Clobetasol promotes neuromuscular plasticity in mice after motoneuronal loss via sonic hedgehog signaling, immunomodulation and metabolic rebalancing

2021

AbstractMotoneuronal loss is the main feature of amyotrophic lateral sclerosis, although pathogenesis is extremely complex involving both neural and muscle cells. In order to translationally engage the sonic hedgehog pathway, which is a promising target for neural regeneration, recent studies have reported on the neuroprotective effects of clobetasol, an FDA-approved glucocorticoid, able to activate this pathway via smoothened. Herein we sought to examine functional, cellular, and metabolic effects of clobetasol in a neurotoxic mouse model of spinal motoneuronal loss. We found that clobetasol reduces muscle denervation and motor impairments in part by restoring sonic hedgehog signaling and …

Off-Target-Based Design of Selective HIV-1 PROTEASE Inhibitors

2021

The approval of the first HIV-1 protease inhibitors (HIV-1 PRIs) marked a fundamental step in the control of AIDS, and this class of agents still represents the mainstay therapy for this illness. Despite the undisputed benefits, the necessary lifelong treatment led to numerous severe side-effects (metabolic syndrome, hepatotoxicity, diabetes, etc.). The HIV-1 PRIs are capable of interacting with “secondary” targets (off-targets) characterized by different biological activities from that of HIV-1 protease. In this scenario, the in-silico techniques undoubtedly contributed to the design of new small molecules with well-fitting selectivity against the main target, analyzing possible undesirabl…

Melatonin in Early Nutrition: Long-Term Effects on Cardiovascular System

2021

Breastfeeding protects against adverse cardiovascular outcomes in the long term. Melatonin is an active molecule that is present in the breast milk produced at night beginning in the first stages of lactation. This indoleamine appears to be a relevant contributor to the benefits of breast milk because it can affect infant health in several ways. The melatonin concentration in breast milk varies in a circadian pattern, making breast milk a chrononutrient. The consumption of melatonin can induce the first circadian stimulation in the infant’s body at an age when his/her own circadian machinery is not functioning yet. This molecule is also a powerful antioxidant with the ability to act on infa…

Cell population analysis of the adult murine subependymal neurogenic lineage by flow cytometry

2021

Summary This protocol provides a flow-cytometry-based procedure to classify and isolate all cells of the adult rodent subependymal zone (SEZ) neurogenic lineage, without the need for reporter mice, into different cell populations, including three neural stem cell (NSC) fractions with molecular signatures that are coherent with single-cell transcriptomics. Additionally, their cycling behavior can be assessed by means of 5-ethynyl-2′-deoxyuridine (EdU) incorporation. Our method allows the isolation of different NSC fractions and the functional assay of their cycling heterogeneity and quiescence-activation transitions. For complete details on the use, execution, and outcomes of this protocol, …

The Role of Personal Biological Resource in the Job Demands-Control-Support Model: Evidence From Stress Reactivity.

2021

Job resources can buffer the deleterious effect of adverse work environments. Extant studies on the interaction pattern between job resources and adverse environments were confined to the diathesis stress model. This traditional perspective has received the challenge from the differential susceptibility model and the vantage sensitivity model. Additionally, stress reactivity may be one of the important job resources at the personal biological level, but its moderating role was short of empirical research. This study aimed to examine how stress reactivity interacts with work environments in predicting job burnouts among 341 Chinese hospital female nurses. This study selected job control and …

Non-Small Cell Lung Cancer Harboring Concurrent EGFR Genomic Alterations: A Systematic Review and Critical Appraisal of the Double Dilemma

2021

The molecular pathways which promote lung cancer cell features have been broadly explored, leading to significant improvement in prognostic and diagnostic strategies. Epidermal growth factor receptor (EGFR) tyrosine kinase inhibitors (TKIs) have dramatically altered the treatment approach for patients with metastatic non-small cell lung cancer (NSCLC). Latest investigations by using next-generation sequencing (NGS) have shown that other oncogenic driver mutations, believed mutually exclusive for decades, could coexist in EGFR-mutated NSCLC patients. However, the exact clinical and pathological role of concomitant genomic aberrations needs to be investigated. In this systematic review, we ai…

Heterochromatin Networks: Topology, Dynamics, and Function (a Working Hypothesis)

2021

Open systems can only exist by self-organization as pulsing structures exchanging matter and energy with the outer world. This review is an attempt to reveal the organizational principles of the heterochromatin supra-intra-chromosomal network in terms of nonlinear thermodynamics. The accessibility of the linear information of the genetic code is regulated by constitutive heterochromatin (CHR) creating the positional information in a system of coordinates. These features include scale-free splitting-fusing of CHR with the boundary constraints of the nucleolus and nuclear envelope. The analysis of both the literature and our own data suggests a radial-concentric network as the main structural…

Comparison of Classroom-based Sedentary Time and Physical Activity in Conventional Classrooms and Open Learning Spaces Among Elementary School Studen…

2021

European children and adolescents spend most of their daily life and especially their school hours being sedentary which may increase their risk for chronic non-communicable diseases later in life. After the curriculum reform of Finnish basic education in 2014, most of the new or renovated comprehensive schools in Finland incorporate open and flexible classroom designs. Their open learning spaces may provide students opportunities to reduce sedentary behavior during school hours. Thus, waist-worn accelerometers were used to assess classroom-based sedentary time (ST), the number of breaks from sedentary time (BST), and physical activity (PA) among cross-sectional samples of 3rd and 5th grade…

A Soil Water Assessment Tool (SWAT) Modeling Approach to Prioritize Soil Conservation Management in River Basin Critical Areas Coupled With Future Cl…

2021

About 44% of the Indian landmass experiences the adverse impact of land degradation. This loss of sediments caused by soil erosion reduces the water quality of local water bodies and decreases agricultural land productivity. Therefore, decision-makers must formulate policies and management practices for sustainable management of basins that are cost-effective and environment friendly. Application of the best management practices (BMPs) to properly manage river basins is difficult and time-consuming. Its implication under various climate change scenarios makes it more complicated but necessary to achieve sustainable development. In this study, the soil and water assessment tool (SWAT) model…

UAV-Aided Multi-Antenna Covert Communication Against Multiple Wardens

2021

In this paper, we propose a UAV-aided covert communication scheme assisted by a multi-antenna jammer to maximize the transmission rate between a ground transmitter and a UAV receiver against several randomly distributed wardens. The transmitter adopts the maximum ratio transmission, while the jammer zero-forces its transmitted signal at the UAV to disturb the monitoring at wardens without interfering the legitimate transmission. First, we analyze the detection performance and derive the optimal threshold for each warden to minimize its detection outage probability (DOP). Then, with the worst situation in which all wardens set their respective optimal thresholds to achieve the minimum global…

Identità russe in transito nella New York di Ljudmila Ulickaja

2021

The essay analyses the atypical concept of border in the povest' by Ljudmila Ulickaja 'Veselye pochorony' (1997). This short novel offers several images related to the symbolic setting represented mainly by the city of New York, which embodies an idealized 'border city', where some Russians immigrants arrive and meet in 1991. First of all my research aims to to examine the role played by a few peculiar places represented in 'Veselye pochorony', in order to highlight the emotional dynamics of some Ulickaja's characters, who have to face the difficulties of integration in a cultural and social environment, totally different from the one they come from. Moreover this essay investigates the dev…

The Origin of Non-thermal Fluctuations in Multiphase Flow in Porous Media

2021

Core flooding experiments to determine multiphase flow in properties of rock such as relative permeability can show significant fluctuations in terms of pressure, saturation, and electrical conductivity. That is typically not considered in the Darcy scale interpretation but treated as noise. However, in recent years, flow regimes that exhibit spatio-temporal variations in pore scale occupancy related to fluid phase pressure changes have been identified. They are associated with topological changes in the fluid configurations caused by pore-scale instabilities such as snap-off. The common understanding of Darcy-scale flow regimes is that pore-scale phenomena and their signature should have a…

Multiple paths to cold tolerance: the role of environmental cues, morphological traits and the circadian clock gene vrille

2021

AbstractBackgroundTracing the association between insect cold tolerance and latitudinally and locally varying environmental conditions, as well as key morphological traits and molecular mechanisms, is essential for understanding the processes involved in adaptation. We explored these issues in two closely-related species, Drosophila montana and Drosophila flavomontana, originating from diverse climatic locations across several latitudes on the coastal and mountainous regions of North America. We also investigated the association between sequence variation in one of the key circadian clock genes, vrille, and cold tolerance in both species. Finally, we studied the impact of vrille on fly cold…

Energy spectral density as valid parameter to compare postural control between subjects with nonspecific chronic low back pain vs healthy subjects : …

2021

Abstract Background Nonspecific chronic low back pain (NSCLBP) is one of the most common and frequent health problems. Objetive to compare postural control (i.e. center of pressure (CoP) displacement and energy spectral density (ESD)) using technological devices (accelerometers and pressure platform) between subjects with NSCLBP and healthy subjects. Methods A cross-sectional case-control study was conducted. Observational study (STROBE). The final sample consisted of 60 subjects (30 NSCLBP subjects and 30 healthy subjects). Triaxial accelerometer and pressure platform were used in order to obtain ESD and CoP displacement measurements during four balance tasks (i.e. with and without vision …

Tritium retention in plasma facing materials of JET ITER-Like-Wall retrieved from the vacuum vessel in 2012 (ILW1), 2014 (ILW2) and 2016 (ILW3)

2021

Abstract ITER-Like-Wall (ILW) project has been carried out at Joint European Torus (JET) to test plasma facing materials relevant to International Thermonuclear Experimental Reactor – ITER [1]. Limiters and an upper dump plate of the vacuum vessel are made of bulk beryllium tiles, whereas for the divertor bulk tungsten and tungsten-coated carbon fibre (CFC) composite tiles are used. During the shutdowns in ILW1 (2012), ILW2 (2014) and ILW3 (2016), selected beryllium tiles were removed from the vacuum vessel. In this study, tiles from three positions were analysed, and analysis results were compared regarding both the tile position in the vacuum vessel and differences in the exploitation con…
