Akute interstitielle Nephritis nach Piperacillin

1989

A 75-year-old woman developed fever, exanthema and nonoliguric renal failure 16 days after the beginning of Piperacillin treatment. Renal biopsy revealed lympho-plasmacellular acute interstitial nephritis (AIN). A lymphocyte-transformation-test showed significant stimulation of patient's lymphocytes by Piperacillin. Corticosteroid-therapy correlated to clinical and renal improvement. Nevertheless the patient died of foudroyant septicemia caused by E. coli. Our report describes the first immunologically documented case of AIN following Piperacillin treatment.

HPLC study on the ‘history’ dependence of gramicidin A conformation in phospholipid model membranes

1989

AbstractA novel HPLC methodology for the study of gramicidin A reconstituted in model membranes has been tested in comparison with circular dichroism data. It is shown that this chromatographic technique not only corroborates most of the recent spectroscopic results but allows one to explain them in terms of mass fractions of different actual conformational species of GA in the phospholipid assemblies. In particular, the dependence of the inserted peptide configuration on the organic solvent and other parameters involved in the ‘history’ of the sample preparation and handling has been analyzed by HPLC in two phospholipid model systems: small unilamellar vesicles and micelles. Moreover, a sl…

GABA receptors are involved in the modulation of the release of 5-hydroxytryptamine from the vascularly perfused small intestine of the guinea-pig

1989

Isolated small intestinal segments of the guinea-pig were perfused arterially and the release of 5-hydroxytryptamine (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) into the portal venous effluent was determined by HPLC with electrochemical detection. Test substances were applied intraarterially. Muscimol (1 microM) time dependently first increased then decreased the release of 5-HT and 5-HIAA. The stimulatory effect was prevented by tetrodotoxin (TTx) or scopolamine, indicating that it was mediated by the release of acetylcholine. Bicuculline concentration dependently decreased (1 microM) or increased (10, 50 microM) the release of 5-HT and 5-HIAA, indicating that endogenous GABA also activ…

Hydrodynamic chromatography of macromolecules on 2 μm non-porous spherical silica gel packings

1989

Non-porous spherical 2μm silica gel particles have been tested as packing for hydrodynamic chromatography of macromolecules (HDC). Columns packed with these particles in 250×4.6mm i.d. columns can be operated very efficiently (e.g. a reduced plate height of about 2) when the detection cell is miniaturized. The packing is suitable for the separation of polystyrenes of Mw of 104–107 by hydrodynamic chromatography and the retention behaviour of polystyrenes agrees well with the known theoretical models. Preliminary results on the applicability of HDC for the separation of biopolymers are presented.

Antibodies to hepatitis B virus x-protein in sera of patients with acute and chronic acitve hepatitis

1989

Sera of patients with acute (AH) and chronic active hepatitis (CAH) were tested for anti-hepatitis B virus (HBV) x-protein (HBx) by immunoblotting, using recombinant MS2- and beta gal-HBx fusion proteins as substrate. Antibodies against HBx were detected in 5 out of 17 patients with AH at an early stage of infection, and in 13 out of 35 patients with CAH. Positive sera from AH patients showed a relatively weak anti-HBx reactivity when compared to sera from CAH patients. In follow up studies we tested serial serum samples from patients positive for anti-HBx. Patients with AH were observed for 3 to 6 weeks and CAH patients for up to 51 months. In general anti-HBx reactivities appeared to be s…

Effects of melatonin on spontaneous electrical activity of neurons in rat suprachiasmatic nuclei: an in vitro iontophoretic study.

1989

Circadian rhythms, endogenously generated in suprachiasmatic nuclei (SCN), seem to be under the direct influence of melatonin. Therefore, the effect of iontophoretically applied melatonin on electrical activity of SCN neurons was investigated in vitro. Usually, melatonin had an inhibitory effect. In the 3-h periods before (2.00-5.00 p.m.) or after (5.00-8.00 p.m.) the light-dark transition the percentage of SCN neurons sensitive to melatonin was very high (80% and 100%, respectively). However, efficacy of melatonin was low in the periods preceeding (20%) and following (33%) this 6-h time interval.

Examination and treatment of sleep-related painful erections--a case report.

1989

The case of a patient with sleep-related painful erections is described. Insomnia and a slight depressive syndrome occurred along with a long history of this disorder. No physical abnormality was found. At a baseline recording of sleep electroencephalography (EEG) and nocturnal penile tumescence (NPT), a disturbed sleep pattern and impaired NPT were recorded. Attempts to treat the disorder with diazepam, amitriptyline, trimipramine, and biperidene did not prompt a stable improvement of the disorder, but a dosage of 25 mg clozapine was sufficient to achieve normalized sleep architecture, remission of the depressive symptomatology, and normalization of NPT. It is likely that marked sedation i…

Liquid viscosity and flow rate effects on interfacial area in packed columns

1989

Abstract The influences of liquid kinematic viscosity and liquid flow rate on effective interfacial area for chemical absorption in a Raschig ring packed column were studied by absorbing carbon dioxide into potassium carbonate-bicarbonate-arsenite solutions. The viscosities of the absorbing liquid were varied by the addition of sugar. Analysis of the data shows that an increase in liquid flow rate increases the effective interfacial area over the whole viscosity range considered. The influence of liquid viscosity is more complex as the interfacial area increases with viscosity for low values, while decreasing at higher viscosities. Two correlations have been developed to express this behavi…

Restriction analysis of lambda EMBL3 background recombinants: occurrence of lambda phages carrying a head to tail oriented left arm DNA sequence.

1989

Eight representative recombinant background clones of lambda EMBL3 were analysed using KpnI, BamHI, SalI, EcoRI and HindIII digestion. We found that lambda EMBL3 carries its own left arm in the BamHI cloning site. In the way, recombinant molecules were found to be generated which can grow on Escherichia coli strain NM539. In all cases analysed, the left arm DNA was inserted in a head to tail orientation. Seven clones carried a restored BamHI site at the cos site-BamHI site connection. In the region where the inserted left arm and the right arm were ligated, BamHI cloning produces a large palindromic sequence consisting of two polylinkers. This BamHI site was incompletely cleaved in all case…

Frequency-dependent effects of activation and inhibition of protein kinase C on neurohypophysial release of oxytocin and vasopressin

1989

Isolated rat neurohypophyses were superfused in vitro and the release of vasopressin and oxytocin into the medium was determined by specific radioimmunoassays. Hormone secretion was increased by electrical stimulation of the pituitary stalk at different frequencies. The effects of several phorbol esters, known to activate (phorbol 12,13-dibutyrate, PDB) or not to affect (4a-phorbol 12,13-dideconate and phorbol 12-monoacetate) protein kinase C, and of the direct protein kinase C inhibitor 1-(5-isoquinolinylsulfonyl)-2-methylpiperazine (H7) were tested. Electrical stimulation with 450 pulses caused the release of about 45 μU vasopressin and 55 μU oxytocin, when a frequency of 3 Hz was applied…

Some structurally disordered models for poly(tetrafluoroethylene) were elaborated and the corresponding calculated X-ray diffraction patterns compared with experimental patterns. This and differential scanning calorimetry results suggest some differences between the virgin powder and a sintered polycrystalline plate of poly(tetrafluoroethylene). These differences may be related to the crystal phase transitions that occur in a range of temperatures close to room temperature.

Self-assembly properties of the proteinaceous coat secreted by the ?slime? variant of Neurospora crassa

1989

The proteinaceous extracellular material (PEM) synthesized by the cells of the ‘slime” strain of Neurospora crassa (see Martinez et al. 1989) was solubilized by treatment with urea or guanidine. Removal of these chemicals by dialysis, caused reassembly of the solubilized proteins into material with the same microscopic appearance as the original PEM. Polypeptide patterns from both native and reassembled structures were identical. Dialysis-mediated reassembly of the solubilized proteins appeared to be dependent on both concentration of the soluble macromolecules and time. Gel chromatography of PEM solubilized with different agents revealed two discrete populations of complexes with molecular…

Imaging of non-glandular superficial metastases: A comparison of xeroradiography, ultrasonography and computed tomography

1989

Between January 1983 and July 1987, 1,622 cancer patients were evaluated to outline the possible applications and limits of various investigative techniques for evaluating metastases. Seventy-three cases with superficial metastases of the dermal-hypodermal (59) layers, of the muscles (11) and of other target organs (3) were included in the study. Although non-glandular superficial metastases are a relatively rare occurrence, diagnostic imaging techniques are indicated for evaluating their extent and anatomical connections. Secondary neoplasms were studied using the imaging techniques of sonography, computed tomography and xeroradiography. The diagnostic reliability of these three techniques…

Classical Heisenberg antiferromagnets with nearest and next-nearest neighbor interactions on the face-centered cubic lattice: a model for EuTe?

1989

Magnetic properties of the Heisenberg antiferromagnet with spin quantum numberS→∞ on the face-centered cubic lattice are studied as function of temperature and magnetic field, using molecular field approximation and Monte Carlo methods. In order to model Europiumtelluride, we use isotropic exchange interactions between nearest- and nextnearest neighbors; the values of these exchange constants are taken from experiments. In addition, a pseudo-dipolar anisotropy (truncated after the next-nearest neighbor distance) is included; the molecular field calculations also are performed with the full dipolar of real EuTe in two respects: the structure in zero magnetic field involves 8 sublattices in t…

Chromatin structure of the 5′ flanking region of the yeastLEU2 gene

1989

The chromatin structure of theLEU2 gene and its flanks has been studied by means of nuclease digestion, both with micrococcal nuclease and DNase I. The gene is organized in an array of positioned nucleosomes. Within the promoter region, the nucleosome positioning places the regulatory sequences, putative TATA box and upstream activator sequence outside the nucleosomal cores. The tRNA3 Leu gene possesses a characteristic structure and is protected against nucleases. Most of the 5′ flank is sensitive to DNase I digestion, although no clear hypersensitive sites were found. The chromatin structure is independent of either the transcriptional state of the gene or the chromosomal or episomal loca…

Cross-reactivity of anti-ssDNA antibodies with heparan sulfate in patients with type I diabetes mellitus

1989

Anti-single-stranded–DNA antibodies cross-reactive with heparan sulfate were detected in serums of patients with type I (insulin-dependent) diabetes mellitus. The results suggested that heparan sulfate, the major glycosaminoglycan constituent of the glomerular basement membrane, may serve as a target antigen in vivo for cross-reactive anti-DNA antibodies. These polyreactive antibodies, directed toward repeating negatively charged units, may neutralize the heparan sulfate–associated polyanionic sites in the glomerulus, leading to an abnormal permeability of anionic plasma proteins.

Comparison of aggregate stability indices for soil classification and assessment of soil management practices

1989

Summary In view of soil structure analysis and land-use and management history evaluation, several mechanical indices of soil structure were tested on 13 Italian soils. From the wet and dry sieving aggregate distributions, a pseudo-textural aggregation index (Ipta) and a mechanical aggregation index (Ima)) were determined and compared with other soil structure stability indices based on single-sieve analysis. The single-sieve indices S and WSI were good correlated with Ipta and Ima, showing the possibility of substituting the time-consuming aggregate-size distribution determinations. All the soil structure indices were correlated with selected semi-permanent soil characteristics. Soil struc…

Endoscopic microsurgical dissection of the esophagus. Results in an animal model.

1989

Blunt dissection of the esophagus is considered the least invasive technique in the treatment of either benign or malignant diseases of the esophagus. Its disadvantage is that it has to be carried out blindly. The results may be uncontrollable hemorrhage, unrecognized injuries to the trachea, and damage to the recurrent laryngeal nerve. In order to reduce the degree of invasiveness a new endoscopic microsurgical technique for the dissection of the esophagus has been developed and tried out in animals. This paper presents the operative technique. Our new endoscopic microsurgical technique obviates a thoracotomy, while direct endoscopic vision results in improved dissection. The magnified end…

Die Skaphoidpseudarthrose: Häufigkeit, Pathogenese und Verlauf

1989

Eight-three scaphoid pseudo-arthroses were found amongst 1.104 scaphoid examinations. Sixty-seven were present at the first examination and 16 pseudoarthroses developed amongst 252 scaphoid fractures. Men were affected predominantly, particularly in the 20 to 40-year old group. Fractures in the proximal third of the scaphoid and vertical oblique fractures had a particular tendency to pseudo-arthrosis formation. The operative treatment of choice is a Matti-Russe bone graft. Only one patient in seven with definite scaphoid pseudo-arthrosis showed firm fusion.

Main posterior watershed zone of the choroid

1989

The main posterior watershed zone of the choroid is located between the nasal edge of the optic disc and the fovea and represents the area situated between the territories supplied by the temporal and nasal posterior ciliary arteries. In the fluorescein angiographies of 800 normal subjects a watershed zone was not observed in 33.1% due to technical reasons and in 22.3% due to the simultaneous filling of the peripapillar and macular choriocapillaris. In the remaining 44.6% the watershed zone was well outlined: it was straddling the optic disc in about half of these cases and involved the temporal half of the optic disc and the close choroid in the other half. Lifetime measurements in99Sr and100Zr

1989

The lifetime of the first excited state in99Sr has been determined by the delayed γ-γ coincidence method. The value of t/2=0.58(9) ns corresponds to a deformation parameter s=0.38(4). The deduced value confirms the previously proposed ν[411]3/2 Nilsson assignment for the99Sr ground state. In addition, lifetime values for the 2+ (t1/2=0.40(8) ns) and 0+ (t1/2=5.36(23) ns) states in100Zr are obtained, which deviate from older measurements and indicate a larger deformation of s=0.39(4).

Continuous monitoring of mixed venous oxygen saturation in infants after cardiac surgery

1989

Continuous mixed venous oxygen saturation (SvO2c) was measured in 16 infants immediately after cardiac surgery. A polyurethane 4F, dual channel catheter (Opticath, Modell U440, Oximetrix) with fiberoptic filaments was introduced into the pulmonary artery during cardiothoracic surgery. The catheters were left in place for an average of 67.5 h (range 27 h -125 h) and there were no catheter-related complications. Correlation between continuous in vivo SvO2 values and in vitro values was satisfactory (r = 0.85), whereas a correlation between SvO2c and arterial oxygen saturation (SaO2) was not found (r = 0.07). The sampled arterial lactate values were inversely correlated to the simultaneously m…

Mössbauer relaxation spectra in arbitrarily ordered absorbers—Line shape analysis for an iron(II) spincrossover complex in the presence of texture

1989

The stochastic theory of Mossbauer line shapes is formulated in a fashion which allows the evaluation of the spectral shapes for absorbers of arbitrary thickness, texture, and an anisotropic Lamb—Mossbauer factor. The results are specialized to a two-state-relaxation model of fluctuating electric hyperfine interaction in the case of an absorber of axially symmetric texture. The formalism is applied to the line shape analysis of Mossbauer spectra of a textured sample of the spin-crossover complex [Fe(mtz)6] (PF6)2 (mtz=1-methyltetrazole). It is found that between 185 and 240 K the rate constants for the HS→LS conversion are temperature independent, whereas an Arrhenius behaviour is found for…
