Isolation and purification of plasmids from Bacteroides fragilis using rubidium trichloroacetate density gradient centrifugation.

A rapid and easy final purification method is described for the isolation of plasmids from B. fragilis. Using RbTCA density gradient centrifugation in an airfuge ultracentrifuge ccc plasmid DNA can be separated from RNA, residual chromosomal DNA, linear and oc plasmid DNA. Pure ccc plasmid DNA is obtained from cultures of between 1 ml and 2 l in less than one day.

Abstract [(CH 3 ) 3 Pl 2 Ru(CO) 2 Cl 2 is an effective catalyst for the reaction of alcohols with triethylsilane. The rate of this reaction varies with the substituents in the hydroxy compound; it is fastest for short primary alcohols. Even in the presence of groups which can be hydrosilated (CC triple or double bonds, as well as ketones), a hydroxy group reacts preferably and completely with the silane. Kinetic experiments have been performed with the system Et 3 SiH/CH 3 CH(OH)COOEt/Ru complex.

Medium-spin levels in the N=83 nucleus147Gd

Levels of theN=83 nucleus147Gd have been investigated by methods of in-beamγ-ray and conversion electron spectroscopy using the147Sm(3He, 3n) and144Sm(α, n) reactions. Sixteen new low- and medium-spin levels have been observed in147Gd. We have located six members of thevf7/2×3− particle-octupole septuplet and several neutron single-particle states in147Gd. The levels of147Gd are discussed on the basis of the spherical shell model.

Positron Surface States on Clean and Oxidized Al and in Surface Vacancies

This Letter reports on the first discrete-lattice calculation of positron surface states on the surfaces of Al. The authors reproduce the observed values and anisotropy of the binding energies on clean surfaces, and predict the surface-state lifetimes. The temperature-independent lateral diffusion constant is calculated. Monovacancies on surfaces are predicted not to trap positrons. The effect of ordered chemisorbed monolayers of oxygen is investigated: Oxidation makes the surface state unstable with respect to positronium emission. Peer reviewed

Vinblastine-induced autophagocytosis: effects on liver glycogen

The possible similarities of the mechanism by which vinblastine induces autophagocytosis in liver were compared with the known effects of glucagon in glucagon-induced autophagocytosis. A single intraperitoneal injection of vinblastine produced a wave of autophagocytosis in less than 0.5 h in mouse hepatocytes. Liver glycogen content decreases simultaneously and blood glucose first increased and then decreased below control values. Both liver cAMP concentration and the activity of glycogen phosphorylase remained unchanged. Summary The faecal coliforms at different sites of a hypereutrophic lake near Valencia (Albufera) were identified and their relative amounts established along an annual cycle. Using lauryl tryptose broth at 35°C, followed by incubation at 44.4°C in 2% brilliant green bile, Escherichia coli and Klebsiella pneumoniae are practically the only coliforms present. A positive correlation was found between the water temperature and the relative amount of these two coliforms: K. pneumoniae predominates at high water temperatures, whereas E. coli shows preponderance during the cold period. A Study of the Nuclear Structure at High Energy and Low Spin

A novel method to study nuclear structure at low spin as a function of temperature is developed and used on 146,148Sm, 154,156Gd and 160,162Dy, by means of the (3He, α) reaction. The nuclear level density for a wide energy range is also studied. The γ-multiplicities and the first generation γ-ray spectra indicate a structure change in deformed nuclei at about 6 MeV.

Effects of Kinetin and Different Light Qualities on the Content of Carbohydrates

Summary The effects of kinetin on the content of soluble redueing sugars, starch, soluble proteins and various components of the photosynthetic apparatus were studied with primary leaves of Sinapis alba, which were grown under different light qualities (white, red, blue Jight conditions). It was especially the content of soluble reducing sugars that was most influenced by kinetin. The enhancement of the sugar content was dependent on the duration of application as well as on the concentration of kinetin. For the positive muon implanted in a metal the precession frequency shift due to hyperfine fields can be measured with high precision. This provides means to obtain information about the local electronic structure of a hydrogen like impurity in any metal in the indefinitely dilute impurity concentration.
