situation in which all wardens set their respective optimal thresholds to achieve the minimum global…
Subnational mapping of HIV incidence and mortality among individuals aged 15-49 years in sub-Saharan Africa, 2000-18: a modelling study

We aimed to assess HIV incidence and HIV mortality for all second-level administrative units across sub-Saharan Africa. Methods. In this modelling study, we developed a framework that used the geographically specific HIV prevalence data collected in seroprevalence surveys and antenatal care clinics to train a model that estimates HIV incidence and mortality among individuals aged 15–49 years. We used a model-based geostatistical framework to e…
Comparison of gap-filling techniques applied to the CCI soil moisture database in Southern Europe
2021
Abstract Soil moisture (SM) is a key variable that plays an important role in land-atmosphere interactions. Monitoring SM is crucial for many applications and can help to determine the impact of climate change. Therefore, it is essential to have continuous and long-term databases for this variable. Satellite missions have contributed to this; however, the continuity of the series is compromised due to the data gaps derived by different factors, including revisit time, presence of seasonal ice or Radio Frequency Interference (RFI) contamination. In this work, the applicability of different gap-filling techniques is evaluated on the ESA Climate Change Initiative (CCI) SM combined product, whi…

Taste Perception Profiles Are Associated With Adherence to Empirically Derived Dietary Patterns Among Older Adults With Metabolic Syndrome
2021
OBJECTIVES: Understanding individual-level drivers of food intake is critical when personalizing dietary guidance. One likely driver is taste perception; yet, limited data relate perception of all 5 tastes (sweet, salt, sour, bitter, umami) to dietary patterns (DPs). Our aim was to determine whether a multivariable measure of taste perception, taste perception profiles, was associated with adherence to empirically derived DPs. METHODS: Participants were 367 community-dwelling adults with metabolic syndrome from PREDIMED-Plus, Valencia (55–75 years; 55% female). Six taste perception profiles were derived from 5 taste perception scores via a data-driven clustering approach: Low All (23%), Hig…
Clobetasol promotes neuromuscular plasticity in mice after motoneuronal loss via sonic hedgehog signaling, immunomodulation and metabolic rebalancing
2021
AbstractMotoneuronal loss is the main feature of amyotrophic lateral sclerosis, although pathogenesis is extremely complex involving both neural and muscle cells. In order to translationally engage the sonic hedgehog pathway, which is a promising target for neural regeneration, recent studies have reported on the neuroprotective effects of clobetasol, an FDA-approved glucocorticoid, able to activate this pathway via smoothened. Herein we sought to examine functional, cellular, and metabolic effects of clobetasol in a neurotoxic mouse model of spinal motoneuronal loss. We found that clobetasol reduces muscle denervation and motor impairments in part by restoring sonic hedgehog signaling and …

Novel clinical insights into spinal hemangioblastoma in adults: a systematic review
2021
ABSTRACT Background Hemangioblastomas are well vascularized, benign CNS tumors and the third most common primary spinal cord tumor after astrocytoma/ependymoma, occurring sporadically or as a part of an autosomal dominant von Hippel-Lindau disease in which tumors are often multiple and prone to relapse. Spinal hemangioblastomas are commonly located in the cervical cord and associated with a syrinx formation. Due to location and growth trends, they may cause significant neurological deficit, impairing patient quality of life. We conducted a systematic review to understand better clinical insights of spinal hemangioblastoma in adults and compare spinal hemangioblastoma versus posterior crania…
